Description
本特許出願は、CorMedix Inc.社及びRobert DiLuccioによる係属中の先行米国仮特許出願第62/211,912号(2015年8月31日出願)「抗菌性組成物及びナノファイバーウェブを用いる方法(ANTIMICROBIAL COMPOSITIONS AND METHODS USING NANOFIBER WEBS)」(代理人整理番号CORMEDIX-11 PROV)に基づく利益を主張し、上記特許出願は引用によって本明細書に援用する。
本発明は、一般的に、患者への活性剤の送達に関し、さらに詳しくは、患者へのナノファイバーウェブを用いる活性剤の送達に関する。
本発明の別の好適な形態において、活性剤を患者に送達する方法を提供し、該方法は、
ナノファイバーウェブと、そのナノファイバーウェブに保持された活性剤とを含む活性剤送達システムを用意し;そして
該活性剤送達システムを患者の体内又は体表面に配置する
ことを含む。
本発明の目的のためのナノファイバーウェブ又はマットは、好ましくは、不織布の、ランダム配列又は整列したナノファイバーの集合体を含む。これらのナノファイバーウェブ又はマットは、典型的には、目の粗い織物(open texture)又は多孔性を特徴とする厚手の絡み合った塊(tangled mass)の形態である。本発明の目的のために、“ナノファイバーウェブ”、“ナノファイバーマット”、“ナノファイバーメッシュ”及び“ナノファイバー膜”という用語は、すべて互換的に使用できる(ナノファイバーウェブ又はマットは膜のようなものと見なすこともできる - 巨視的には膜はナノファイバー構造のネットワークである)。
本発明のナノファイバーの製造に使用されるポリマーは生体適合性である。本発明の目的のために、生体適合性とは、有毒でも、有害でも、又は生理的に反応性でもなく、そして免疫学的拒絶も起こさないことにより、害を及ぼすことなく生組織又は生体との共存が可能なことを意味する。本発明のナノファイバーの製造に使用されるポリマーは、生分解性でも非生分解性でも、合成でも天然でもよい。
前述のように、本発明は、患者の体内で制御放出される活性剤を保持するナノファイバーウェブの提供及び使用を含む。
本発明で使用できる活性剤又は成分は、生物学的製剤及び薬剤を含む。
本発明を用いて送達できる薬物は、抗感染薬、抗生物質、抗結核薬、抗真菌薬、抗ウィルス薬、抗寄生虫薬、抗リウマチ薬、非ステロイド性抗炎症薬(NSAID)、コルチコステロイド、免疫調節剤、生物学的製剤、抗新生物薬などの多数の異なるクラスの薬物を含むが、これらに限定されない。
本発明の一つの好適な形態において、ナノファイバーウェブによって送達される活性剤はタウロリジンである。
タウロリジンは、線毛及び鞭毛を破壊することによって細菌及び真菌の宿主細胞への接着を低減するので、バイオフィルムの形成を防止する。
ナノファイバーウェブへの活性剤の組込み
活性剤は、(ポリマー性ナノファイバーの)不織布構造中に包埋(すなわち“含浸”)されるか、又は不織布構造の開口部に配置される、もしくはポリマー性ナノファイバーの表面に配置される、もしくはポリマー性ナノファイバーの側壁に組み込まれる、のいずれかその他の方法で不織布構造に保持される。こうすることにより、不織布構造が解剖学的部位に送達されて体液に暴露されると、活性剤は不織布構造から放出される。
本発明の活性剤送達組成物は、いくつかの方法で投与できる。一般に、活性成分を含有しているナノファイバーウェブは、ナノファイバーウェブが体液と接触し、活性成分が体液中に一定期間にわたって制御された様式で放出されるように、組織の内部又は上部に導入される。組織又は体液の場合、ナノファイバーウェブは、治療される組織の機能をなるべく損なわないような様式で配置又は設置される必要がある。
本発明の性質を説明するために本明細書中に記載及び例示されている詳細、材料、工程
及び部品の配列には、多数の追加の変更が当業者によって可能であるが、それらもなお本
発明の原理及び範囲に含まれることは理解されるはずである。

ポリ-(ビニルアルコール)、生分解性ポリエステル、キトサン、ポリ-(プロピレンカ
ーボネート)、及びポリ-(ラクチド-グリコリド)、ポリ-e-カプロラクトン、ポリ
-トリメチレンカーボネート、ポリ-グリコリド、ポリ-p-ジオキサノンからなる群か
ら選ばれるポリマーから形成される、請求項6に記載の活性剤送達システム。

ポリマーが、ホモポリマー、コポリマー又はマルチマーでありうる、請求項10に記載
の活性剤送達システム。

Claims (11)
- ナノファイバーウェブと、そのナノファイバーウェブに保持された活性剤とを含む活性剤送達システムであって、当該ナノファイバーウェブが、ポリ-(ラクチド)(PLA)、ポリ-(ラクチド-グリコリド)、及びポリ-グリコリドからなる群から選ばれるポリマーから形成されるポリマー性ナノファイバーを含み、当該活性剤がタウロリジンを含み、前記活性剤が前記ポリマー性ナノファイバーの側壁に組み込まれている、前記システム。
- ナノファイバーウェブが不織布構造を含む、請求項1に記載の活性剤送達システム。
- 不織布構造が10mm未満の厚さを有する、請求項2に記載の活性剤送達システム。
- 不織布構造が5mm未満の厚さを有する、請求項2に記載の活性剤送達システム。
