Description
本出願は、2015年4月24日に出願された米国仮特許出願第62/152,417号の利益を請求し、この出願の開示全体を参照することができ、ここに引用することで本明細書の一部をなすものとする。

〈分子イメージング及び多重療法のための低分子量マンガンビスホスホネート錯体〉
(高スピンMn(II)錯体の合成例)
所望の最終容量の再(double)蒸留水に絶えず撹拌しながら、2当量のエチドロン酸及び4当量の炭酸水素ナトリウムを添加する。溶液のpHは、10分後に約3.8〜4.0となる。その後、pHは、希NaOHによって約5.5〜7.0に更に調整することができる。この後、1当量のMnCl2を添加する。MnCl2の添加後、溶液は予想通り、より酸性(pH約3〜4)となるが、pHは中程度に希釈した塩基、例えば、NaOHで7.0〜7.5に上昇させるべきである。最も一貫性のある有効な結果は、Mn(II)添加前にpHが5〜7であり且つアルカリ金属カチオン(例えば、Na)が予め存在して、pKaが0.70及び1.46のエチドロネート上の2つのホスフェート酸素と少なくとも配位結合している場合に、達成された。
〈造影剤中のMnの錯体形成の差異〉
図3Aで、1−OHビスホスホネートとMn2+の2:1錯体が熱力学的に有利であることが判明した。合成中に化学量論を変えることによって、ビスホスホネート:Mn2+の比が2:1より高くなっても、Mn2+の追加の錯体形成は観察されない(図3)。
〈体内分布の変更、画像診断及び治療活性の向上のための、金属と錯体形成した1−ヒドロキシエタン−1,1−ジホスホン酸由来の小分子薬物〉
Mnビスホスホネート錯体は、MRイメージング及び画像誘導療法のための一連の新規分子イメージングプローブを創出する新しい機会を提供する。1−ヒドロキシルエチレンジホスホネートは、中心炭素から離れたR2基を介して又はリン酸エステル化によって容易に結合することができ、したがって、公知のターゲティング及び/又は治療活性を有する多くの既存の小分子薬物を用いて誘導体化させることができる。
〈診断及び治療用の高スピン鉄ピロホスフェート錯体及びその誘導体〉
鉄(III)とピロホスフェート(P2O7)との相互作用を探求した。遍在性(ubiquitous)のジホスフェートテトラアニオンであるP2O7は、Fe(III)の最も強い公知のキレート剤の1つである。1つのFe(III)と共に2つのP2O7アニオンを組み込んでいる常磁性スキャフォールド(scaffold)の合成のためのプロトコールを開発し、2つの内圏水を配位している高スピン八面体Fe(III)錯体(q=2)を生成した(図7A)。7Tの磁界強度で、この低分子量錯体のr1は5.2mM−1秒−1であり、これは、同一磁界強度において最も強い市販Gd(III)薬剤ガドブトロールの緩和度に等しい。1.5Tのより臨床的に関連する磁界強度では、Fe(P2O7)2錯体のr1は35mM−1秒−1まで増加する。このような緩和度は、いかなるFe(III)造影剤でもこれまで報告されてない。このスキャフォールドの印象的なコントラスト強調は、2つの内水の並外れて安定な配位及びリン酸基の広範な水和によって媒介される著しい外圏寄与と関連している可能性がある。Fe(P2O7)2錯体の安定度定数(log K1)は、EDTA(log K1=26)を用いた競合実験に基づき、中性pH及び室温において22超と推定される。したがって、配位強度は、典型的に約17のlog K1を有する市販の大環状Gd(III)錯体より高い。Fe(P2O7)2の安定性は、種々の化学量論でピロホスフェート及びFe(III)についてこれまでに報告されている錯体と一致している。
1当量の第二鉄塩(塩化第二鉄、酢酸第二鉄、クエン酸第二鉄など)を、所望の最終容量の再蒸留水中の2当量の二塩基性ピロリン酸ナトリウムに、絶えず撹拌しながら添加する。溶液は、曇りのままであるはずである。激しく撹拌しながら溶液の温度を80又は90℃に上昇させ、次いで3当量の炭酸水素ナトリウムを添加する。これは、結果として起こるCO2生成により測定される方法で行われるはずである。加熱下で継続的に激しく撹拌するにつれて、溶液は最終的に約2〜20分にわたって透明になり、かすかな黄緑色の色相を保持するであろう。透明になるまでの時間及び色相度は、試薬の出発濃度及び適用される相対的熱量に左右される。溶液が透明になるとき、試料が熱から取り除かれるので撹拌は継続すべきである。溶液が室温に達したとき、最終pHは約7.0〜7.2となるはずである。
40mMチアミンピロホスフェート及び40mMメグルミンを、室温で絶えず撹拌しながらddH2Oに溶解させる。絶えず撹拌しながら、H2O中20mMのFeCl3をゆっくり添加する。次いで、60mMのNaHCO3を添加する。最終pHは、6.5〜7である。試料を冷却し、ゴールドないしオレンジないしブラウン色の分散ガラス状ミクロビーズが得られるまで、フリーズドライ/凍結乾燥する。乾燥されると、錯体は、室温で数カ月を超えて安定であり続ける。

- 請求項1から12のいずれか1項に記載のMRI造影剤を、医薬として許容される担体中に含むMRI造影用組成物。
- 静脈内又は動脈内投与のために製剤化されている、請求項13に記載のMRI造影用組成物。
- 前記医薬として許容される担体が、水又はリン酸緩衝生理食塩水である、請求項13又は14に記載のMRI造影用組成物。
- 6.0〜8.0のpHを有する、請求項13から15のいずれか1項に記載のMRI造影用組成物。
