Description
大部分は周知であるがいくらか説明を必要とするものもある、ある特定の用語が、本明細書及び特許請求の範囲の全体を通じて使用されている。本明細書において使用する用語は以下のとおり理解されるべきである。
閉塞層は、液体及び少なくとも一部の気体の通過に対する不透過性バリアを提供するのに有用である。閉塞層は、多孔性又は非多孔性であり得る。代表的なバリアとしては、不織布及び織布繊維ウェブ、編み布地、フィルム、発泡体、ポリマーフィルム並びに他の一般的なバッキング材料が挙げられる。いくつかの実施形態では、下にある対象が見える透明な閉塞層が望ましい。好適な閉塞層としては、その開示が参照により本明細書に援用される国際公開第WO2014/149718号に記載のものが挙げられる。
図2は、ナノ構造表面を有する例示的な基材の概略図である。基材600のナノ構造表面602は、典型的には、様々な高さ及びアスペクト比のナノ構造(ナノスケール形成部(nanoscale feature))605を含み得る。一般に、ナノ構造表面は、ナノ構造異方性表面を有し得る。ナノ構造異方性表面は典型的には、ナノスケール形成部を含み得る。いくつかの実施形態では、ナノ構造異方性表面は、異方性のナノスケール形成部を含み得る。いくつかの実施形態では、ナノ構造異方性表面は、ランダムな異方性のナノスケール形成部を含み得る。ナノ構造異方性表面は典型的には、約2:1以上、好ましくは約5:1以上の高さと幅の比(アスペクト比)を有するナノスケール形成部を含み得る。いくつかの実施形態では、高さと幅の比は、更には50:1以上、100:1以上、又は200:1以上であり得る。ナノ構造異方性表面は、例えば、ナノピラー若しくはナノカラム、又はナノピラー若しくはナノカラムを含む連続ナノウォールなどのナノスケール形成部を含み得る。典型的には、ナノスケール形成部は、基材に対して実質的に垂直な、急勾配の側壁を有する。
本開示の金属酸化物層は、金属酸化物を含む。金属酸化物は、抗菌効果を有することが知られているものであり得る。ほとんどの医療用途では、金属酸化物は生体適合性でもあり得る。いくつかの実施形態では、金属酸化物層において使用される金属酸化物としては、酸化銀、酸化銅、酸化金、酸化亜鉛、酸化マグネシウム、酸化チタン、酸化クロム、及びこれらの組み合わせを挙げることができるが、これらに限定されない。これらの実施形態のうちのいくつかでは、金属酸化物は、Ag2Oを含むがこれに限定されない酸化銀であり得る。いくつかの実施形態では、金属酸化物層は、99重量%未満、95重量%未満、90重量%未満、80重量%未満、40重量%未満、20重量%未満、10重量%未満、5重量%未満、1重量%未満の非酸化金属を含み得る。金属酸化物層が40重量%を超える非酸化金属を含む場合、物品は、導電性がより高くなり、すなわち、物品の抵抗率が減少し、抗菌剤の放出もまた減少する。
吸収層において使用される吸収材料は、織布又は不織布の綿又はレーヨンを含むが、これらに限定されない任意の好適な材料から製造できる。吸収パッドは、吸収層として使用でき、経皮薬物送達のための薬物、患者においてホルモン又は他の物質をモニタリングするための化学指標を含んでいてもよい、複数の物質を含有するのに有用であり得る。
物品の抗菌効果は、例えば、物品をアルコール又は体液若しくは体組織などの水性電解質と接触させ、その結果、Ag+などの金属イオン、原子、分子又はクラスターが放出されるときに達成され得る。抗菌効果を生じるのに必要な金属の濃度は、金属によって変わる。一般に、抗菌効果は、血漿、血清又は尿などの体液では、10ppm未満の濃度で達成される。いくつかの実施形態では、物品からのAg+放出濃度は、0.1ppm、0.5ppm、1ppm、2ppm、2.5ppm、3ppm、4ppm、5ppm、6ppm、7ppm、8ppm、9ppm、10ppm又はこれらの値のうちの任意の2つの間の、それら2つの値を含む範囲であり得る。上で議論したとおり、金属酸化物層における金属酸化物の量が増加すると、今度は物品から放出される金属イオンが増加する。例えば、60重量%を超える金属酸化物は、物品からの金属イオンの放出を増強する。したがって、本開示の物品は、非常に効果的な抗菌効果を提供できる。いくつかの実施形態では、物品は、7日以内に細菌増殖の4logを超える減少を呈し得る。いくつかの実施形態では、物品は、7日以内に細菌増殖の6logを超える減少を呈し得る。

物品は、7日以内に細菌増殖の4logを超える減少を呈する、方法。
米国特許第5,888,594号及び国際公開第WO2015/013387号に記載されている自作のプラズマ処理システムを本質的に使用して、ナノ構造を得た。全てのポンピングをターボ分子ポンプによって行い、それによりプラズマ加工で従来行われていたよりもはるかに低い作動圧力で作動させるために、円筒電極の幅を42.5インチ(108cm)まで大きくし、プラズマシステム内の2つのコンパートメント間の仕切りを取り外した。ポリマー基材のロールをチャンバー内に取り付け、基材を円筒電極周囲に巻き付け、円筒の反対側の巻取りロールに固定した。巻出し及び巻取り張力は、3ポンド(13.3N)に維持した。チャンバーのドアを閉め、0.5ミリトルのベース圧力までチャンバーをポンプダウンした。次に、酸素及びヘキサメチルジシロキサン(HMDSO)などの気体をチャンバー中に導入した。作動圧力は、公称で(nominally)5ミリトル又は10ミリトルであった。高周波エネルギー6000ワットの電力を円筒電極に印加することによって、プラズマを発生させた。特定の実施例に記述されている特定の処理時間のために所望される速度でフィルムが送られるように、円筒電極を回転させた。基材片(piece−part substrate)に関しては、サンプルを、ウェブキャリア(web carrier)又は円筒電極表面のいずれかに貼り付けて、特定の実施例に記述されている特定のエッチング時間のために所望される速度で処理した。
BYK Haze−Gard Plus(BYK Gardiner(Columbia,MD)製)を用いて、ASTM D1003及びD1004に従い、透過率の測定を実施した。
Fluke 175 True RMS Multimeter又は非接触式抵抗計、Delcomモデル717Bコンダクタンスモニタを使用して、表面抵抗率を測定した。
電極(Orion Sure−flow IonPlus Silver/Sulfide組み合わせイオン選択性電極、モデル9616BN)スロープを調べた。Ag+標準溶液を調製した。電極は、0.3、1、10、及び100ppmのAg+標準溶液中に浸漬することにより毎日校正した。物品の銀イオン放出を以下のとおり評価した。60mLの水、1mLのISA、及び50μLの1000ppm銀標準溶液を、100mLの使い捨てビーカーに加え、撹拌子を加えた。Ag ISE上の初期電位を記録した。3cm2の物品をビーカーに加え、タイマーを開始した。溶液中の遊離銀イオン濃度を、10秒間隔で、Tiamo 2.4ソフトウェア(Metrohm(Herisau,Switzerland)製)により60分間記録した。
修正JIS Z 2801試験法(日本工業規格、日本規格協会(東京、日本))を使用して、物品の抗菌活性を評価した。細菌接種材料を、一部のNutrient Broth(NB)及び499部のリン酸緩衝液の溶液中で調製した。細菌懸濁液の一部(150μl)を、物品の表面に置き、接種した物品を特定の接触時間、27+/−1℃でインキュベートした。インキュベーション後、物品を20mlのD/E Neutralizing Broth中に置いた。Neutralizing broth中の生存菌数を、3M Petrifilm(3M(St.Paul,MN))を使用して測定した。
阻害領域試験(Kirby−Bauerディスク拡散感受性試験)を使用して、物品の抗菌性浸出化合物に対する細菌の感受性又は抵抗性を測定した。材料のディスクに含浸させた抗菌剤の存在下、Mueller−Hinton寒天培地上で、微生物を増殖させた。ディスクの周囲に領域が存在するか又は存在しないかが、その化合物がその微生物を阻害する能力の間接的な尺度である。
銀を被覆した吸収性物品を重量測定し[W(0)]、次に、T時間水中に浸漬した。特定の時間(T)で水中から物品を取り出し、再度重量測定した[W(T)]。吸収された水の重量[W(T)−W(0)]を、吸収性物品の初期重量[W(0)]で割って吸収力を計算し、これを特定の時間での吸収力%として報告した。
着色した水の液滴を銀被覆吸収性物品に添加し、その水滴が物品に完全に吸収された時間を記録することにより、濡れ時間を測定した。
400gmの75nmシリカ粒子(Nalco Chemical Co.から「NALCO 2329K」という商品名で入手)を、1クォート(0.95リットル)の広口瓶に入れた。450グラムの1−メトキシ−2−プロパノール、9.2グラムの3−(メタクリロイルオキシ)プロピルトリメトキシシラン、及び0.23グラムのヒンダードアミン窒素酸化物阻害剤(BASF Specialty Chemicalsから「PROSTAB 5128」という商品名で入手)(水溶液、5重量%阻害剤)を一緒に混合し、撹拌しながら広口瓶に加えた。広口瓶を密封し、16時間80℃に加熱して、表面改質されたシリカ分散体を生成した。ロータリーエバポレーター(rotary evaporation)により混合物から水分を更に除去して、42.4重量%の75nm SiO2の1−メトキシ−2−プロパノール溶液を生成した。
ポリプロピレン布を、Midwest Filtration CoからUNIPRO 150 SMSという商品名で入手し、上記のナノ構造処理により前処理した。第1のガス種は、20sccmの流速で供給されたヘキサメチルジシロキサン(HMDSO)蒸気であり、第2のガス種は、500sccmの流速で供給された酸素であった。曝露中の圧力はおよそ10ミリトルであり、6000ワットの電力でプラズマを発生させた。処理時間は約54秒であった。次に、ナノ構造処理有り及び無しのポリプロピレン布を、20ミリトルで6.8%O2のガス混合物を使用してスパッタリング堆積によって、銀で被覆した。それらのAg+放出及びlog増殖試験の結果を、表2に示す。
3MウレタンAeroフォームを、実施例2のプラズマ処理によって前処理し、20ミリトルで29%O2のガス混合物を使用してスパッタリング堆積によって、Agで更に被覆した。被覆サンプルをlog減少試験によって試験したところ、試験時間2時間の時点で4.5logを超える減少を達成し、4.5logを超える減少という殺菌効果を7日間まで維持した。被覆サンプルの吸収力は500%を超える。
