Description
精製水1kgに以下の物質を溶解し、シース液を調製した。
トリス(ヒドロキシメチル)アミノメタン 1.51g
塩酸 6mol
EDTA−2K 0.2g
レオドールTW0120V 0.75g
トミサイドS 0.2g
ProxelGXL 0.2g
屈折率調整剤としてチオグリセリンを用いた場合、シース液に57g/Lの濃度で添加すればよいことが判った。
屈折率調整剤としてジグリセリンを用いた場合、シース液に69g/Lの濃度で添加すれば屈折率が1.342に調整されることが判った。
屈折率調整剤としてポリグリセリン#310を用いた場合、シース液に71g/Lの濃度で添加すれば屈折率が1.342に調整されることが判った。
屈折率調整剤としてポリグリセリン#750を用いた場合、シース液に73g/Lの濃度で添加すれば屈折率が1.342に調整されることが判った。
屈折率調整剤としてポリグリセリン#500を用いた場合、シース液に75g/Lの濃度で添加すれば屈折率が1.342に調整されることが判った。
屈折率調整剤として硫酸ナトリウムを用いた場合、シース液に62g/Lの濃度で添加すれば屈折率が1.342に調整されることが判った。
屈折率調整剤として塩化ナトリウムとグリセリンとを用いた場合、塩化ナトリウムを7.1g/L、グリセリンを69.1g/Lの濃度で添加すれば屈折率が1.342に調整されることが判った。
屈折率調整剤として塩化ナトリウムと硫酸ナトリウムとを用いた場合、塩化ナトリウムを7.1g/L、硫酸ナトリウムを54.44g/Lの濃度で添加すれば屈折率が1.342に調整されることが判った。
屈折率調整剤として塩化ナトリウムと硫酸ナトリウムとグリセリンとを用いた場合、塩化ナトリウムを7.1g/L、硫酸ナトリウムを28.45g/L、グリセリンを34g/Lの濃度で添加すれば屈折率が1.342に調整されることが判った。
実施例1のシース液に種々の屈折率調整剤を添加してシース液の屈折率を1.342に調整し、電気伝導度を測定した。電気伝導度の測定には、電気伝導度計DS−8F(堀場製作所)を用いた。
上記シース液に屈折率調整剤として7.1g/Lの塩化ナトリウムと28.45g/Lの硫酸ナトリウムと34g/Lのグリセリンとを添加してシース液Bを調整した。シース液Bの電気伝導度は、35mSであった。
上記シース液に屈折率調整剤として79g/Lのグリセリンを添加してシース液Cを調整した。シース液Cの電気伝導度は、1.1mSであった。
実施例2で調整したシース液Aを用いて、塩の析出が見られるか否かを検討した。
実施例2で調整したシース液Bを用いて、塩の析出が見られるか否かを検討した。対照として、実施例3で調製したシース液Dを用いた。
実施例2で調製したシース液Bを細菌分析装置BACSYS-40i(シスメックス製)に適用し、精度管理物質であるバクトチェック(シスメックス製)を試料として、試料中の細菌に相当する疑似粒子(以下、疑似細菌とする)と白血球に相当する疑似粒子(以下、疑似白血球とする)を測定した。
シース液Bを用い、実施例5と同様にして尿試料116検体(検体番号1〜116)の細菌数及び白血球数を測定した。また、対照としてバクトシースを用い、116検体の細菌数及び白血球数を測定した。
Claims (5)
- 尿中の粒子を分析する装置に用いられる粒子分析装置用シース液であって、
グリセリン類及び塩化ナトリウムを含む屈折率調整剤と、
水と、
pHを6.0〜8.5に調整するための緩衝剤と、を含有し、
塩化ナトリウムの濃度が20g/L以下であり、
25℃でのナトリウムD線(λ=589.3nm)の波長における屈折率が1.338〜1.345であり、
25℃での電気伝導度が40mS以下である、ことを特徴とするシース液。 - 前記グリセリン類が、グリセリン、チオグリセリン及びポリグリセリンからなる群より選択される、請求項1記載のシース液。
- 塩化ナトリウムの濃度が10g/L以下である、請求項1又は2に記載のシース液。
- 前記緩衝剤が、トリス(ヒドロキシメチル)アミノメタン−塩酸及びトリス(ヒドロキシメチル)アミノメタン−マレイン酸からなる群より選択される、請求項1〜3の何れかに記載のシース液。
- 界面活性剤と、キレート剤と、防カビ剤又は防菌剤とをさらに含む、請求項1〜4の何れかに記載のシース液。
