Abstract
Description
(b)前記(a)段階で透明化させた生体組織試料に酵素反応混合液およびプライマーを添加して、検出しようとするバイオマーカーを等温で増幅させる段階と、
(c)前記(b)段階で増幅させたバイオマーカーを検出する段階と、を含む、生体組織の3次元核酸映像診断方法を提供する。
(ロ)前記固定された試料を組織クリアリング溶液に入れて反応させる段階と、
(ハ)前記組織クリアリング溶液に反応させた試料を洗浄溶液に入れて試料に付着している有機物を洗い落とす段階と、を含むことができる。
(b)前記(a)段階で透明化させた生体組織試料に酵素反応混合液およびプライマーを添加して、検出しようとするバイオマーカーを等温で増幅させる段階と、
(c)前記(b)段階で増幅させたバイオマーカーを検出する段階と、を含む、生体組織の3次元核酸映像診断方法を提供する。
(ロ)前記固定された試料を組織クリアリング溶液に入れて反応させる段階と、
(ハ)前記組織クリアリング溶液に反応させた試料を洗浄溶液に入れて試料に付着している有機物を洗い落とす段階と、を含むことができる。
マウスから脳を摘出して3mmのサイズにスライスした後、スライスした脳試料を固定溶液に入れ、4℃で12時間の間反応させた。
(2−1.マウス脳でのThy−1 mRNA等温核酸増幅結果の確認)
前記実施例1で透明化させたマウス脳試料にBst DNAポリメラーゼおよび逆転写酵素を含む酵素反応混合液を添加して、4℃で24時間の間振とうし、組織で等温増幅が可能であるかを判断するために、ニューロン、胸腺細胞、T細胞、および幹細胞マーカーとしてマウス脳組織神経で多く発現するThy−1(Thymocyte differentiation antigen 1)mRNAを等温で増幅させた。
前記実施例2−1の方法と同じ方法でマウス脳試料のGAD−67(Glutamic acid decarboxylase 67)mRNAを等温で増幅させた。この際に使用したGAD67プライマー(プローブを含む)は、下記表4に示した。
Claims (13)
- (a)生体組織試料を透明化させる段階と、
(b)前記(a)段階で透明化させた生体組織試料に酵素反応混合液およびプライマーを添加して、検出しようとするバイオマーカーを等温で増幅させる段階と、
(c)前記(b)段階で増幅させたバイオマーカーを検出する段階と、を含む、生体組織の3次元核酸映像診断方法。 - 前記(b)段階で使用されたプライマーは、増幅させようとするバイオマーカーのプライマーであることを特徴とする、請求項1に記載の生体組織の3次元核酸映像診断方法。
- 前記プライマーは、プローブを含むことを特徴とする、請求項2に記載の生体組織の3次元核酸映像診断方法。
- 前記生体組織は、脳、肝臓、肺、腎臓、腸、心臓、筋肉、または血管であることを特徴とする、請求項1に記載の生体組織の3次元核酸映像診断方法。
- 前記(a)で透明化させる段階は、(イ)生体組織試料を固定溶液に入れて固定させる段階と、
(ロ)前記固定された試料を組織クリアリング溶液に入れて反応させる段階と、
(ハ)前記組織クリアリング溶液に反応させた試料を洗浄溶液に入れて試料に付着している有機物を洗い落とす段階と、を含むことを特徴とする、請求項1に記載の生体組織の3次元核酸映像診断方法。 - 前記固定溶液は、スクロースを含むことを特徴とする、請求項5に記載の生体組織の3次元核酸映像診断方法。
- 前記スクロースは、濃度が20%(w/v)〜100%(w/v)であることを特徴とする、請求項6に記載の生体組織の3次元核酸映像診断方法。
- 前記組織クリアリング溶液は、CHAPS(3−[(3−コラミドプロピル)ジメチルアンモニオ]−1−プロパンスルホナート)、尿素、および塩化ナトリウム(NaCl)よりなる群から選ばれた一つ以上を含むことを特徴とする、請求項5に記載の生体組織の3次元核酸映像診断方法。
- 前記塩化ナトリウム(NaCl)は、濃度が0.001〜1.0%(w/v)であることを特徴とする、請求項8に記載の生体組織の3次元核酸映像診断方法。
- 前記洗浄溶液は、リン酸緩衝生理食塩水(PBS)およびアジ化ナトリウムを含むことを特徴とする、請求項5に記載の生体組織の3次元核酸映像診断方法。
- 前記アジ化ナトリウムは、濃度が0.001〜0.5%(w/v)であることを特徴とする、請求項10に記載の生体組織の3次元核酸映像診断方法。
- 前記バイオマーカーは、分子バイオマーカーであることを特徴とする、請求項1に記載の生体組織の3次元核酸映像診断方法。
- 前記分子バイオマーカーは、DNAまたはRNAであることを特徴とする、請求項12に記載の生体組織の3次元核酸映像診断方法。
