Abstract

PURPOSE: To prepare immobilized enzyme having high enzymatic activity and stable for a long period, by using a specific blocked isocyanate compound hardenable with water at normal temperature, as the gel substrate.
CONSTITUTION: A urethane prepolymer obtained by the reaction of a polyisocyanate with a hydrophilic polyol prepolymer having a molecular weight of 1,000W20,000 is made to react with an isocyanate blocking agent selected from a heterocyclic compound having the skeleton of formula in the molecule, e.g. imidazole, sym-triazole, etc., hydroxypyridine, hydroxyquinoline, and substituted phenol having a pKa of 5W9.5. The equivalent ratio of the OH group and/or the NH group of the blocking agent to the NCO group of the urethane prepolymer is 0.2W2. The cold-curing blocked isocyanate compound thus obtained is mixed with a substance having enzymatic activity, and the aqueous mixture is gelatinized to obtain the immobilized enzyme.
