Claims (6)
- 효모 한세눌라 폴리모르파 DL-1 유래의 MOX 프로모터와 알부민 자체의 분비 시그날을 포함하는 알부민 cDNA를 이용하여 발현벡터를 조립하고, 이를 효모 한세눌라 폴리모르파에 도입하여 조립된 발현벡터가 염색체에 삽입(integration)된 형질 전환 효모를 선별, 발효 배양시켜 인간 혈청 알부민을 고효율로 발현시킴을 특징으로 하는 인간 혈청 알부민의 제조 방법
- 제 1항에 있어서, 발효 배양시 형질전환 효모를 메탄올이 함유된 배지에서 배양시킴을 특징으로 하는 인간 혈청 알부민의 제조 방법
- 제 1항에 있어서, 알부민 cDNA는 한국인 태아 간세포로부터 만든 cDNA 라이브러리에서 유래된 pBlue-HSA1 으로부터 분리된 유전자임을 특징으로 하는 인간 혈청 알부민의 제조 방법
- 알부민을 코딩하는 cDNA에서 5'UTR(5' 비해독 영역)이 제거되고 알부민 개시코돈이 직접 MOX 프로모터에 연결된 발현 벡터 pYHSA12 (KCTC-0503BP호)
- 숙주세포의 염색체로 발현벡터 pYHSA12가 삽입(integration)된 형질전환 효모 한세눌라 폴리모르파 DL1-L/T7 (KCTC-0502BP호)
- 숙주세포의 염색체로 발현벡터 pYHSA12가 삽입(integration)된 형질전환 효모 한세눌라 폴리모르파 DL1-L/T8 (KCTC-0501BP호)
