Description
1.第1の実施形態
図1は、本発明の第1の実施形態のイムノクロマトグラフィー用試験具の断面図である。このイムノクロマトグラフィー用試験具は、表面に粘着層を有するプラスチック板からなる基材1上に、レーヨンの不織布からなる試料添加用部材3と、グラスファイバーの不織布からなる標識保持部材5と、レーヨンの不織布からなる展開用部材7と、ニトロセルロースの多孔体からなるクロマト用膜担体9と、セルロースの不織布からなる吸収部材11とを備える。標識保持部材5は、試料添加用部材3に接触して配置され、試料中の測定対象と抗原抗体反応する標識物質を保持する。クロマト用膜担体9は、標識保持部材と間隔を介して配置され、測定対象と抗原抗体反応する固定化用物質が固定された判定部を有する。展開用部材7は、標識保持部材5およびクロマト用膜担体9と接触するように配置されている。吸収部材11は、クロマト用膜担体9と接触するように配置されている。
図3は、本発明の第2の実施形態のイムノクロマトグラフィー用試験具の断面図である。この試験具は、第1の実施形態と比べて、展開用部材7を有しない点と、試料添加用部材3が標識保持部材5を覆ってクロマト用膜担体9と接触するように配置されている点が異なっている。これ以外の点は、第1の実施形態と共通している。
以下の方法に従って、クロマト用のクロマト用膜担体、標識保持部材を調製し、さらにこれらを用いて、イムノクロマトグラフィー用試験具を調製した。
図4(クロマト用膜担体の平面図)に示すように、ニトロセルロースメンブレンからなるクロマト用膜担体9の第1判定部9A、第2判定部9B及び対照部9Cに、抗体塗布機(BioDot社)を用いて、それぞれ、リン酸緩衝液(pH7.0)で2.0mg/mLの濃度になるように希釈した抗インフルエンザA型モノクローナル抗体、1.5mg/mLの濃度になるように希釈した抗インフルエンザB型モノクローナル抗体、及びビオチンBSAを塗布し、50℃で30分間乾燥させた。
抗インフルエンザA型モノクローナル抗体を青色着色ポリスチレンラテックス粒子(粒径0.3μm)に感作し、分散用緩衝液(BSA及びシュークロースを含有するリン酸緩衝液(pH7.0))に懸濁し、抗インフルエンザA型モノクローナル抗体感作ラテックス粒子を調製した。
ここで、図5(a)〜(c)を用いて、各部材の基材への貼り付け方法について説明する。図5(a)は、標識保持部材5の右端とクロマト用膜担体9の左端の距離が−3mm(すなわち、両者が3mm重なっている。)の場合であり、図5(b)は、標識保持部材5の右端とクロマト用膜担体9の左端の距離が0mm(すなわち、両者の端が同じ位置にある。)の場合であり、図5(c)は、標識保持部材5の右端とクロマト用膜担体9の左端の距離が3,5,10,20mmの場合を示す。それぞれの場合での各部材の幅(試料が流れる方向の長さ)は、表1に示す。
次に、上記方法で調製した図5の試験具を用いて、測定対象の検出にかかる時間について試験を行った。
(4)次に、(3)の試験管に上記方法で調製した図5の試験具を入れた。
(5)次に、試験具を試験管に入れたまま、しばらく放置し、次に示す項目について測定を行った。その結果を表2に示す。なお、以下の測定項目において、BioDot社のTSR3000メンブレンストリップリーダーで測定したラインの強度が0.015以上0.03未満のものを1+、0.03以上0.08未満のものを2+、0.08以上のものを3+となるような目視判定見本を作製し、試験具に現れたライン強度を該目視判定見本と比較することにより判定を行った。
(a)バックグラウンドがクリアになるまでの時間(BG Clear)

(c)対照部9Cのライン強度が3+になるまでの時間
(d)第1判定部9Aのライン強度が1+になるまでの時間

(f)第2判定部9Bのライン強度が1+になるまでの時間
(g)第2判定部9Bのライン強度が2+になるまでの時間
(h)設定判定時間(試験具を入れてから10分後)の第1判定部9A及び第2判定部9Bの目視スコア
(i)設定判定時間以降のバックグラウンド及び感度の上昇の有無
表2を参照すると、以下のことが分かる。
(1)標識保持部材5とクロマト用膜担体9の間隔を開けるほど、バックグランドがクリアになるまでの時間が早くなった。
(2)標識保持部材5とクロマト用膜担体9の間隔を開けるほど、ラテックスが標識保持材5から溶出完了するまでの時間が早くなった。
(3)標識保持部材5とクロマト用膜担体9の間隔を開けるほど、対照部9C、第1判定部9A、第2判定部9Bのライン出現時間が早くなった。
(4)標識保持部材5とクロマト用膜担体9の間隔を開けるほど、設定判定時間(10分後)以降のバックグラウンドの上昇及び感度の上昇がなくなり、5mm以上間隔を開けることで設定時間内に設定感度まで各ラインのシグナルの上昇が完了した。
以上より、本発明の試験具を用いると、迅速に判定結果が得られることが実験的に確認された。

- 試料添加用部材と、試料中の測定対象と抗原抗体反応する標識物質を保持する標識保持部材と、標識保持部材から試料の展開方向に間隔を介して配置され、測定対象と抗原抗体反応する固定化用物質が固定された判定部を有するクロマト用膜担体とを備え、試料添加用部材が標識保持部材を覆ってクロマト用膜担体と接触するように配置されていることを特徴とするイムノクロマトグラフィー用試験具。
