Description
50℃で1カ月、分子量60〜90%、特に70〜85%の減少;および
37℃で6カ月、分子量30〜80%、特に40〜70%の減少。
(a)カプロラクトンおよびポリ(アルキレンオキシド)部分の共重合単位を含むプレポリマーと;
(b)カプロラクトンおよびC2〜C6ジオールの共重合単位を含むポリカプロラクトンジオールと;
(c)ジイソシアネートと
を共に反応させることにより得ることができる。

(a)カプロラクトンおよびポリ(アルキレンオキシド)部分の共重合単位を含むプレポリマーと;
(b)カプロラクトンおよびC2〜C6ジオールの共重合単位を含むポリカプロラクトンジオールと;
(c)ジイソシアネートと
を用意するステップと:
(2)成分(a)、(b)および(c)を共に反応させるステップと
により調製され得る。
PEGブロック(400、2000および8000g/mol)とカプロラクトンブロック(500〜3500g/mol)の長さを変化させた。標的プレポリマー分子量は、7000〜11000g/molの間となるように選択した。プレポリマーバッチサイズは約500〜600gとした。プレポリマーを、以下の通りその組成を変化させて調製した(表1参照):バッチA)プレポリマーAを、7418g/molの理論分子量を標的にして、PEG400 32.01g(16.0mol%)、カプロラクトン561.58g(98.4mol%)およびスズ(II)オクトアート0.608g(0.03mol%)から作った、バッチB)プレポリマーBを、7592g/molの理論分子量を標的にして、PEG2000 149.81g(2.0mol%)、カプロラクトン418.84g(97.9mol%)およびスズ(II)オクトアート0.45g(0.03mol%)から作った、バッチC)プレポリマーCを、9027g/molの理論分子量を標的にして、PEG8000 461.93g(10.0mol%)、カプロラクトン59.30g(90.0mol%)およびスズ(II)オクトアート0.07g(0.03mol%)から作った、バッチD)プレポリマーDを、7592g/molの理論分子量を標的にして、PEG2000 394.86g(2.0mol%)、カプロラクトン1103.95g(97.97mol%)およびスズ(II)オクトアート1.20g(0.03mol%)から作った。
撹拌タンク型反応器に、乾燥PEG400(MW400g/mol)32.01g(16.0mol%)、カプロラクトン561.58g(98.4mol%)およびスズ(II)オクトアート0.608g(0.03mol%)をこの順で供給した。乾燥窒素を反応器に連続的にパージした。反応器を油浴および60rpmの混合速度を使用して155℃に予熱した。PEG400を反応器に添加する前にロータリーエバポレーター(rota−evaporator)で乾燥および溶融させた。次いで、ε−カプロラクトンを添加し、最後に触媒スズ(II)オクトアートを添加した。PEG−PCLプレポリマーの予備重合時間は5時間とした。プレポリマーの理論分子量は7418g/molであった。
プレポリマーB(実施例1の表1)およびポリカプロラクトンジオール(MW約530g/mol)を、予熱した(110℃)反応器に供給する前に、真空下90℃で少なくとも1時間混合、乾燥および溶融させた。反応混合物を窒素下で混合(75rpm)した。1,4−ブタンジイソシアネートを反応器に供給した。プレポリマー、ポリ(ε−カプロラクトン)ジオールおよびBDI間のモル比は1:1:2とした。反応時間は13分とした。
プレポリマーが実施例1の表1のプレポリマーCであることを除いて、実施例3のように鎖延長重合を行った。反応時間は15分とした。
プレポリマーが実施例1の表1のプレポリマーCであることとプレポリマー、ポリ(ε−カプロラクトン)ジオールおよびBDI間のモル比が0.25:1.75:2であることを除いて、実施例3のように鎖延長重合を行った。反応時間は12分とした。
プレポリマーが実施例1の表1のプレポリマーCであることとプレポリマー、ポリ(ε−カプロラクトン)ジオールおよびBDI間のモル比が0.05:1.95:2であることを除いて、実施例3のように鎖延長重合を行った。反応時間は20分とした。
プレポリマーが実施例2の表1のプレポリマーDであることを除いて、実施例3のように鎖延長重合を行った。反応時間は20分とした。
表3の生体吸収性ポリマー1、2、3、4および6を、ホットプレスを使用して加工する前に真空下で一晩乾燥させた。上および下プレート温度を160℃に設定した。2枚のテフロンシートを型とホットプレートの間に置いた。溶融時間は2分とし、引き続いて圧力(約170bar)下で30秒維持した。正確な量のポリマーを使用して型に充填した。室温に冷却した後、試料を機械的に打ち抜き、さらなる分析のために冷凍庫に保った。
合成ポリマーの生体吸収性およびその生理活性薬剤を放出する能力を証明するために、いくつかのポリマーを選択して生分解および膨潤試験を行った。
ポリマー6 0.5gをDCM10gに溶解し、油相(O)を形成した。ウシ血清アルブミン(BSA)0.1gを蒸留水(dH2O)0.5gに溶解して内水相(W1)を形成した。Tween 80(登録商標)1.5gをdH2O48.5gに溶解して外水相(W2)を形成した。W1およびOを、高せん断ミキサーを使用して4000rpmで5.5分間ホモジナイズして油中水型(W1/O)エマルジョンを形成した。結果として生じたW1/Oエマルジョン5gを外水相(W2)に移し、7000rpmでホモジナイズして最終水中油中水型(W1/O/W2)エマルジョンを形成した。油相から溶媒を除去するために、エマルジョンを、マグネチックスターラーを使用して650rpmで24時間撹拌した。
外水相がdH2O 50gのみからなることを除いて実施例11のように配合物を調製した。
BSAを水相に相対して固体配合物に使用した(バリンとの共結晶として)ことを除いて実施例11のように配合物を調製した。この固体BSA配合物0.1gを油相に直接添加し、ホモジナイズして油中固体型(s/o)エマルジョンを形成した。

2.5%ポリマー溶液を形成するためのポリマー0.25gをDCM10gに溶解したことを除いて実施例11のように配合物を調製した。
(W1/O)エマルジョンのW2への添加速度を減少させたことを除いて実施例11のように配合物を調製した。
ポリマー0.1gをEA10gに溶解したことを除いて実施例11のように配合物を調製した。
ポリマー0.25gをEA10gに溶解したことを除いて実施例11のように配合物を調製した。
ポリマー0.5gをEA10gに溶解したことを除いて実施例11のように配合物を調製した。
ポリマー0.1gをEA10gに添加し、外水相がdH2O 48.5g中PEG6000 1.5gからなることを除いて実施例11のように配合物を調製した。
ポリマー0.1gをEA10gに添加し、外水相がdH2O 48.5g中PVP1.5gからなることを除いて実施例11のように配合物を調製した。
ポリマー0.1gをEA10gに添加し、外水相がdH2O 48.5g中PVA1.5gからなることを除いて実施例11のように配合物を調製した。
動的光散乱は、形成した微粒子の粒度分布を決定するために使用することができる方法である。動的光散乱粒度測定では、体積中位径D(v、0.5)は粒度分布の50%が上にあり、50%が下にある直径である。D(v、0.9)は、体積分布の90%がこの値より下にある値である。D(v、0.1)は、体積分布の10%がこの値より下にある値である。スパンは、以下の式で示される、10%、50%および90%分位数(quantile)に基づく分布の幅である:
走査型電子顕微鏡法(SEM)は、粒子形態を研究するために一般的に使用される技術である。実施例17で調製した微粒子をPolaron SC515 SEMコーティングシステムを使用した画像法の前に金コーティングした。次いで、これらをJEOL 6400走査型電子顕微鏡で観察した。画像を、Scandiumソフトウェアを使用してとらえた。図2は、10ミクロン未満の大きさの平均粒径を有する概して球形の粒子の形成を示している。
Claims (19)
- ポリマーがポリ(アルキレンオキシド)部分、カプロラクトン部分およびウレタン部分を含む構造単位由来のポリウレタンであるポリマー微粒子であって、0.01〜100ミクロンの粒径を有し、1〜3の範囲の分散性スパンを有する、ポリマー微粒子。
- 活性物質をさらに含む、請求項1に記載の微粒子。
- 活性物質がタンパク質またはペプチドである、請求項2に記載の微粒子。
- 15〜80ミクロンの粒径を有する、注射用の請求項1に記載の微粒子。
- 1〜3ミクロンの範囲の見かけの空気動力学径を有する、エアロゾルとして使用するための請求項1に記載の微粒子。
- 0.02〜2ミクロンの粒径を有する、眼内使用のための請求項1に記載の微粒子。
- 動物の生理学的環境中で分解する、請求項1〜6のいずれかに記載の微粒子。
- ポリマーが完全に分解されるのにかかる時間が、1日〜250週程度である、請求項1〜7のいずれかに記載の微粒子。
- リン酸緩衝食塩水中50℃で1カ月後にポリマーの平均分子量の減少が60〜90%、特に70〜85%である、請求項1から8のいずれかに記載の微粒子。
- リン酸緩衝食塩水中37℃で6カ月後にポリマーの平均分子量の減少が30〜80%、特に40〜70%である、請求項1から8のいずれかに記載の微粒子。
- リン酸緩衝食塩水中における37℃での膨潤性が、1〜500%である、請求項1〜10のいずれかに記載の微粒子。
- リン酸緩衝食塩水中における37℃での膨潤性が10〜100%である、請求項1から11のいずれかに記載の微粒子。
- 微粒子が活性物質をさらに含み、活性物質が2時間、24時間、10日、30日、60日、90日または120日までに放出されるものである、請求項2〜12のいずれかに記載の微粒子。
- 分散性スパン(dispersivity span)が1.1〜2.5、特に1.2〜2.0の範囲にある、請求項1から10のいずれかに記載の微粒子。
- ヒトまたは動物患者の体内で完全に吸収される、請求項1〜14のいずれかに記載の微粒子。
- ポリマーが
(a)カプロラクトンおよびポリ(アルキレンオキシド)部分の共重合単位を含むプレポリマーと;
(b)カプロラクトンおよびC2〜C6ジオールの共重合単位を含むポリカプロラクトンジオールと;
(c)ジイソシアネートと
を共に反応させることにより得ることができる、請求項1〜15のいずれかに記載の微粒子。 - ポリ(アルキレンオキシド)部分が、ポリ(C2−アルキレンオキシド)若しくはポリ(C3−アルキレンオキシド)またはその混合物から選択される、請求項16に記載の微粒子。
- C 2 〜C 6 ジオールが、エチレングリコールまたはジエチレングリコールから選択される、請求項16または17に記載の微粒子。
- 活性物質を患者に送達するための組成物であって、請求項2〜18のいずれかに記載の微粒子を含む組成物。
