Description
本出願は、米国仮出願第61/016,265号(2007年12月21出願)の利益を請求するものである。
本発明の方法において使用される試料は、液体培地内に懸濁される液体試料又は固体試料であり得る。好ましくは、固体試料は、物理的に(例えば、均質化)及び/又は化学的に(例えば、界面活性剤と混合することによって)のいずれかで処理して、液体培地内に標的微生物を懸濁することができる。液体試料は、浮遊固体の濃度又は大きさが、試料が表面型膜フィルタを通って濾過されるのを防止しないという条件で、浮遊固体を含有してもよい。液体又は浮遊固体試料は、濾過の前に好適な溶媒(例えば、滅菌水又は緩衝液)中で希釈されてもよい。
標的微生物は、液体若しくは固体試料をフィルタの表面上に移動させることによって、又は膜フィルタを通して液体試料を濾過することによって、膜フィルタ上に収集することができる。試料を膜フィルタ上に収集した後、フィルタは、培養デバイスに移動させることができる。膜フィルタは、標的微生物の成長又は代謝活性に対して抑制的ではない、表面型微多孔性膜フィルタであり得る。膜フィルタは、例えば、セラミック酸化アルミニウム、トラックエッチポリカーボネート、又はトラックエッチポリエステルから作製されてよい。好適な膜フィルタは、培養デバイス内の水和培地等の水和培地と接触している時に実質的に透明であるフィルタをも含む。本明細書において使用される時、「実質的に透明な」膜フィルタは、培養デバイス内の微生物成長の観測結果又は兆候の画像化(例えば、コロニー、pHインジケータ、気泡、酵素反応の生成物又は中間体)を顕著に歪ませないか、又は損なわない膜フィルタを指す。好適な膜フィルタの非限定的な例としては、約60μmの厚さ、約25〜50%の多孔性、及び約1.6の屈折率を有する、商標名ANOPOREでWhatman Inc.(Florham Park、NJ)によって販売されるセラミック膜フィルタ、並びに約10〜20μmの厚さ、約15%の多孔性、及び約1.6の屈折率を有する、商標名NUCLEOPOREでWhatman Inc.によって販売されるトラックエッチポリカーボネートフィルタが挙げられる。
様々な培養デバイスが、本発明の方法において使用できる。いくつかの実施形態では、培養デバイスは、細菌の存在を検出することができる。代替の実施形態では、培養デバイスは、酵母及び/又はかびの存在を検出することができる。ある実施形態では、培養デバイスは、ウイルスの存在を検出することができる。
表面フィルタ、濾過ユニット、及び培養デバイスは、梱包材料と組み合わせ、試料内の微生物を検出するための試料調製システム(キット)として販売することができる。例えば、試料調製システムは、併せてパッケージ化された、2つ以上の構成要素(例えば、表面フィルタ及び培養デバイス)、又は3つ以上の構成要素(例えば、表面フィルタ、濾過ユニット、及び培養デバイス)を含んでもよい。ある実施形態では、濾過ユニットは、表面フィルタの取り外し用に構成されることができる。
培養デバイス内の微生物コロニーを数えるための自動システムは、当該技術分野において既知である。そのような自動システムは、概して、撮像システム、コロニー数を決定する画像分析アルゴリズム、並びにコロニー数データ及び画像を表示し、任意に、格納し、操作するためのデータ管理システムを含む。寒天平板上のコロニーを数えるための例示的なシステムは、商標名PROTOCOLで、Synbiosis(Cambridge、UK)によって販売され、米国特許第6,002,789号に見られる。PETRIFILMプレート上のコロニーを数えるためのシステムは、米国特許第5,403,722号、同第7,298,885号、及び同第7,298,886号に記載される。
本開示のいくつかの培養デバイスは、培養デバイス内に存在する微生物を明確に特定するための、選択的及び/又は分化試薬等の手段を提供する。他の培養デバイスは、培養デバイス内に存在する微生物の暫定的特定を提供することができる。そのような暫定的特定が行われる時、時折、付加的な試験を行うことによって、微生物の同一性を確認することが望ましい。本開示の方法は、確証試験を提供する。

Claims (1)
- ガス産生微生物を検出するための方法であって、
ガス産生微生物を含有する疑いがある試料、表面フィルタ、及び発酵性栄養素を含む培地を含有する平坦フィルム培養デバイスを提供する工程と、
前記試料からのガス産生微生物を前記表面フィルタ上に収集する工程と、
前記表面フィルタを前記培地と接触させる工程と、
前記培地と接触している前記表面フィルタを一定期間培養する工程と、
ガス産生微生物の存在を検出する工程と、
を含み、
前記表面フィルタが、セラミック酸化アルミニウムを含み、
前記ガス産生微生物の存在を検出する工程が、コロニーを検出する工程を含み、
前記コロニーが、光学的に検出され、
前記ガス産生微生物の存在を検出する工程が、前記コロニーに隣接した気泡を検出する工程を更に含む、方法。
