Description
本出願は、米国特許法第119条(e)の定めにより、2014年9月23日に出願された「Systems and Methods for Integration of Microfluidic Tear Collection and Lateral Flow Analysis of Analytes of Interest」という表題の仮出願第62/053,923の出願日を本出願の有効出願日とする利益を受けることを請求し、当該仮出願の全体の開示は参照により本明細書に組み込まれる。
基材構成、および、対象の分析物の存在及び/又は濃度を検出するために基材を製作し使用する方法は、例えば米国特許第6,319,676:5,141,850;5,602,040;5,656,503;5,714,389;5,591,645;5,989,921;6,319,676;6,485,982;7,763,454号に開示され、米国特許出願公開第2015/0017068号、米国特許第5,714,389;5,989,921および6,485,982号には、アッセイ基材について記述され、それによれば、対象の分析物が、試験領域に固定された分析物(限定されない例として、抗分析物抗体)用の固定されたバインダーによって試験領域内に直接捕捉される。このアプローチにおいて、分析物用のラベルが付された第2のバインダーに結合された分析物(限定されない例として、第2の抗分析物抗体)は、「半サンドイッチ」の形態で、試験領域内で結合される。米国特許第6,319,676および5,141,850号には、アッセイ基材について記載され、それによれば、対象の分析物(限定されない例として、抗分析物抗体)が、対象の分析物用のバインダーと共有結合したバインダーパートナー(ビオチン)に結合する、固定されたバインダー(限定されない例として、アビディンまたはストレプトアビジン)によって、試験領域に間接的に捕捉される。このアプローチにおいて、捕捉可能な結合部分(限定されない例として、ビオチン付加された抗分析物抗体)−分析物−検知可能な結合部分(限定されない例として、ラベルが付された抗分析物抗体)の複合体を含む「完全サンドイッチ」は、流体が基材を通過するように形成され、そして、捕捉可能な結合部分(限定されない例として、アビディンまたはストレプトアビジン)と結合する結合部分によって試験領域内で捕捉される。

(e)前記第2のサンプル領域は第1のサンプル領域と流体連通するとともに、流体サンプルの少なくとも一部を受け取るように形成され、第2のサンプル読み取りを生成するために流体サンプル中で1つ以上の分析物を検出するように構成された検出基材を含み、
前記デバイスは、更に、
(f)前記垂直の入口に流体的に接続された流体リザーバであって、該流体リザーバは移動流体を含み、流体リザーバから第1のサンプル領域への移動流体の導入は、前記流体サンプルの少なくとも一部を第1のサンプル領域から第2のサンプル領域へ移動させる、流体リザーバと、を含む、デバイス。 - 前記第2のバルブが、第1のサンプル領域から第2のサンプル領域への流体流動を制御するパッシブバルブ又はアクティブバルである、請求項1に記載のデバイス。
- 前記第1のバルブがパッシブバルブである、請求項1に記載のデバイス。
- 前記第2のバルブがパッシブバルブ又はアクティブバルであり、
a)前記パッシブバルブは第2のサンプル領域への前記流体サンプルの流れを抑制する1つ以上の幾何学的形状を含み、あるいは、b)前記アクティブバルブは、疎水性のコーティングを有する電極を含み、電極に電圧が加えられることにより、疎水性のコーティングの溶解が引き起こされ、それによって、第2のサンプル領域へ前記流体サンプルの少なくとも一部を流れさせる、請求項1に記載のデバイス。 - 疎水性のコーティングはアルカンチオール自己組織化単分子膜を有する、請求項4に記載のデバイス。
- 第1のサンプル領域は毛細管流路を含む、請求項1に記載のデバイス。
- 毛細管流路の少なくとも1つの壁は、感圧性接着剤の層を含む、請求項6に記載のデバイス。
- 感圧性接着剤の層は毛細管流路を通って前記流体サンプルの少なくとも一部の流れを遅らせるために前記流体サンプルの少なくとも一部と相互に作用する、請求項7に記載のデバイス。
- 少なくとも1つのトランスデューサは毛細管流路の壁に位置する、請求項7または8のいずれか1つに記載のデバイス。
- 第2のサンプル読み取り値は、流体サンプル中の1つ以上の分析物の存在または濃度を示す、請求項1−9のいずれか1つに記載のデバイス。
- 前記流体サンプルの量は10nLから10μLまでの範囲内である、請求項1−10のいずれか1つに記載のデバイス。
- 検出基材は微小多孔性の基材を含み、操作中に、前記流体サンプルの少なくとも一部が微小多孔性の基材を通過する、請求項1−11のいずれか1つに記載のデバイス。
- 微小多孔性の基材は、前記流体サンプルの少なくとも一部が流れの方向に微小多孔性の基材を通過する際に、流体の最前部の実質的な均一化を促すように構成された幾何学的形状を有する、請求項12に記載のデバイス。
- 微小多孔性の基材は、a)砂時計の形をした幾何学的形状、b)検出基材の中心方向の卵形の開口部、またはc)流体の最前部の実質的な均一化を促すための複数の開口部を有する、請求項13に記載のデバイス。
- 微小多孔性の基材は、流体の最前部の実質的な均一化を促すための複数の開口部を含む、請求項12に記載のデバイス。
