Description
本発明は、一般的には、抗菌組成物に関する。特に、本発明は、無水クエン酸二水素銀とクエン酸とを含む組成物を提供する。本発明は、その組成物を生成するプロセスを提供する。本発明は、上記固体組成物を使用する抗菌方法を提供する。
抗菌金属イオン溶液が、殺菌剤として使用されている。銀溶液は、冷却塔(cooling tower)、用水プール(swimming pool)、病院の温水システム、携帯型水システム、およびスパプール(spa pool)において、殺菌剤として使用されている。さらに、銀イオン溶液は、創傷の処置のために調製されている。しかし、提唱される方法において使用される銀イオンは、不安定である。その銀イオンは、創傷部位へと治療用量を送達するためには、その創傷付近にて生成されなければならない。
本発明は、無水クエン酸二水素銀とクエン酸とを含む組成物を提供し、その組成物を生成するプロセスを提供する。そのプロセスは、水性クエン酸二水素銀のストック溶液を凍結させる工程、その後、その凍結ストック溶液を凍結乾燥させて、無水クエン酸二水素銀を生成する工程を包含する。
本発明は、無水クエン酸二水素銀とクエン酸とを含む組成物を提供する。驚くべきことに、この無水クエン酸二水素銀組成物は、有効な抗菌活性を有する安定なクエン酸二水素銀溶液を生成するために、一定期間後に再構成され得ることが、見出された。この無水組成物は、保存が容易であり経済的であるという利点を提供する。この無水組成物はまた、クエン酸二水素銀水溶液よりも輸送が経済的であるという利点を提供する。この無水組成物は、水または別の水性希釈剤を用いて簡便に再構成され得るので、この無水組成物は、クエン酸二水素銀水溶液に対する簡便な代替法を提供する。
水を、逆浸透ユニット中に導入し、半透膜を通して通過させて不純物を除去し、そして脱イオン水を生成した。99%純度の無水クエン酸を、上記水と混合し、200ガロンの20%(重量/体積)(水1ガロン当たりクエン酸796g)溶液を生成した。この200ガロンの20%クエン酸を、陽極および陰極を有するイオンチャンバ中に入れた。これらの電極の各々は、200トロイオンスの999純銀からなった。これらの陽極および陰極を、少なくとも2.0mm離して間隔をあけ、上記クエン酸溶液がこの2つの電極の間を通過することを可能にした。陽性伝導体および陰性伝導体を備えるイオン発生コントローラ(ion generation controller(IGC))電源を、上記陽極および陰極に取り付けた。そのIGCで、17ボルトで5アンペアの電流を印加し、9秒毎にパルス化し、1分間間隔で極性を変化させた。上記プロセス全体にわたって、5アンペア−17ボルトの出力を維持するために、電極間隙を調整した。電流フローは、イオン電流が上記陽極と陰極との間を流れることを引き起こし、上記希釈されたクエン酸溶液中に遊離の銀イオンを生成した。この銀イオンは、上記クエン酸溶液中のクエン酸と反応して、クエン酸二水素銀溶液を生成した。20%のクエン酸溶液を、所望の銀イオン濃度が得られるまで、1分間あたり50ガロンで、144時間、上記イオンチャンバを通して再循環させた。次いで、このクエン酸二水素銀溶液を、上記手順の間に形成されたあらゆる固体を沈殿させるために、静置した。得られた生成物は、2400ppmの銀イオン濃度を有するクエン酸二水素銀溶液であった。本明細書中で今後は、この溶液をストック溶液と呼ぶ。このストック溶液は、以下の実施例ですぐに使用され得るし、後の使用のために保存もされ得る。
上記実施例1で調製したストック溶液の1,000mlサンプルを、SSAバッチ番号04.06.03から入手し、イオン選択性電極(ion specific electrode(ISE))を備えるOrionTM 290Aプロセッサを使用して、このサンプルが2400ppmのイオン性銀の濃度を有することを確認した。
次いで、実施例2からの結晶の粉末を、約794mlの薬学等級の純水を使用して、1,000ml Pyrexガラスフラスコ中で再構成した。次いで、この溶液を、磁気的スターラーによって30分間撹拌し、遮光性保存キャビネット中に24時間置いた。
クエン酸二水素銀組成物の防腐剤としての効力を実証するために、実施例1に記載したクエン酸二水素銀のストック溶液を、防腐剤負荷試験に供した。この防腐剤負荷試験を、カテゴリー2の生成物(水性基剤またはビヒクルを用いて生成された局所的に使用される生成物、無菌性でない経鼻生成物、粘膜に適用されるエマルジョンを含むエマルジョン)についてのヨーロッパ薬局方試験法4.04/5.01.03.00に従って、実施した。

Claims (14)
- クエン酸二水素銀とクエン酸とを含む、無水の抗菌用組成物であって、該組成物は、クエン酸二水素銀と該クエン酸二水素銀に対して少なくとも5倍モル過剰のクエン酸とを含むストック水溶液を凍結乾燥することにより調製され、
該組成物が該ストック水溶液の体積と等しい体積の水中での希釈により再構成された場合に、該ストック水溶液と等価な濃度のクエン酸二水素銀を含む安定な水溶液が得られることを特徴とする、組成物。 - 請求項1に記載の組成物であって、該組成物は、0.5%未満の水を含む、組成物。
- 請求項1に記載の組成物であって、該組成物は、結晶性である、組成物。
- 請求項1に記載の組成物であって、前記ストック水溶液は2,400ppmの銀イオン濃度を有するクエン酸二水素銀溶液である、組成物。
- 請求項1〜4のうちのいずれか1項に記載のクエン酸二水素銀とクエン酸とを含む無水の抗菌用組成物を生成するプロセスであって、該プロセスは、
(a)クエン酸二水素銀とクエン酸と水とを含む前記ストック水溶液を提供する工程;
(b)該ストック水溶液を凍結させて凍結溶液を形成する工程;および
(c)該凍結溶液を凍結乾燥させて、該無水組成物を形成する工程;
を包含する、プロセス。 - 請求項5に記載のプロセスであって、前記工程(c)は、前記凍結溶液を加熱する工程をさらに包含する、プロセス。
- 抗菌量のクエン酸二水素銀を含む安定な抗菌水溶液を調製する方法であって、該方法は、
請求項1〜4のうちのいずれか1項に記載の組成物を、水性希釈剤で希釈する工程
を包含する、方法。 - 請求項7に記載の方法であって、前記水性希釈剤は、実質的に純粋な水である、方法。
