NEW MATERIAL:An alpha-mannosidase having the following properties. Action: hydrolyzing yeast mannan under acidic condition and liberate mannose. Substrate specificity: hydrolyzing specifically alpha-1,2-mannoside bond of yeast mannan and liberate mannose. Fittest pH: 5.0. Stable pH range: 3.0-0.9. Action temperature: 30-70 deg.C. Heat stability: stable <=40 deg.C. Molecular weight: 51,000. Isoelectric point: 4.5 or 3.9. USE:A biochemical reagent for determining alpha-mannan structure, and a reagent for detecting mannosidosis, a recessive hereditary disease caused by abnormal human saccharometabolism, in its early stage and diagnosing it. PROCESS:A funfus capable of producing alpha-1,2-mannosidase, belonging to the genus ''Aspergillus'' (e.g., ''Aspergillus saitoi'' ATCC 14332) is subjected to liquid or solid cultivation.
