Abstract
Description
Claims (30)
- 1.液体サンプルを検定するための装置であって、その中を流れる液体流れに適 する中空の光透過性導管手段;および前記導管手段に設けられた光透過性物質の 多孔性塊状物であって、前記光透過性物質の塊状物の多孔度は前記サンプルの液 体流れを許すように選択され、前記塊状物はリガンド/結合複合体の少なくとも 一部をその表面上に固定しているもの; とを、組み合わせて含むことからなる前記装置に使用するためのフローセル。
- 2.前記導管手段内に設けられこれにより前記導管手段における室の少なくとも 一つの壁を画成する流動性多孔質バリヤー手段を含み、その際、前記多孔性塊状 物が特定範囲の直径に寸法が決められた複数の粒子を含み、前記バリヤー手段の 多孔度が前記範囲より十分小さく前記粒子が前記室における前記バリヤー手段に より捕捉され前記多孔性塊状物を形成することからなる請求項1記載のフローセ ル。
- 3.前記バリヤー手段が相互に間隔を空けて離れた少なくとも一対のスクリーン を含み、前記スクリーンのメッシュが前記範囲の直径より十分小さく前記粒子が 前記室における前記スクリーンにより捕捉され前記多孔性塊状物を形成し、前記 粒子が前記リガンド/結合複合体の少なくとも各々の部分をその表面上に固定し ている請求項2記載の装置。
- 4.前記バリヤー手段が一対のスクリーンを複数個含み、各々の前記一対のスク リーンが相互に間隔を空けて離れておりこれにより前記導管手段内にそれぞれの 反応室を画成し; 各々の前記室は特定範囲の直径に寸法が決められた複数の粒子を含み、前記スク リーンのメッシュが前記範囲の直径より十分小さく前記粒子が各々の前記室にお ける前記スクリーンにより捕捉されそれぞれの多孔性塊状物を形成し、各々の前 記塊状物の粒子が前記それぞれのリガンド/結合複合体の少なくともそれぞれの 部分をその表面上に固定している請求項2記載の装置。
- 5.液体サンプルを検定するための装置であって、集束光学レンズ手段; 液体流れを通すのに適しそして前記レンズ手段の焦点領域を通過し前記レンズ手 段の光軸を横断して伸びる前記レンズ手段を通る中空の円筒状通路とを、組み合 わせて含むことからなる前記装置に使用するためのフローセル。
- 6.前記レンズ手段が複数のレンズを含みそして前記通路が前記レンズの一つを 通って伸びる導管手段を含む請求項5記載の装置。
- 7.前記導管手段に設けられた光透過性物の多孔性塊状物を含み、前記透過性物 の塊状物の多孔度を前記液体サンプルの流れを許すよう選択し、前記塊状物はリ ガンド/結合複合体の少なくとも一部をその表面上に固定している請求項6記載 の装置。
- 8.前記導管手段を通る予め決められた大きさの粒子の流れを遮断するように寸 法を決められそして前記焦点領域に隣接して設けられた機械的、流動性多孔質バ リヤー手段を含む請求項5記載のフローセル。
- 9.前記レンズの前記一つにおける第一の細長い半一管状チャンネルおよび第二 の細長い半一管状チャンネルであって、前記チャンネルが互いに合わさって前記 導管手段を面成するものを含む請求項6記載の装置。
- 10.前記第二の半一管状チャンネルがその凹面部において光反射性表面を有す る請求項9記載の装置。
- 11.前記第一のチャンネルの前記一つの伸長端部が前記第二のチャンネルの伸 長端部と蝶番式に接続している請求項9記載の装置。
- 12.前記中空導管手段が前記円筒状通路に位置する光透過性管を含み、前記バ リヤー手段が前記管に設けられている請求項8記載の装置。
- 13.前記バリヤー手段が相互に間隔を空けた一対のスクリーンを含み、これに より前記管内に反応室を画成し;そして特定範囲の直径に寸法が決められた複数 の粒子を含み、前記スクリーンのメッシュは前記範囲より十分小さくこれにより 前記粒子が前記室の前記スクリーンにより捕捉され実質的に前記焦点領域に位置 可能な多孔性塊状物を形成する請求項12記載の装置。
- 14.前記レンズ手段および前記管の屈折率が実質的に適合する請求項12記載 の装置。
- 15.前記管の周囲にそして前記管と前記通路の中間に設けられた屈折率適合性 液体を含む請求項14記載の装置。
- 16.前記通路の一部が前記レンズ手段における細長い半管状の第一のチャンネ ルとして形成され、そして前記通路の残りの部分が前記第一のチャンネルと適合 する細長い半管状の第二のチャンネルとして形成される請求項7記載の装置。
- 17.前記第二のチャンネルが前記通路の内側表面の少なくとも一部を形成する 反射性表面を有する請求項16記載の装置。
- 18.特定範囲の直径に寸法が決められた複数の粒子を含み、前記流動性多孔質 手段が前記範囲より小さな直径の孔を有しこれにより前記粒子が前記バリヤー手 段に対向して前記導管手段に付着して実質的に前記焦点領域に設けられた多孔性 塊状物を形成する請求項8記載の装置。
- 19.前記粒子が実質的に光透過性である請求項18記載の装置。
- 20.前記粒子がその表面の少なくとも一部にリガンド/結合複合体の少なくと も一部で被覆された請求項19記載の装置。
- 21.励起したとき輻射線を放出しうる標識物を用いた液体サンプルを検定する ための装置であって、 前記輻射線を集束することができそしてその中で前記レンズ手段の焦点領域を通 り前記レンズ手段の光学軸を横断して伸びるレンズ手段であって、前記導管手段 は液体流れに適するものであり; 粒子径の関数としての前記多孔性手段を通る粒子の通路を制限するために前記導 管手段において前記焦点領域に隣接して設けられた流動性多孔質バリヤーとを含 む前記装置。
- 22.前記輻射線の放出を励起するための手段を含む請求項21の装置。
- 23.前記放出を励起するための前記手段が励起輻射線の供給源および前記励起 輻射線を前記焦点領域で前記導管手段に向けさせる手段とを含む請求項22記載 の装置。
- 24.前記放出が蛍光である請求項23記載の装置。
- 25.特定範囲の直径に寸法が決められた複数の粒子を含み、前記流動性多孔質 手段が前記範囲より小さな直径の孔を有し、これにより前記粒子を前記多孔性バ リヤーに対向して前記導管手段に付着して実質的に前記焦点領域に設けられた多 孔性塊状物を形成することからなる請求項23記載の装置。
- 26.前記粒子が前記励起輻射線と前記蛍光線の両方ともに対し透過性であり、 そして固定した特異的結合リガンドで被覆された請求項25記載の装置。
- 27.前記リガンドがリガンド/結合反応で複合体を形成し、前記複合体が適当 な励起輻射線により励起された場合蛍光を発光しうる分子で標識化される請求項 26記載の装置。
- 28.リガンド/結合複合体から放出される輻射線の検出により液体サンプルを 検定する方法であって、該方法が次の工程;特定範囲の直径に寸法が決められた 複数の粒子を準備し;前記粒子の懸濁液を後者の焦点領域を通りレンズ手段の光 軸を横断して伸びる導管手段に流し; 前記範囲より小さい直径の孔を有する多孔性手段により前記導管を通る前記粒子 の流れを阻止し、これにより前記粒子が実質的に前記焦点領域で付着し;少なく とも前記液体サンプルを流すことを含む粒子の前記付着を処理し、これにより前 記粒子の表面上に前記リガンド/結合複合体を作りだし;前記複合体を刺激して これにより特徴的輻射線をここから起こさせ;そして前記レンズ手段により伝達 される前記特徴的輻射線の放出を検出することからなる前記方法。
- 29.前記処理が、前記複合体の少なくとも一部でその表面の少なくとも一部分 において前記粒子を被覆し; 前記複合体を蛍光標識物で標識化し; 前記導管手段に通して前記粒子を付着し洗液を流して標識化複合体から未反応お よび蛍光標識物を分離する; ことからなる請求項28記載の検定方法。
- 30.前記刺激が前記標識化複合体上へ励起輻射線を向かわせ;前記特徴的輻射 線は励起した標識化複合体から生じそして前記レンズ手段を通過する蛍光からな る請求項28記載の検定方法。
