Description
[0001]本出願は、開示が参照により本明細書に組み込まれている、2010年5月26日に出願された米国特許仮出願第61/396372号の優先権を主張する。
[0017]「動物」という用語は、カタラーゼの産生又は活性の増大が有益であり得るあらゆる動物又は他の生物(例えば、菌類)を意味し、ヒト、鳥類、ウシ属、イヌ科、ウマ科、ネコ科、ヤギ属(hicrine)、オオカミ、ネズミ科、ヒツジ属、又はブタの動物が挙げられる。
本発明
[0093]イヌ科動物の膀胱移行上皮癌(cbTCC)細胞を、5%のFBS、5%の新生仔ウシ血清、及びペニシリン−ストレプトマイシン(GIBCO BRL、Grand Island、NY、USA)を補ったDMEM培地中で、5%のCO2の加湿雰囲気中、37℃で培養した。細胞を、6ウェルの培養プレート(Corning Incorporated Life Sciences、Acton、MA、USA)中に50,000細胞/mlで播種して、処理前に24時間インキュベートした。ウエスタンブロット解析用には、細胞を、10−7M及び10−9Mのカルシトリオール(1α,25−ジヒドロキシビタミンD3)並びに10−9Mのセオカルシトール(EB1089、1α,25−ジヒドロキシ−22,24−ジエン−24,26,27−トリホモビタミンD)で24時間処理した。酵素活性解析用には、細胞を、10−7M及び10−9Mのカルシトリオール、10−9Mのセオカルシトール並びに10−9Mの類似体V(1α,25−ジヒドロキシ−16−エン−23−イン−ビタミンD3)で24時間処理した。遺伝子発現解析用には、細胞を、10−7M及び10−9Mのカルシトリオール、並びに10−7M及び10−9Mのセオカルシトールで24時間処理した。処理は、同じものを少なくとも3つずつ行った。全ての対照におけるビヒクル(エタノール)の最終濃度は、培地の0.1%未満であった。
[0094]回収した細胞を均質化して、マンガンスーパーオキシドジスムターゼ(MnSOD)、銅/亜鉛スーパーオキシドジスムターゼ(Cu/ZnSOD)、カタラーゼ(CAT)、及びグルタチオンペルオキシダーゼ(GPX)の産生について解析した。遠心分離後に細胞溶解液を収集して、全タンパク質を定量化した。同等の量(20又は40μg)の全タンパク質を、プレキャスト10%ポリアシルアミドゲル上で分離させて、電気的にニトロセルロース膜に転写した。トランスブロットした膜を、Tris−緩衝食塩水(TBS)中5%のスキムミルクを添加して4℃で一晩ブロッキングした。この膜を、37kD(緑色のバンド)及び25kD(桃色のバンド)の分子量のところで切断した。この膜の上部をCAT、GPX、及びβ−アクチンについて染色し、下部をMnSOD及びCu/ZnSODについて染色した。特定のタンパク質用の一次抗体を以下の希釈率:ウサギ抗ヒトMnSODを1:2000(Upstate Biotech);ヒツジ抗ヒトCu/ZnSODを1:2000(Calbiochem);ヒツジ抗ヒトカタラーゼを1:1000(Bidesign);及びヒツジ抗ヒトGPXを1:500(Biodesign)で使用した。二次抗体は、1:2000で使用されるこれらのそれぞれのIgG−HRPコンジュゲートである。データは、(β−アクチンレベルに標準化された)対照に対するタンパク質バンドの強度として報告しており、対照を100パーセントとみなした。CAT、MnSOD及びCu/ZnSOD、n=7;GPX、n=4。対照のパーセントとしてデータを変換する前に、有意性を決定するために、等分散を仮定した対応のある片側t−検定を行った。結果を表1に示している。
[0095]カタラーゼの酵素活性は、Cayman Chemical Co.、Ann Arbor、MI製のアッセイキットを製造業者の指示に従って使用して決定した。このアッセイは、酵素活性の決定のためにCATの過酸化作用を利用し、最適濃度のH2O2の存在下におけるメタノールとの酵素の反応に基づいた。産生されたホルムアルデヒドを、4−アミノ−3−ヒドラジノ−5メルカプト−1,2,4−トリアゾール(パーパルド)をクロマゲンとして用いて分光光度法で測定した。パーパルドは、アルデヒドと二環式ヘテロ環を特異的に形成し、これは、酸化を受けて無色から紫色に変わる。この吸光度を、分光光度的プレートリーダーを使用して540nmで読み取った。結果は、与えられた式を使用することによって既知の試料の標準曲線から算出した。そこから、CATの酵素活性を、nmol/分/ml CATとして導いた。試料は、同じものを3つずつで行った。有意性を決定するために、等分散を仮定した片側t−検定を行った。結果を表2に示している。
[0096]Ambion Inc.(Austin、TX、USA)製のRNAクエオス(RNAqueous)(登録商標)キットを使用して全RNAを単離した。簡単に説明すると、処理した細胞を、(オーブン中、37℃で)トリプシン処理して、手持ち式電気ホモジナイザーによって(4℃で)均質化した。次いで、RNAクエオス(登録商標)キットによって提供されるプロトコールに従って細胞溶解液の全RNAの単離を行い、−80℃で保管した。得られたRNAを、エタノール沈澱を用いて精製して約1000ng/ulに濃縮した。このRNAを、DEPC水中に再懸濁し、DNアーゼI処理して、残留したDNAを除去した。Ambion製のDNAフリー(DNA−free)(商標)DNアーゼ処理キット(カタログ#1906)を、製造業者の指示に従ってDNアーゼI処理に使用した。この精製したRNAを、ナノドロップ分光光度計を使用して定量し、RNAナノチップキット(カタログ#5067−1511)のプロトコールに従って、Agilentの2100バイオアナライザ(Bioanalyzer)を使用して完全性の確認をする。
Claims (14)
- 動物においてカタラーゼ欠損によって引き起こされる疾患を予防又は治療するための医薬組成物であって、前記疾患は無カタラーゼ血症又は肝癌であり、前記組成物は、動物においてカタラーゼの産生又は活性を増大させるのに有用であり、1種又は複数の型のビタミンDと、動物においてカタラーゼの産生若しくは活性を増大させるための、又は活性酸素種を減少させる若しくは排除するための少なくとも1種のさらなる作用剤とが配置されている生物学的に許容される媒体を含む、医薬組成物。
- ビタミンDが、カルシトリオール(1a,25−ジヒドロキシビタミンD3)、セオカルシトール(EB1089、1a,25−ジヒドロキシ−22,24−ジエン−24,26,27−トリホモビタミンD)、1α,25−ジヒドロキシ−16−エン−23−イン−ビタミンD3(類似体V)、ビタミンD3、25(OH)ビタミンD3、24,25(OH)2ビタミンD3、パリカルシトール、1,25−ジヒドロキシ−16,23Z−ジエン−26,27−ヘキサフルオロ−D3、1,25−ジヒドロキシ−16,23E−ジエン−26,27−ヘキサフルオロ−19−ノル−D3、22−オキサ−カルシトリオール、1α−(OH)D5、20−エピ−1α,25(OH)2D3、カルシポトリオール、1α24(S)(OH)2D2、1α,24(R)(OH)2D3、及びKH1060のうちの1種又は複数である、請求項1に記載の医薬組成物。
- 1ng/kg体重〜10mg/kg体重をもたらすのに十分な量のビタミンDを含む、請求項1に記載の医薬組成物。
- 前記さらなる作用剤が1種又は複数の抗酸化剤である、請求項1に記載の医薬組成物。
- 1種又は複数の薬学的に許容される担体、賦形剤、又は添加剤を含む医薬組成物として製剤化されている、請求項1に記載の医薬組成物。
- 1種又は複数のプロバイオティクス、プレバイオティクス、又はこれらの組合せをさらに含む、請求項1に記載の医薬組成物。
- プロバイオティクスがエンテロコッカス・フェシウムSF68である、請求項6に記載の医薬組成物。
- 動物においてカタラーゼ欠損によって引き起こされる疾患を予防又は治療するための薬剤を調製するためのビタミンDの使用であって、前記疾患は無カタラーゼ血症又は肝癌であり、前記薬剤は動物においてカタラーゼの産生又は活性を増大させるのに有用な薬剤である、使用。
- ビタミンDが、カルシトリオール(1a,25−ジヒドロキシビタミンD3)、セオカルシトール(EB1089、1a,25−ジヒドロキシ−22,24−ジエン−24,26,27−トリホモビタミンD)、1α,25−ジヒドロキシ−16−エン−23−イン−ビタミンD3(類似体V)、ビタミンD3、25(OH)ビタミンD3、24,25(OH)2ビタミンD3、パリカルシトール、1,25−ジヒドロキシ−16,23Z−ジエン−26,27−ヘキサフルオロ−D3、1,25−ジヒドロキシ−16,23E−ジエン−26,27−ヘキサフルオロ−19−ノル−D3、22−オキサ−カルシトリオール、1α−(OH)D5、20−エピ−1α,25(OH)2D3、カルシポトリオール、1α24(S)(OH)2D2、1α,24(R)(OH)2D3、及びKH1060のうちの1種又は複数である、請求項8に記載の使用。
- 前記薬剤が、プロバイオティクス、プレバイオティクス、又はこれらの組合せをさらに含む、請求項8に記載の使用。
