Claims (11)
- 試料を格納する試料格納容器と、染料を格納する染料格納容器と、濃度の相異なる既知濃度の試料を格納する複数の標準試料格納容器とを含み、当該試料格納容器、当該染料格納容器、及び当該標準試料格納容器のそれぞれには、圧縮空気流入口と連結されており大気圧下で空気は通過させるが液体は通過させない上部疎水性隔壁と、液体流出口と連結されており大気圧下で空気は通過させるが液体は通過させない下部疎水性隔壁と、液体流入口とが設けられて構成されたカートリッジ格納容器部と、
前記試料格納容器の液体流出口と連結された試料流入口、内面に付着された抗体、及び液体放出ポートを有する試料反応部と、前記標準試料格納容器の液体流出口と連結された標準試料流入口、内面に付着された抗体、及び液体放出ポートを有する複数の標準試料反応部と、前記染料格納容器の液体流出口と連結される染料流入口、及び緩衝溶液流入ポートを有する染料/緩衝溶液流入部とを含む微細流動チャンネルを備えるカートリッジと、
前記試料格納容器、染料格納容器、及び標準試料格納容器のそれぞれの圧縮空気流入口と弁を通じて連結される圧縮空気貯蔵部と、
前記緩衝溶液流入ポートと弁を通じて連結される緩衝溶液貯蔵部と、
前記試料反応部及び標準試料反応部での抗原−抗体反応の程度を染料による色変化の程度として測定する検出部と、を含むことを特徴とする生物学的試料中の蛋白質を検出するための蛋白質検出用流動システム。 - 前記上部疎水性隔壁及び下部疎水性隔壁は多孔性であり、前記下部疎水性隔壁の平均気孔が前記上部疎水性隔壁の平均気孔より大きいことを特徴とする請求項1に記載の蛋白質検出用流動システム。
- 前記上部疎水性隔壁の平均気孔の直径は0.2ないし1μmであり、前記下部疎水性隔壁の平均気孔の直径は2ないし20μmであることを特徴とする請求項2に記載の蛋白質検出用流動システム。
- 前記上部疎水性隔壁及び前記下部疎水性隔壁は多孔性ポリテトラフルオロエチレン膜で形成されることを特徴とする請求項1に記載の蛋白質検出用流動システム。
- 前記圧縮空気貯蔵部及び前記緩衝溶液貯蔵部に連結されるポンプをさらに含むことを特徴とする請求項1に記載の蛋白質検出用流動システム。
- 前記試料反応部と前各標準試料反応部とは容積が同一であることを特徴とする請求項1に記載の蛋白質検出用流動システム。
- 前記染料/緩衝溶液流入部の染料流入口から前記試料反応部の液体放出ポートまでの微細チャンネル長と当該染料流入口から前記標準試料反応部の液体放出ポートまでの微細チャンネル長とは同一であることを特徴とする請求項1に記載の蛋白質検出用流動システム。
- 前記染料/緩衝溶液流入部の緩衝溶液流入ポートから前記試料反応部の液体放出ポートまでの微細チャンネル長と当該緩衝溶液流入ポートから前記標準試料反応部の液体放出ポートまでの微細チャンネル長とは同一であることを特徴とする請求項1に記載の蛋白質検出用流動システム。
- 前記圧縮空気流入口と前記圧縮空気貯蔵容器間に連結された弁は、弁が閉じられた場合には圧縮空気流入口を外部空気に通じさせ、弁が開いた場合には圧縮空気流入口を前記圧縮空気貯蔵容器に通じさせる3方向弁であることを特徴とする請求項1に記載の蛋白質検出用流動システム。
- 前記試料反応部及び標準試料反応部のそれぞれの液体放出ポートの開閉を調節する手段をさらに含むことを特徴とする請求項1に記載の蛋白質検出用流動システム。
- 請求項1に記載の蛋白質検出用流動システムを使用して試料中の蛋白質を検出する蛋白質検出方法において、
前記圧縮空気流入口を通じて圧縮空気を供給することによって前記試料格納容器及び標準試料格納容器から試料及び標準試料を試料反応部及び標準試料反応部に供給して抗原−抗体反応を誘導する段階と、
前記緩衝溶液流入ポートを通じて緩衝溶液を供給して前記試料反応部及び標準試料反応部を洗浄する段階と、
前記圧縮空気流入口を通じて圧縮空気を供給することによって前記染料格納容器から染料を染料/緩衝溶液流入部に、次いで、前記試料反応部及び標準試料反応部に供給する段階と、
前記緩衝溶液流入ポートを通じて緩衝溶液を供給して前記試料反応部及び標準試料反応部を洗浄する段階と、
前記試料反応部及び前記標準試料反応部での色変化データを利用して試料中の蛋白質の存在の有無を検出し、かつその濃度を測定する段階と、を含む蛋白質検出方法。
