Description
表1に示す成分をトルエンに混合して、その混合物を攪拌して均一な溶解物を得た。次にこの溶解物をPETフィルム上に展延し、乾燥機によりトルエンを揮発除去させた後に支持体(PET編布)上にかぶせて粘着剤層を支持体上に圧着転写させて貼付剤を得た。なお、表1に示す処方の数値は「質量%」を指す。粘着基剤としてスチレン−イソプレン−スチレンブロック共重合体を、粘着付与剤として脂環族飽和炭化水素樹脂を、可塑剤として飽和炭化水素系可塑剤を、それぞれ使用した。
実施例3及び比較例1の貼付剤の薬物含量安定性を以下の方法で評価した。すなわち、製剤を60℃で1ヶ月、40℃で1ヶ月及び3ヶ月、25℃で3ヶ月間それぞれ保存した後、保存後の製剤中におけるリドカインの含量を液体クロマトグラフィーにより測定し、処方量に対する薬物含有率(%)を算出した。なお、測定は3回繰り返して行い、得られた結果の平均値を表2に示す。表2の結果から明らかなように、溶解剤として2−オクチルドデカノールを添加することにより、薬物含量の低下が抑制されることが確認された。
実施例3及び比較例1の貼付剤の薬物放出率安定性を以下の方法で評価した。すなわち、製剤を60℃で1ヶ月もしくは40℃で1ヶ月それぞれ保存した後、溶出試験機を用いて(溶出液を精製水として)保存後の製剤からリドカインを12時間放出させた。回収した溶出液中のリドカインの含量を液体クロマトグラフィーにより測定し、処方量に対する薬物含有率(%)を算出した。なお、測定は5回繰り返して行い、得られた結果の平均値を図1及び図2に示す。溶解剤として2−オクチルドデカノールを添加することによって、薬物放出率の低下が抑制されることが確認された。
表5に示す化合物に対するリドカインの溶解度(25℃)を次の方法により測定した。すなわち、各溶解剤に飽和以上となるようにリドカインを添加して十分に攪拌した後、その混合液をろ過してろ液中のリドカイン濃度を液体クロマトグラフィーにより測定し、飽和溶解度(%)を算出した。なお、測定は3回繰り返して行い、得られた結果の平均値を表5に示した。
Claims (5)
- 支持体上に粘着剤層を備える貼付剤であって、
前記粘着剤層は、リドカイン及びその薬学的に許容される塩からなる群より選ばれる少なくとも1種の薬物とアルキルアルカノールとを含有し、
前記アルキルアルカノールは、オクチルドデカノール及びイソステアリルアルコールから選択される少なくとも1種であって、その含有量は、前記粘着剤層の全質量基準で4〜12質量%である貼付剤。 - 前記アルキルアルカノールは、2−アルキルアルカノールである請求項1に記載の貼付剤。
- 前記薬物に対する前記アルキルアルカノールの比は、質量比で0.8〜2である請求項1又は2に記載の貼付剤。
- 前記粘着剤層は、熱可塑性エラストマーを含有する請求項1〜3のいずれか一項に記載の貼付剤。
- 前記熱可塑性エラストマーは、スチレン−イソプレン−スチレンブロック共重合体である請求項4記載の貼付剤。
