前記ギペノシドは、ダンマラン系(dammarane type)サポニン、トリテルペン(triterpene)サポニン、ギノサポニン(gynosaponin)など、別名が使われることがあるが、当業界で通常使用されている、ギペノシドを指す別名も、本発明の範囲に含まれる。
(i)甘茶蔓に溶媒を加えて抽出した後、ろ過して、甘茶蔓粗抽出物を得るステップ;及び
(ii)前記甘茶蔓粗抽出物を、吸着剤を用いて溶出した後、乾燥して、甘茶蔓ギペノシド分画物を得るステップを含む方法によって製造することができる。
1)甘茶蔓エタノール粗抽出物の製造
乾燥された甘茶蔓の地上部分(葉と茎)1.0kgを切断/粉砕し、精製水とエタノールの体積比6:4で混合した40%エタノールを14L加え、80℃で4時間攪拌して抽出した後、抽出液を流出させて1次抽出液を得る。抽出残渣が残っている抽出器に再び同じ溶媒を11L加え、同じ温度で2時間抽出し、抽出液を流出させて2次抽出液を得、1次抽出液と合わせる。1、2次抽出液を濾過及び濃縮し、甘茶蔓エタノール粗抽出物を得た。
カラムに多孔性合成吸着剤であるHP−20を充填し、40%エタノールを加えて平衡化した後、実施例1−1)で得られた甘茶蔓エタノール粗抽出物を40%エタノールで希釈させ、徐々にロード(loading)する。次いで、45%エタノールを十分に加えて不純物を除去した後、80%エタノールで溶出して甘茶蔓ギペノシド分画物を得る。溶出液を減圧濃縮してエタノールを蒸発させた後、凍結乾燥して、粉末状の甘茶蔓ギペノシド分画物約30gを得た。
実施例1で得られた甘茶蔓ギペノシド分画物から有効成分を次のように分離した。実施例1で得られた甘茶蔓ギペノシド分画物21gをとり、シリカゲル323gを用いたカラム(直径4.6cm×高さ31cm)に入れた後、クロロホルム:メタノールの混合割合がそれぞれ8:1である混合溶媒(v/v)2L、5:1である混合溶媒(v/v)3L、3:1である混合溶媒(v/v)2L、1:1である混合溶媒(v/v)3Lを順に用いながらクロマトグラフィーを実施した。前記4種類の混合溶媒により得られたFr.A(1.56g)(8:1混合溶媒)、Fr.B(5.99g)(5:1混合溶媒)、Fr.C(3.12g)(3:1混合溶媒)、Fr.D(8.15g)(1:1混合溶媒)分画を得た。各分画をMCI−gelでアセトニトリルの水溶液濃度を5、10、15、20、25%に徐々に上げながらクロマトグラフィーを実施し、Fr.AからギペノシドUL1とギペノシドUL2を、Fr.BからギペノシドUL3とギペノシドUL4を、Fr.CからギペノシドUL5を、Fr.DからギペノシドUL6及びギペノシドUL7を得た。
実施例1によって得られた甘茶蔓ギペノシド分画物をHPLCで分析した。前記実施例2によって確認したギペノシドUL1〜ギペノシドUL7をHPLCクロマトグラムに表示して、図2に示した。
実施例1によって得られた甘茶蔓ギペノシド分画物をアセトニトリル、蒸留水の濃度勾配条件において、直径4.6mm、長さ25cmのC18カラムを用いてHPLC−UVD203nmで分析した結果、実施例1によって得られた甘茶蔓ギペノシド分画物は、粉末1g当たりにギペノシドUL4が121mg、ギペノシドUL5が95.6mg含まれていると分析された。
正常ヒト皮膚繊維芽細胞(Human dermal fibroblasts neonatal,Cascade Co.)を10%の牛胎児血清が補充されたDMEM培地を用いて24ウェルマイクロプレートに接種し(5×104細胞/ウェル)、37℃、5%CO2の条件において24時間培養した。血清が入っていないDMEM培地に交換し、24時間培養した後、各試験化合物の最終濃度が1μg/ml、5μg/mlになるように適量の試料液を処理した。48時間培養した後、細胞培養液を採取し、宝酒造社(Takara Shuzo Co.,Ltd)のコラーゲン測定キットを使用して、プロコラーゲンの量を測定した。まず、1次コラーゲン抗体が均一に塗布された96ウェルマイクロプレートに採取した細胞培養液を入れ、37℃で3時間抗原−抗体反応が起きるようにした。ウェル内の細胞培養液を除去し、PBSで4回洗浄した。各ウェルに発色誘発物質を入れて常温で15分間反応させた後、1N硫酸溶液を加えて反応を終了した。分光光度計で波長450nmにおける吸光度を測定した。標準溶液を用いて標準曲線を作成し、前記方法で得られた吸光度を標準曲線に代入して、各試験化合物が添加された細胞培養液のプロコラーゲン生成量を算出した。
1)細胞毒性テスト
正常ヒト皮膚繊維芽細胞(Human dermal fibroblasts neonatal,Cascade Co.)を10%の牛胎児血清が補充されたDMEM(Dulbeco’s Modified Eagle’s Medium)培地を用いて24ウェルマイクロプレートに接種し(5×104細胞/ウェル)、37℃、5%CO2の条件において24時間培養した。血清が入っていないDMEM培地に交換し、24時間培養した後、実施例1で得られた甘茶蔓ギペノシド分画物とこれから分離したギペノシドUL4及びUL5をそれぞれ処理した。48時間培養した後、MTT solutionを加えて4時間培養した。培地を除去し、DMSOを加えてMTT formazanを溶解した。DMSOに溶解したMTT formazanをとり、570nmで吸光度を測定し、試料を処理していない群と比較して%controlで表記した。
甘茶蔓ギペノシド分画物の皮膚刺激性を確認するために、ヒトの肌に対する刺激試験を行った。詳しくは、成人30人を対象に、背中に5×20cmの大きさで甘茶蔓ギペノシド分画物を0.2、0.4、1.0%含有する水溶液を一定量(20μl)24時間貼付してから除去し、1日、2日、6日後に、肉眼で肌の状態の変化を確認し、その結果を表2に示した。ここで肌の状態の変化程度は、下記の皮膚状態評価基準に基づいて分類し評価した。
−:非刺激性
±:弱い刺激性
+:中レベルの刺激性
++:強い刺激性
甘茶蔓ギペノシド分画物の原料状態における安定性を確認するために、プロピレングリコール、グリセリン、精製水の5:3:2の混合溶媒100mLに甘茶蔓ギペノシド分画物の粉末を4%の濃度で溶解した溶液を用いた。4℃、30℃、45℃、日光の条件において、12週間保管し、定量化のための甘茶蔓ギペノシド指標成分であるギペノシドUL4、ギペノシドUL5の経時変化(初期含有量に対する%)をHPLC分析により観察し、その結果を表3に示した。
前記実験例を基に、下記の表4〜6に表した組成にて、実施例1で得られた甘茶蔓ギペノシド分画物を含有する様々な剤形の化粧料組成物を製造した。下記剤形例は、本発明をより具体的に説明するための例示であり、本発明が下記例示のみに限定されるものではない。
甘茶蔓ギペノシド分画物を含有する高粘度乳化剤形の処方は表4の通りである。
(1)水相と油相を加温して、それぞれ均一に混合及び溶解した。
(2)75℃で水相に油相を入れて混合し可溶化した。
(3)添加Iは、甘茶蔓ギペノシド分画物を溶解した相であり、50℃で投入して混合した後、添加IIを混合した。
甘茶蔓ギペノシド分画物を含有する低粘度乳化剤形の処方は、表5の通りである。
(1)水相と油相を加温して、それぞれ均一に混合及び溶解した。
(2)75℃で水相に油相を入れて混合し可溶化した。
(3)添加相Iは、甘茶蔓ギペノシド分画物を溶解した相であり、50℃で投入して混合した後、添加相IIを混合した。
甘茶蔓ギペノシド分画物を含有する可溶化剤形の処方は、表6の通りである。
(1)水相と可溶化状をそれぞれ均一に混合及び溶解した。
(2)水相に可溶化状を入れて混合し、水中油型乳化剤形を得た。
(3)添加Iは、甘茶蔓ギペノシド分画物を溶解した相であり、可溶化した後投入して混合し、添加IIを混合して完了した。
前記剤形例1、2、及び3の化粧料組成物を、4℃、30℃、45℃、日光の条件において12週間保管し、肉眼評価と官能評価により、剤形の色、匂い、相の変化を観察して、表7〜9に示した。

剤形例1、2、及び3のように甘茶蔓ギペノシド分画物を0.2%含有する高粘度乳化剤形、低粘度乳化剤形、可溶化剤形の化粧料組成物を、4℃、30℃、45℃、日光の条件において12週間に保管し、甘茶蔓ギペノシド成分のうち定量化のための指標成分であるギペノシドUL4とギペノシドUL5の経時変化(初期含有量に対する比%)をHPLC分析により観察し、その結果を表10〜12に示した。
甘茶蔓ギペノシド分画物含有クリームのシワ改善効果を観察するために、甘茶蔓ギペノシド分画物を0.4%含有するクリームを製造した。その製造構成は、下記表13の構成の通りである。
顔を半分に分けて、目尻から2cm離れた地点を測定部位とし、右と左を測定した。測定機器は、Visiometer SV600(Courage−Khazaka electronic GmbH、Germany)を用い、シワレベルのパラメータのうちR2(Maximum roughness)を測定し、シワ改善効果の結果を図5及び図6に示した。12週間甘茶蔓ギペノシド分画物を含有するクリームを使用した結果、対照製品を使用した部位は、使用前に比べて、目元のシワ改善効果が表われなかったが、試験製品を使用した部位の目元のシワは、69.10から43.97に改善されたことを確認した(p<0.05)。
皮膚の弾力は、Cutometer(登録商標、Courage+Khazaka electronic GmbH、Germany)を用いて測定し、測定は、1回ずつ、両方の目元の同じ部位に対して行い、R2(皮膚の再変形能力)値を、皮膚弾力度の評価資料として用いた。R2値は、総体的な弾性であり、1に近い値であるほど、肌がより弾力的であることを意味する。甘茶蔓ギペノシド分画物を含有するクリームの使用による皮膚弾力を測定した結果を図7及び図8に示した。試験製品の使用部位の皮膚の弾力は、使用前が0.58だったことに比べて、使用12週後には0.66まで増加したことが確認でき、対照製品の使用部位の皮膚弾力に比べて有意に(p<0.05)皮膚弾力が増加することを確認した。
皮膚真皮緻密度は、Skin Scanner High Resolution Ultrasound(taberna promedicum、Germany)で、両方の目元の部位を撮影し、その画像を用いて製品の使用前後の皮膚真皮緻密度を分析した。皮膚真皮緻密度の測定単位は%で、測定値が高くなるほど、真皮緻密度が改善されたことを表す。甘茶蔓ギペノシド分画物を含有するクリームの使用による皮膚真皮緻密度の改善効果を測定した結果を図9及び図10に示した。対照製品を使用した部位の皮膚真皮緻密度は、使用12週後に有意なレベルで改善が見られなかったことに対し、試験製品を使用した部位の皮膚真皮緻密度は、使用12週後(20.78%)には使用前(16.53%)に比べて有意に改善されたことを確認した。
甘茶蔓ギペノシド分画物の透明ジワ改善効果を確認するために、ヒトの皮膚に対する臨床試験を行った。
Claims (4)
- 下記定義されたギペノシドUL4、及びギペノシドUL5を含有する甘茶蔓分画物を含有し、
前記ギペノシドUL4、及び前記ギペノシドUL5の含有量は、分画物の総重量に対して20〜30重量%である、
シワ改善用の化粧料組成物:
前記において、
ギペノシドUL4は、((23ξ)−3β,20β,21α−トリヒドロキシ−19−オキソ−21,23−エポキシ−ダンマル−24−エン−3−O−[α−L−ラムノピラノシル(1→2)][β−D−キシロピラノシル(1→3)]−α−L−アラビノピラノシド((23ξ)−3β,20β,21α−trihydroxy−19−oxo−21,23−epoxy−dammar−24−ene−3−O−[α−L−rhamnopyranosyl(1→2)][β−D−xylopyranosyl(1→3)]−α−L− arabinopyranoside);
ギペノシドUL5は、(23ξ)−3β,20β,21α−トリヒドロキシ−19−オキソ−21,23−エポキシ−ダンマル−24−エン−3−O−[α−L−ラムノピラノシル(1→2)][β−D−グルコピラノシル(1→3)]−α−L−アラビノピラノシド((23ξ)−3β,20β,21α−trihydroxy−19−oxo−21,23−epoxy−dammar−24−ene−3−O−[α−L−rhamnopyranosyl(1→2)][β−D− glucopyranosyl(1→3)]−α−L− arabinopyranoside)を意味する。 - 前記組成物は、組成物の総重量に対し、前記ギペノシドを0.0001〜30重量%で含有することを特徴とする、請求項1に記載のシワ改善用の化粧料組成物。
- 前記組成物は、組成物の総重量に対し、前記ギペノシドUL4及び前記ギペノシドUL5のいずれか一つ以上を0.0002〜9重量%で含有することを特徴とする、請求項1に記載のシワ改善用の化粧料組成物。
- 前記シワは、透明ジワであることを特徴とする、請求項1に記載のシワ改善用の化粧料組成物。
