Description
HLB=(E+P)/5
式中、Eはオキシエチレン含有量の重量パーセントであり、Pは多価アルコール含有量(グリセロール、ソルビトールなど)の重量パーセントである。本明細書中の化合物について、2つのヒドロキシル基を有するグリセロール区分、1つのヒドロキシル基を有するグリセロール区分、及び任意の追加の多価分子のヒドロキシル含有区分は、Pの定義に含まれた。
2.一晩増殖プレートから、リン酸緩衝水(PBW)中の108CFU/mLの霊菌懸濁液を調製する。PBW中に1:10で希釈し、107CFU/mL作業懸濁液を作成する。これを、皮膚に接種するために使用する。
3.DURAPREP 8635塗布器(3M Co.から入手可能)のスポンジを、セラチア作業懸濁液で飽和させる。区域の中央で始め、皮膚にわたり前後にスポンジを拭い、中心から両方向に動く。同じスポンジを再飽和させ、90°方向を変え、新しい懸濁液で繰り返す。次に、同じスポンジを使用するが、再飽和させない。中心から第3の方向に拭う。皮膚を完全に乾燥させる(30分間)。
4.滅菌皮膚マーカーでスクラブ環の底に印を付け、皮膚に触れることにより、覆われるべき区域の中心の3つの区域に印を付ける。覆布片からライナを除去し、皮膚に適用する。滅菌ガーゼで上面にわたり滑らかにする。皮膚上の印を付けた区域に対応するように、覆布上の区域に印を付ける。
5.カップスクラブ法(ASTM E1874−09)により、覆布の周りの接種区域から2つの基準試料を回収する。
6.覆布を定位置に35℃で4時間置く。
7.皮膚上での4時間後、各覆布を除去し、清潔なライナ上に置く。覆布の下の皮膚の印を付けた区域をカップスクラブ法(ASTM E1874−09)を用いてこする。回収した溶液を系列希釈する。純溶液及び希釈溶液をトリプチカーゼ大豆寒天(TSA)プレート上に広げる。
8.各覆布上の印を付けた区域から、3つの25mm試料を打ち抜く。0.1%チオ硫酸ナトリウムを含有する20mLの中和緩衝液(Difco)に、それぞれ別個に移す。1分間超音波処理し、2分間激しく渦動させる。系列希釈し、全ての純溶液及び希釈溶液をTSAプレート上に広げる。
9.32℃で3日間、プレートをインキュベートする。
10.プレート上の赤色コロニーのみ(セラチア)を計数する。平方センチメートル当たりの取り出した細菌を計算する。関連する皮膚及び覆布部位からの細菌を合計し、ログ10に変換する。覆布当たり3回の再現部位の平均を計算する。
2.一晩増殖プレートから、リン酸緩衝水(PWB)中の108CFU/mL霊菌懸濁液を調製する。PBW中に1:10で希釈し、107CFU/mL作業懸濁液を作成する。これを、皮膚に接種するために使用する。

4.細菌の基準試料を回収する。滅菌金属型板を(覆布により覆われていない区域の)皮膚上に置く。1ml SSS中で滅菌綿棒を事前に濡らし、型板内の皮膚上で綿棒を30秒間こする。1ml管に綿棒を戻し、30秒間渦動させ、系列希釈し、TSAに広げる。
5.覆布を滑らかに置き、覆布が皮膚に接着するように、滅菌ガーゼを用いて皮膚に覆布を適用する。
6.各覆布を5分間、置く。
7.滅菌外科用メス刃(10番)で、覆布を通して6センチメートルの切開を行う。脂肪層を通して、筋肉まで切断するが、皮膚区画(約1〜2cmの深さ)の下面からの汚染を避けるために皮膚片を完全に通しては切断しない。
8.滅菌手袋を着けた指で、切開を1分間処置する。切開の内側の周りを移動し、創傷縁部で引くために、各手の初めの2本指を使用する。
9.切開縁部に沿った覆布の持ち上がりについて点検する。
10.切開を、滅菌ステンレススチール5.5’’ウェイトラナー開創器で約3センチメートル開創する。
11.滅菌ガーゼ(2片の12重ガーゼ)を25ml滅菌リン酸緩衝水(PBW)で湿らせ、開創した切開上に緩く置く。
12.各覆布区画上の3つの切開の全てについて工程を繰り返す。
13.皮膚をホイル鍋で(緩く)覆い、35℃で全部で4時間インキュベートする。
14.1時間後、インキュベーターから皮膚を除去し、ガーゼ及び開創器を除去する。(覆布の上面上の)切開の周りを手袋を着けた指でこすることにより、切開縁部を15秒間処置する。開創器を元のところに置き(開創器が元の切開に合うように保つ)、新しい湿らせたガーゼで覆う。ホイル鍋で覆い、35℃インキュベーターに戻す。
15.全部で2時間後、ステップ14を繰り返し、全部で3時間後、再び繰り返す。
16.4時間後、ガーゼ及び開創器を除去する。各切開を、覆布の持ち上がり及び覆布の下に見える流体について評価する。
17.各切開の縁部に沿って細菌を回収する。SSSの1ml管中で滅菌綿棒を事前に濡らし、この綿棒を切開の内側の周りで2回、回転させる。SSSの管中に綿棒を折り入れ、30秒間渦動させる。(「切開内」試料)
18.皮膚から覆布を引き剥がす。覆布の下に移動した皮膚上の水分を探す。
19.切開縁部の周りの皮膚から細菌を回収する。SSSの1ml管中で滅菌綿棒を事前に濡らし、この綿棒を皮膚表面上の切開の縁部の周りで1回、回転させる。SSSの管中に綿棒を折り入れ、30秒間渦動させる。(「周囲皮膚」試料)
20.綿棒で溶液を系列希釈し、それぞれ、トリプチカーゼ大豆寒天(TSA)プレート上に広げる。
21.32℃で3日間プレートをインキュベートする。
22.mL(綿棒)当たりの細菌を計算する。覆布当たりの切開の平均値を計算する。ログ10CFU/綿棒に変換する。

前記モノアシルグリセロールのアルキル基が、C8〜C18アルキル基である、項目4に記載の組成物。

前記モノアシルグリセロールが、グリセロールモノカプリレート、グリセロールモノラウレート、グリセロールモノイソステアレート、グリセロールモノオレエート、及びこれらの組み合わせからなる群から選択される、項目5に記載の組成物。

前記疎水性ビヒクルが、エーテル基を含む、項目1に記載の組成物。

Claims (11)
- HLB法を用いて決定されるときに10以下の親水性−親油性バランスを有する疎水性ビヒクル中に可溶化されたグルコン酸クロルヘキシジンを含み、前記疎水性ビヒクルは2つの近接した、ヒドロキシル基、エステル基及びエーテル基から選択される水素結合基を含み、前記水素結合基のうちの少なくとも1つはヒドロキシル基である組成物であって、
グルコン酸クロルヘキシジン1重量部当たり1重量部以下の、HLB法を用いて決定されるときに10を超える親水性−親油性バランスを有する親水性ビヒクルを含む、組成物。 - 前記疎水性ビヒクルが、エステル基を含む、請求項1に記載の組成物。
- 前記疎水性ビヒクルが、エーテル基を含む、請求項1又は2に記載の組成物。
- 前記疎水性ビヒクルが、ジプロピレングリコールを含む、請求項3に記載の組成物。
- 前記疎水性ビヒクルが、グリセリルモノアルキルエーテルを含む、請求項3に記載の組成物。
- 前記グリセリルモノアルキルエーテルが、エチルヘキシルグリセリンである、請求項5に記載の組成物。
- 前記疎水性ビヒクルが、近接したヒドロキシル基を有するアルコールを含む、請求項1に記載の組成物。
- 前記疎水性ビヒクルが1,2−ジオールである、請求項1に記載の組成物。
- 前記アルコールが、1,2−オクタンジオール、1,2−デカンジオール、及びこれらの組み合わせからなる群から選択される、請求項7又は8に記載の組成物。
- 前記組成物が、グルコン酸クロルヘキシジン1重量部当たり0.1重量部以下の親水性ビヒクルを含む、請求項1〜9のいずれか一項に記載の組成物。
- 前記組成物が、グルコン酸クロルヘキシジン1重量部当たり0.1重量部以下の水を含む、請求項1〜10のいずれか一項に記載の組成物。
