Description
本出願は、全体を引用例として本明細書に取り込む、2003年6月3日に出願された同時係属中の米国特許仮出願第60/475,288号(事件番号BYRK−27PR)に基づく優先権を主張する。
本開示は、体液試料を光学的に検査するための装置及び方法に関し、具体的には、装置で使用するための読み取りヘッドに関する。さらに具体的には、本開示は、蛍光分光分析及び反射分光分析の両方を実施するための部品を含む読み取りヘッドに関する。
様々な医学的診断目的にとって、反射分光器を使用して体液試料を分析して、たとえば人の尿又は血液の色を測定することが有用である。公知のとおり、分光分析は、吸収性種の吸光度と濃度との間の直線関係(ベールの法則)を使用して試料の含有量を測定する。未知の分析対象物濃度は、試料が吸収する光の量を計測し、ベールの法則を適用することによって測定することができる。分析対象物の吸光係数が未知であるならば、標準から導出した吸光度対濃度の検量線を使用することにより、未知の濃度を測定することができる。
本開示は、蛍光分光分析及び反射分光分析の両方を簡単かつ好都合な方法で実施することを可能にする、試料キャリヤ(たとえば、液体試料を有するストリップ又はカセット)に光を照射し、試料キャリヤから光を受けるための、診断機器のための新規で改良された読み取りヘッドの典型的な実施態様に関する。
図1は、医学的診断目的のために体液試料を光学的に検査するための装置の一部として使用するための、本開示にしたがって構成された新規で改良された読み取りヘッド200の典型的な実施態様を示す。
単一回使用のカセットであってもよい。試薬カセット122は、尿のような体液試料が中に配置される開口又はウェル124を有する。試薬カセット122の内部には、ウェル124中に配置された体液試料と反応することができる試薬ストリップ(図示せず)がある。試験の結果に依存して、試薬ストリップは色を変えることができ(たとえば、カラーストライプが現れる)、それは、試薬カセット122の窓128を介して試薬ストリップを見ることによって決定することができる。図示しないが、支持トレー120は、較正を容易にするため、その上面に配置された一定の色、たとえば白の較正チップを有することができる。本開示の新規で改良された読み取りヘッドはまた、蛍光粒子を標識として使用する側方流動イムノアッセイで使用することもできる。
Claims (12)
- 試料キャリヤに光を照射し、試料キャリヤから光を受けるための診断機器のための読み取りヘッドであって、
診断機器に組み込まれるように適合され、試料キャリヤを受けるための照射チャンバを含むハウジングと、
ハウジング内で照射チャンバに対して固定された位置に取り付けられた発光ダイオードを含む反射光源と、
発光ダイオードからの実質的にすべての光を照射チャンバに運んで照射チャンバが実質的に均一に照射を受けるように適合された発光ダイオードから光を受けるようにハウジング中に取り付けられた導光体と、
照射チャンバから光を受けるように結合された光検出器と、
ハウジング中に配置され、所定の波長の励起光を照射チャンバに送るように配置された蛍光励起光源と、
照射チャンバと光検出器との間に配置され、蛍光励起光源からの励起光の通過を防ぐが、励起光の所定の波長と異なる波長を有する、照射チャンバ中の試料キャリヤから出た光の通過を許すように適合された光フィルタと
を含み、
蛍光励起光源が紫外線光源を含み、紫外線光源が、金属箔でライニングされているハウジングのチャンバの中に配置されることを特徴とする、読み取りヘッド。 - 紫外線光源が、364ナノメートル、405ナノメートル及び436ナノメートルのライン出力ならびに330〜385ナノメートルの広帯域出力を有するブラックランプを含む、請求項1記載の読み取りヘッド。
- 光検出器と光フィルタとの間にミラーが配置されている、請求項1又は2記載の読み取りヘッド。
- レンズが、光検出器と光フィルタとの間に配置され、光を光検出器に合焦させるように適合されている、請求項1〜3のいずれか1項記載の読み取りヘッド。
- 導光体が、入口に隣接する地点で比較的小さな幅を有し、出口に隣接する地点で比較的大きな幅を有するダイバージング導光体を含む、請求項1〜4のいずれか1項記載の読み取りヘッド。
- 紫外線光源が発光ダイオードを含む、請求項1記載の読み取りヘッド。
- 発光ダイオードが高輝度緑色発光ダイオードを含む、請求項6記載の読み取りヘッド。
- 反射光源のアレイが、ハウジング内で照射チャンバに対して固定された位置に取り付けられ、第一の波長の実質的に単色性の光を発するための第一の発光ダイオード及び第一の波長と実質的に異なる第二の波長の実質的に単色性の光を発するための第二の発光ダイオードを含み、及び
導光体が、発光ダイオードの一方だけが点灯しているとき、その一方の発光ダイオードからの実質的にすべての光を照射チャンバに運んで照射チャンバが実質的に均一に照射を受けるようにするために、発光ダイオードそれぞれから光を受けるようにハウジング中に取り付けられる、請求項1〜7のいずれか1項記載の読み取りヘッド。 - 一つ以上の反射光源が、一つ以上のレンズレス発光ダイオードを含む、請求項1〜8のいずれか1項記載の読み取りヘッド。
- 請求項1〜9のいずれか1項記載の読み取りヘッドを含み、光検出器に接続され、光検出器からの電気信号を処理して、読み取りヘッドの照射チャンバに受けられた試料キャリヤ上の試料の物理的性質を測定するようにプログラムされたプロセッサをさらに含む診断機器。
- プロセッサが、予測モデルを使用して物理的性質を測定するようにプログラムされている、請求項10記載の診断機器。
- 物理的性質がグルコース濃度を含む、請求項10又は11記載の診断機器。
