Abstract
Description
古典的遺伝子再集合及び逆遺伝学の要素を組み合わせる、遺伝子再集合インフルエンザウイルスを産生する方法は、これまでに議論されてきた。例えば、宿主細胞が第1のインフルエンザ株に感染し、第2のインフルエンザ株からの少なくとも1つのセグメントをコードする1つ以上の発現構築物でトランスフェクトされる方法は、参考文献11で論じられている。この方法は、siRNAなどのインフルエンザウイルスセグメントの翻訳及び/または転写を阻害する剤を使用して、siRNAによって標的とされるセグメントが存在しない遺伝子再集合ウイルスを生成するという状況でのみ記載されている。
したがって、本発明の目的は、古典的遺伝子再集合法及び逆遺伝学法の両方に関連する欠点を克服する、遺伝子再集合体インフルエンザウイルスを生成する改善された方法を提供することである。
インフルエンザウイルスは、セグメント化されたマイナス鎖RNAウイルスである。インフルエンザA及びBウイルスのゲノムには、8つの一本鎖ウイルスRNA(vRNA)セグメントが含まれている。インフルエンザA及びBウイルスの8つのゲノムセグメントは、サイズの順に、PB2、PB1、PA、HA、NP、NA、M、NSである。PB2、PB1、PA、NP、M、及びNSは、内部及び非構造タンパク質をコードし、バックボーンセグメントと呼ばれ得る。HA及びNAセグメントは、表面糖タンパク質をコードする。本発明の方法は、遺伝子再集合インフルエンザAウイルスを産生するために使用され得る。あるいは、本発明の方法は、遺伝子再集合インフルエンザBウイルスを産生するために使用され得る。
本発明の方法では、培養宿主が接触する親インフルエンザウイルス株は、ドナー株である。ドナー株は、複製可能である(すなわち、感染後培養宿主内でドナー株自身が増殖できる)。培養宿主をドナー株と接触させることにより、ウイルスが培養宿主に感染したときに、ドナー株のゲノムセグメントが培養宿主に送達される。
本発明の方法において、1つ以上のインフルエンザ遺伝子を含む1つ以上の発現構築物は、培養宿主に導入される。1つ以上の発現構築物は、既知の逆遺伝学技術からの発現構築物(複数可)を導入するための任意の方法を使用して、細胞培養物に導入され得る。
ドナー株と接触した培養宿主を培養することにより産生され、第2のHA遺伝子または第2のNA遺伝子を含む1つ以上の発現構築物が導入された遺伝子再集合ウイルスは、ドナー株からのインフルエンザ遺伝子及び1つ以上の発現構築物によってコードされる第2のHA遺伝子または第2のNA遺伝子を含む。好ましい実施形態では、ドナー株と接触した培養宿主を培養することにより産生され、第2のHA遺伝子及び第2のNA遺伝子を含む1つ以上の発現構築物が導入された遺伝子再集合ウイルスは、ドナー株からのインフルエンザ遺伝子及び1つ以上の発現構築物によってコードされる第2のHA遺伝子及び第2のNA遺伝子を含む。さらなる実施形態では、ドナー株と接触した培養宿主を培養することにより産生され、第2のHA遺伝子、第2のNA遺伝子、及び第2のバックボーン遺伝子(すなわち、PB1、PB2、PA、M、NSまたはNP遺伝子)を含む1つ以上の発現構築物が導入された遺伝子再集合ウイルスは、ドナー株からのインフルエンザ遺伝子及び1つ以上の発現構築物によってコードされる第2のHA遺伝子、第2のNA遺伝子、及び第2のバックボーン遺伝子を含む。
本発明の方法で使用するための培養宿主は、発育鶏卵または細胞であり得る。好ましい実施形態では、培養宿主は、細胞培養物である。
ウイルスが細胞株上で単離及び/または成長している場合、DNAの任意の潜在的な発がん活性を最小限に抑えるために、最終ワクチン中の残存細胞株DNAの量を最小限に抑えることが標準的な実施である。
本発明は、ワクチンを製造するための方法に従って製造されたウイルスを利用する。
インフルエンザワクチンは通常、生ウイルスまたは不活化ウイルスのいずれかに基づく。不活化ワクチンは、全ビリオン、「スプリット」ビリオン、または精製された表面抗原に基づいてもよい。抗原は、ビロソームの形態で提示され得る。本発明は、これらの種類のワクチンのいずれかを製造するために使用することができる。

水中油型エマルジョンは、インフルエンザウイルスワクチンのアジュバント添加での使用に特に好適であることが見出されている。様々なそのようなエマルジョンが知られており、それらは典型的には、少なくとも1つの油及び少なくとも1つの界面活性剤を含み、油(複数可)及び界面活性剤(複数可)は、生分解性(代謝可能)及び生体適合性である。エマルジョン中の油滴は一般に直径5μm未満であり、サブミクロンの直径を有し得る。これらの小さいサイズは、安定したエマルジョンを提供するためにマイクロフルイダイザーで達成される。平均サイズが220nm未満の液滴は、フィルタ滅菌を行うことができるため、好ましい。
界面活性剤の好ましい量(重量%)は、ポリオキシエチレンソルビタンエステル(Tween(登録商標)80など)0.01~1%、特に約0.1%;オクチルまたはノニルフェノキシポリオキシエタノール(Triton(登録商標) X-100、またはTriton(登録商標)シリーズの他の洗浄剤など)0.001~0.1%、特に0.005~0.02%;ポリオキシエチレンエーテル(ラウレス9など)0.1~20%、好ましくは0.1~10%、特に0.1~1%または約0.5%である。
スクアレン、トコフェロール、及びTriton(登録商標)洗浄剤(Triton(登録商標) X-100など)のエマルジョン。本エマルジョンはまた、3d-MPLを含み得る(下記参照)。本エマルジョンは、リン酸緩衝液を含み得る。
本発明の組成物(またはキット構成要素)に好適な容器としては、バイアル、シリンジ(例えば、使い捨てシリンジ)、鼻スプレーなどが挙げられる。これらの容器は、滅菌されているべきである。

293T細胞は、Melbourne Universityから入手し、10%FBS、1XGlutaMAX(Gibco)及び1X抗生物質/抗真菌剤(Gibco)を含むDMEM中で維持した。MDCK(WHO)細胞は、WHOから入手し、10%FBS、1XGlutaMAX(Gibco)及び1X抗生物質/抗真菌剤(Gibco)を含むDMEM中で維持した。MDCK33016PF細胞[79]は、化学的に定義された培地(Lonza)で維持した。
PR8(H1N1)、A/Wyoming/3/2003(H3N2)、及びA/Indonesia/NIHRD11771/2011(H5N1)のHA及びNAをpHW2000ベクター[80]に含むプラスミドは、標準分子生物学技術を使用して生成した。H6N1の場合、H6(A/turkey/Massachusetts/3740/1965)及びN1(A/Brisbane/59/2007及びA/California/07/2009)をコードするプラスミドは、標準分子生物学技術を使用して生成した。
高成長の親ウイルス(PR8及びA/Texas/1/1977)は、培養宿主の感染前に精製した。各親インフルエンザウイルス株について、感染した尿膜液2mlを0.45μmフィルタでろ過し、Microsep 300K Omega遠心分離装置(PALL)を使用して約1400xgで1時間遠心分離することによって、精製した。保持されたウイルスをリン酸緩衝生理食塩水(PBS)-で希釈し、きれいなMicrosep装置を使用して再度遠心分離した。保持されたウイルスをPBS-を使用して1mlまで希釈し、100μlのアリコートで-80℃で凍結した。
遺伝子再集合インフルエンザウイルスを産生するためのインフルエンザ遺伝子の時差送達は、プラスミドDNAによる293T/MDCK共培養物のトランスフェクション、次いで、トランスフェクション1~24時間後のHGPによる感染によって行った。TransIT-293(Mirus)、Lipofectamine 2000(Invitrogen)、またはLipofectamine 3000(Invitrogen)トランスフェクション試薬を製造業者の指示に従って使用して、ウイルスHA及びNAを発現する1~2μgのプラスミドによる293T/MDCK細胞のトランスフェクションを行った。
293T細胞及びMDCK細胞(WHOまたはMDCK 33016PF、浮遊または接着)の共培養物に、精製HGPウイルス(10~20μl/トランスフェクション(6ウェルプレート)のニート精製HGPウイルス)と1~2μgのウイルスのHA及びNAをコードするプラスミドDNAのそれぞれと同時にトランスフェクトした。トランスフェクション試薬TransIT-293(Mirus)、Lipofectamine 2000(Invitrogen)、またはLipofectamine 3000(Invitrogen)は、製造業者の指示に従って使用した。TransIT-293を使用してトランスフェクトさせた細胞は、トランスフェクションの約3時間後に洗浄した。TPCK-トリプシン(1μg/ml)は、前述のとおりトランスフェクション後1日目、またはトランスフェクションの4~8時間後に添加した。トランスフェクション後3~6日目に上清を回収した。
その後、トランスフェクション上清は、HGPウイルスに対して作成した抗血清の存在下で卵内で継代させた。簡潔に説明すると、1回目の抗血清継代では、発育卵に200μl/卵のニートトランスフェクション上清を接種し、1時間後に200μl/卵のトリプシン過ヨウ素酸塩処理抗血清(適宜、PR8またはA/Texas/1/1977に対して作成)を接種した。2回目の抗血清継代では、1回目の抗血清継代から回収した感染尿膜液を、HA力価に従って希釈し、等量の抗血清と共にRTで1時間インキュベート後、卵を200μl/卵で接種した。一部のトランスフェクションでは、限界希釈によってウイルスをクローン化した。
H5N1遺伝子再集合ウイルスは、高成長親株(A/Texas/1/1977)と、A/Indonesia/NIHRD11771/2011のHA及びNA遺伝子をコードするプラスミドを使用して生成した。
遺伝子再集合H1N1ウイルス(A/PR/8/1934)は、高成長親株(A/Texas/1/77)と、A/PR/8/1934のHA及びNA遺伝子をコードするプラスミドを使用して生成した。
遺伝子再集合H3N1ウイルスは、H1N1 HGPウイルス(A/PR/8/1934)と、A/Wyoming/3/2003のHA及びNA遺伝子をコードするプラスミドを使用して生成した。
遺伝子再集合H6N1ウイルスは、H3N2 HGPウイルス(A/Texas/1/1977)と、H6(A/turkey/Massachusetts/3740/1965)及びN1(A/Brisbane/59/2007及びA/California/07/2009)遺伝子をコードしているプラスミドを使用して生成した。
293T/接着MDCK 33016PF細胞の共培養物は、HA及びNAコードプラスミド及びHGPで同時にトランスフェクトさせた(表4)。
準種を産生するためにインフルエンザウイルスを複製する状況で発生し得る遺伝的多様性は、古典的遺伝子再集合または逆遺伝学(RG)によって得られたインフルエンザAウイルス中に存在する非表面インフルエンザタンパク質の配列を比較することによって評価した。10個のRGウイルスのプール(元はSeqirus、CDC、NIBSCによってレスキューされた)からのバックボーン遺伝子の核酸配列のシーケンシングを行い、次に翻訳してタンパク質配列を決定した。これらの配列は、古典的遺伝子再集合(Seqirus、NYMC、及びNIBSCによって生成された遺伝子再集合)によって得られた16のH1N1及び32のH3N2ウイルスの対応する配列と比較した。

Claims (42)
- 遺伝子再集合インフルエンザウイルスを生成するための方法であって、
(i)培養宿主を、第1の赤血球凝集素(HA)遺伝子及び第1のノイラミニダーゼ(NA)遺伝子を含む親インフルエンザウイルス株と接触させるステップと;
(ii)前記培養宿主に、1つ以上のインフルエンザ遺伝子を含む1つ以上の発現構築物を導入するステップであって、前記インフルエンザ遺伝子が、第2のHA遺伝子または第2のNA遺伝子を含む、前記ステップと;
(iii)遺伝子再集合ウイルスを産生するために前記培養宿主を培養するステップと;
(iv)前記第2のHA遺伝子または前記第2のNA遺伝子を含む遺伝子再集合ウイルスを選択するステップと;を含む、前記方法。 - 前記第2のHA遺伝子または前記第2のNA遺伝子を含む遺伝子再集合インフルエンザウイルスを単離するステップ(v)をさらに含む、請求項1に記載の方法。
- 前記単離された遺伝子再集合インフルエンザウイルスが、7:1の遺伝子再集合インフルエンザウイルスである、請求項2に記載の方法。
- 前記1つ以上のインフルエンザ遺伝子が、第2のHA遺伝子及び第2のNA遺伝子を含み、ステップ(iv)が、前記第2のHA遺伝子及び/または前記第2のNA遺伝子を含む遺伝子再集合ウイルスを選択することを含む、請求項1に記載の方法。
- (i)前記第2のHA遺伝子及び/または(ii)前記第2のNA遺伝子を含む遺伝子再集合インフルエンザウイルスを単離するステップ(v)をさらに含む、請求項4に記載の方法。

- (a)請求項1~38のいずれか1項に記載の方法によって、遺伝子再集合インフルエンザウイルスを調製するステップと、(b)前記遺伝子再集合インフルエンザウイルスからワクチンを調製するステップと、を含む、ワクチンを調製する方法。
