また、本発明は、SOCS(suppressor of cytokine signaling)の発現抑制剤または活性抑制剤を含む、幹細胞の免疫抑制活性誘導用組成物に関する。
また、本発明は、SOCS(suppressor of cytokine signaling)の発現または活性抑制を通じて幹細胞の免疫抑制能の活性を誘導する方法に関する。
1−1.ヒト組織由来間葉系幹細胞の分離および培養
本実験は、サムスン医療院の研究審査委員会(Institutional Review Board;IRB)の承認(IRB No.2011−10−134)を受け、すべてのサンプルは、事前同意を得て収集した。間葉系幹細胞の分離は、従来知られている方法で分離した。分離した細胞は、10%FBS(fetal bovine serum、Invitrogen−Gibco)および100U/mLペニシリン/ストレプトマイシン(Invitrogen−Gibco)が含まれているDMEM(Dulbecco’s Modified Eagle’s Medium、Invitrogen−Gibco、Rockville、MD)培地を利用して2×103cells/cm2の密度でシーディングして、37℃および5%CO2条件下で培養した。
siRNA形質感染24時間前に、MSCsを播種(plating)して、形質感染日に50%のコンフルエンスになるようにした。細胞を製造社の指示に応じてLipofectamine2000試薬(Gibco−Invitrogen、Rockville、MD)で形質感染させた。
SOCS1の発現を阻害するために、脂肪組織由来間葉系幹細胞(AT−MSC)にSOCS1 shRNA(short hairpin RNA)および赤色蛍光タンパク質(RFP)を発現するアデノウイルスを処理した。具体的に、SOCS1をターゲットとするshRNA配列は、ヒトU6プロモーターとTagRFPマーカー遺伝子含有シャトルベクター(pO6A5−U6−mPGK−TagRFP)にクローニングされて、pO6A5−U6−shSOCS1−mPGK−TagRFP発現ベクターとして準備し、シャトルベクターのU6−shRNA−SV40−pA領域は、BACベクターに組換えしてトランスファーした。回収した組換えアデノウイルス(Ad5−U6−shSOCS1−mPGK−TagRFP)は、HEK−293細胞で増殖させ、PTD(protein transduction domain)であるHP4は、ペプトロン社から購入した。間葉系幹細胞にアデノウイルスパーティクルをトランスフェクションするために、HP4(100nM)とともにアデノウイルスパーティクルをMOI(multiplicity of infection)100で処理し、室温で30分間無血清培地に培養させた。以後、培養された細胞をPBSで洗い、Ad−RFP−shSOCS1およびHP4 preparationで培養し、2時間後に細胞をPBSで洗って、血清含有培地で培養させた。選別された間葉系幹細胞は、RFP(red fluorescent protein)を用いた蛍光観察およびウェスタンブロッティングを通じて確認した。
細胞を冷たいPBS(Gibco−Invitrogen)で洗浄し、300μLの冷たいRIPAバッファー[1%Triton X−100、150mMのNaCl、0.1%SDS(sodium dodecyl sulfate)、および1%デオキシコール酸ナトリウムを含む50mMのTris−HCl、pH7.5]でプロテアーゼ抑制剤カクテル(Thermo Fisher Scientific、Rockford、IL、USA)で溶出した。
間葉系幹細胞に固定溶液である4%ホルムアルデヒドを処理し、光を遮断させた状態の室温で30分間反応させた後、PBSで3回洗った。細胞の内部に発現するタンパク質(PTGES、CXCR7)を検出するために、細胞に0.25%Triton X−100を処理し、光を遮断させた状態の室温で5分間反応させて、細胞透過性を高めた。以後、さらに細胞を3回洗い、5%FBSブロッキング溶液を処理して、室温で1時間反応させた後、さらに細胞を洗い、Santa Cruz Biotechnology(Santa Cruz、CA)で購入した1次抗体を処理(1:100)した後、やはり室温で1時間反応させた。次に、細胞をさらに3回洗い、Alexa Fluor 488が付着しているgoat anti−mouse IgG(Invitrogen−Gibco)2次抗体を処理して、室温で1時間反応させ、以後、Carl Zeiss LSM 700 confocal microscope system(Jena、Germany)を利用して細胞イメージを得た。
96−ウェルプレートに10%FBSが補充された高グルコースDMEM内にMSCsを1.25×104cells/mLでシーディングした。24時間後、細胞増殖を抑制するために、10μg/mLのミトマイシン−C(Sigma−Aldrich)を添加した。処理された細胞をさらなる2時間の間に37℃でインキュベーションした後、培養培地で5回洗浄した。引き続いて、1×105hPBMCs(human peripheral blood mononuclear cells)を勾配遠心で分離し、各ウェルに添加し、1μg/mLのPHA(phytohemagglutinin;Sigma−Aldrich)でT−細胞増殖を促進した。引き続いて、BrdU(5−bromo−2−deoxyuridine)を添加する前に、3〜4日間PHA−活性化したhPBMCsを異なる条件のMSCs上で培養した。増殖水準は、Roche Applied Science(Mannheim、Germany)のアッセイキットを利用して18時間後に測定した。
8〜9週齢のNOD/SCID免疫欠乏マウス(Jackson Laboratories、Bar Harbor、ME)に300cGyの全身照射法を実施し、24時間後にヒト末梢血液単球細胞を静脈投与した。具体的に、各マウスに2×107個のヒト末梢血液単球細胞を1×106個の間葉系幹細胞とともに投与した。以後、同じ個数の間葉系幹細胞を投与7日目に反復投与した。
MSCでSOCSを抑制するために、siRNAを利用してSOCS1またはSOCS3の発現を抑制した。その結果、図1のAに示したように、SOCS1またはSOCS3は、それぞれのMSCで有意的に発現抑制されることを確認することができた。
SOCS1をターゲットとするshRNA発現アデノウイルスを脂肪組織由来間葉系幹細胞(AT−MSC)に形質感染させることにより、MSCでSOCSの発現抑制を確認した。その結果、図2に示したように、赤色蛍光タンパク質(RFP)の標識(A)およびウェスタンブロッティング(B)の結果においてMSC内SOCS1タンパク質の発現が有意的に抑制されることを確認することができた。
免疫抑制能を増進させるためにIFN−γをMSCに処理した後、SOCS(Suppressors of cytokine signaling)の発現を免疫ブロッティングを利用して分析した。
前記実施例2で獲得されたSOCS下向き調節されたMSCと、これにIFN−γをさらに処理したMSC間の免疫抑制能をin vivo移植片対宿主病(GVHD)の動物モデルで比較評価した。
MSCを高密度培養(5,000cells/cm2)する場合、PTGES(Prostaglandin E synthase)とCXCR7(chemokine(C−X−C motif)receptor type 7)の遺伝子発現が増加するかを確認することにより、高密度培養により免疫抑制能がさらに増進されるかを評価した。
PTGESに特異的な抗体を利用してウェスタンブロッティング(A)および免疫細胞化学染色(B)した結果、図6に示したように、低密度培養(200cells/cm2)に比べて高密度培養(5,000cells/cm2)時にPTGESのタンパク質発現が顕著に増加することを確認した。
CXCR7に特異的な抗体を利用してウェスタンブロッティング(A)および免疫細胞化学染色(B)した結果、図7に示したように、低密度培養(200cells/cm2)に比べて高密度培養(5,000cells/cm2)時にCXCR7のタンパク質発現が顕著に増加することを確認した。

<6> 前記SOCS(suppressor of cytokine signaling)の発現抑制剤または活性抑制剤は、SOCS(suppressor of cytokine signaling)またはこれをコードする遺伝子に特異的なsiRNA(small interfering RNA)、shRNA(short hairpin RNA)、miRNA(microRNA)、リボザイム、DNAzyme、PNA(peptide nucleic acids)、アンチセンスオリゴヌクレオチド、抗体、アプタマー、拮抗剤、抽出物および化合物からなる群より選択されるものであることを特徴とする、<5>に記載の組成物。
<7><1>〜<4>のいずれか一項に記載の幹細胞を有効成分として含む、免疫抑制用薬学的組成物。
<8> 前記組成物は、体液性拒否反応、移植片対宿主疾患、臓器移植時の拒否反応、自己免疫疾患またはアレルギー性疾患の予防または治療用であることを特徴とする、<7>に記載の組成物。
<9> 前記自己免疫疾患は、クローン病、紅斑病、アトピー、関節リウマチ、橋本甲状腺炎、悪性貧血、アディソン病、第1型糖尿、ループス、慢性疲労症候群、繊維筋肉痛、甲状腺機能低下症と亢進症、硬皮症、ベーチェット病、炎症性腸疾患、多発性硬化症、重症筋無力症、メニエール症候群(Meniere’s syndrome)、ギラン・バレー症候群(Guilian−Barre syndrome)、シェーグレン症候群(Sjogren’s syndrome)、白斑症、子宮内膜症、乾癬、全身性硬皮症、喘息および潰瘍性大腸炎からなる群より選択されるものであることを特徴とする、<8>に記載の組成物。
<10> 幹細胞にSOCS(suppressor of cytokine signaling)の発現または活性を抑制する段階を含む、幹細胞の免疫抑制能の活性を誘導する方法。
<11> インターフェロンガンマを幹細胞に処理する段階をさらに含むことを特徴とする、<10>に記載の方法。
<12> 前記幹細胞を高密度培養する段階をさらに含むことを特徴とする、<10>に記載の方法。
<13> 前記高密度培養は、5,000cells/cm 2 〜20,000cells/cm 2 の密度で培養することを特徴とする、<12>に記載の方法。
<14> 前記幹細胞は、自家由来、他家由来または同種異系由来であることを特徴とする、<10>に記載の方法。
Claims (3)
- (a)ヒト由来の間葉系幹細胞においてSOCS(suppressor of cytokine signaling)1またはSOCS3の発現または活性を抑制すること、
(b)前記間葉系幹細胞をインターフェロンガンマで処理すること、および
(c)前記間葉系幹細胞を5,000cells/cm2〜20,000cells/cm2の密度で培養すること、
を含む、in vitroでヒト由来の間葉系幹細胞の免疫抑制活性を誘導する方法。 - (a)ヒト由来の間葉系幹細胞においてSOCS(suppressor of cytokine signaling)1またはSOCS3の発現または活性を抑制すること、
(b)前記間葉系幹細胞をインターフェロンガンマで処理すること、および
(c)前記間葉系幹細胞を5,000cells/cm 2 〜20,000cells/cm 2 の密度で培養すること、
を含む、免疫抑制活性が誘導されたヒト由来の間葉系幹細胞をin vitroで製造する方法。 - 前記間葉系幹細胞は、臍帯、臍帯血、骨髄、脂肪、筋肉、神経、皮膚、羊膜、絨毛膜、脱落膜および胎盤からなる群より選択されるものに由来する、請求項2に記載の方法。
