Description
疎水性のタンパク性物質のコーティングされた粒子を製造する本発明の方法は、タンパク性物質を脱油されたリン脂質組成物と接触させる特徴がある。本発明の方法及び製品の好ましい実施態様は添付の特許請求の範囲に要約され、以下にさらに詳述する。
本明細書における「タンパク性物質(proteinaceous material)」は、あらゆるタンパク質、ペプチド、アミノ酸及びアミノ酸エステルと理解され、パルミチン酸、ミリスチン酸、炭水化物等と共役したタンパク質等の糖化タンパク質及びリポタンパク質等の、さらに少量の他の物質(例えば25重量%未満、好ましくは5重量%未満)を含むことができる。かかる他の物質は、タンパク性部分のみがタンパク性物質として見なされる。好ましくは、かかるタンパク性物質は10個以下のアミノ酸残基の長さの鎖を有する遊離アミノ酸及び/又はペプチドを少なくとも80重量%、好ましくは少なくとも90重量%を含有する。すなわち、アミノ酸残基が10個を超えるペプチド及びタンパク質が20重量%未満、好ましくは10重量%未満である。より好ましくは、タンパク性物質は遊離アミノ酸及びジペプチドの合計を少なくとも60重量%、特には少なくとも80重量%、さらに少なくとも95重量%、最も好ましくは少なくとも98重量%含む。特別な実施態様においては、タンパク性物質は遊離アミノ酸を少なくとも50重量%、好ましくは少なくとも75重量%、より好ましくは95重量%含む。特には、原則的に遊離アミノ酸のみを含む。コーティング前には、タンパク性物質が結晶形を有していることが好ましい。
本明細書中で使用される「リン脂質組成物」は、次に規定される、例えば少なくとも40重量%又は具体的には少なくとも50重量%のかなり大きな割合のリン脂質を含むあらゆる組成物である。かかるリン脂質組成物は、さらにリン脂質以外の脂質、例えば、トリグリセリド、ジグリセリド、モノグリセリド、脂肪酸、糖脂質、炭水化物、水等を含んでもよい。しかしながら、重量%が示されている箇所では、無水ベースで示されており、すなわち水の不在下であることを仮定して示されている。リン脂質組成物の実例はレシチンである。中性脂肪(トリグリセリド等)と水は、通常レシチンのアセトン可溶性部分の一部であるが、極性脂質(リン脂質、糖脂質)は通常アセトン不溶性である。リン脂質組成物は好ましくは水に分散させて使用される。
本発明の方法は、パンコーター(pan coater)、流動床コーター(fluid bed coater)等の従来機を使用して実施することができる。その後、アミノ酸又はペプチドの粉末を、適した溶剤中でレシチンの分散とに接触させる。適した溶剤は、任意で追加成分が溶解、分散されている水を含む。かかる追加の成分は、加工助剤、緩衝材、ミネラル、微量元素、一部のビタミン及びスクロース又は他の非還元糖等の炭水化物を含む。溶剤中のリン脂質の濃度は、例えば1重量%と臨界ミセル濃度との間であるべきで、それは追加される成分によって異なり、当業者には既知の方法によって決定することができる。好ましくは、リン脂質濃度が15重量%未満であり、特に好ましくは2〜10重量%である。
コーティングされた粒子は包装され、製品として使用することができる。製品は摂取前に摂取者が、単独、或いは他の成分と組合わせて、食品又は飲料に溶解又は懸濁することができる。
[実施例]
96%を超えるアセトン不溶性物質を含む脱油された大豆レシチン(Cargill Texturizing Solutions社製)を最終濃度が5.9重量%になるまで水に分散した。99%純ロイシン50kg(Rexim社製)を流動床造粒機(Aeromatic AG、Niro Inc.社製)に配置した。流動化を床温34℃にて開始し、0.143l/分の速度で分散されたレシチンを30分間噴霧した。その後、粉末を熱風(70℃)に10分間くぐらせることで乾燥させ、その後26℃まで冷却した。その後、造粒機を停止し、粉末を2000μmメッシュの篩を使用して篩過し、袋詰めにした。
図2は、本実施例によりコーティングされたロイシンの粉末の電子顕微鏡像(200倍)を示す。湿潤時間は120秒よりも長かった。国際IDFスタンダード87:1979法を用いた測定によれば、粉末は水において良好な分散性を有した。
粉末のサプリメントがフェニールケトン尿症及び高フェニルアラニン血症の食事管理のために調製された。サプリメントは栄養的に完全ではない。100gあたり以下を含む:

Claims (12)
- 遊離アミノ酸とジペプチドとの合計を少なくとも60重量%含み、1.0kJ/molより高い疎水性を有する疎水性のタンパク性物質のコーティングされた粒子の製造方法であって、前記タンパク性物質を、脱油されたリン脂質組成物の全乾燥重量に対し20%未満のトリグリセリドを含む脱油されたリン脂質組成物と接触させることを含む方法。
- タンパク性物質が、遊離アミノ酸とジペプチドとの合計を少なくとも80重量%含むことを特徴とする、請求項1に記載の方法。
- タンパク性物質が、メチオニン、トリプトファン、フェニルアラニン、ロイシン、イソロイシン及びバリンから選択されるアミノ酸を総タンパク量に対して少なくとも25重量%、及び/又は上記アミノ酸を数で少なくとも50%含むペプチドを含むことを特徴とする、請求項1又は2に記載の方法。
- タンパク性物質が、総タンパク量に対して、遊離アミノ酸又はそれらの塩を少なくとも75重量%含むことを特徴とする、請求項1〜3のいずれか1項に記載の方法。
- 脱油されたリン脂質組成物が、トリグリセリドを10重量%未満含むことを特徴とする、請求項1〜4のいずれか1項に記載の方法。
- 脱油されたリン脂質組成物が、リン脂質を少なくとも65重量%含むことを特徴とする、請求項1〜5のいずれか1項に記載の方法。
- 脱油されたリン脂質組成物が、リン脂質と糖脂質とを合わせて少なくとも70重量%含むことを特徴とする、請求項1〜6のいずれか1項に記載の方法。
- 遊離アミノ酸及びジペプチドの合計を少なくとも60重量%含み、1.0kJ/molよりも高い疎水性を有するコーティングされたタンパク性物質であって、前記タンパク性物質の合計重量の0.1〜1.5%の脱油されたリン脂質組成物のコーティングを有し、前記脱油されたリン脂質組成物が乾燥重量の20%未満のトリグリセリドを含むことを特徴とするコーティングされたタンパク性物質。
- 0.001〜0.1重量%のリン含有量を有する、請求項8に記載のコーティングされたタンパク性物質。
- 1又は複数の炭水化物、タンパク質、ビタミン及びミネラルから選択される食物成分と、請求項8又は9に記載のコーティングされたタンパク性物質、又は請求項1〜7のいずれか1項に記載の方法で得られた、コーティングされたタンパク性物質とを組み合わせることを含む、栄養組成物の製造方法。
- 炭水化物、タンパク質、ビタミン及び/又はミネラルを含む栄養組成物であって、さらに請求項8又は9に記載のコーティングされたタンパク性物質を炭水化物、タンパク質及び任意の脂質の合計に対して、0.1〜50重量%含む栄養組成物。
- 炭水化物、タンパク質及び任意の脂質の合計に対して、請求項8又は9に記載のコーティングされたタンパク性物質を0.5〜30重量%含む、請求項11に記載の栄養組成物。
