Description
本願は、2010年12月3日出願の米国特許仮出願第61/419,660号の利益を請求し、参照により全体として本明細書に組み込まれる。
本明細書に開示されるのは、網膜の疾患の治療方法であって、クロザピン、n−デスメチルクロザピン、オランザピン、オランザピンのある代謝産物、および後述の本明細書に記載されるような種々の他の化合物を、そのような治療の必要がある患者へ投与することによって、網膜に作用する方法である。
から成る群から選択される化合物を、それを必要とする患者に投与することを含む、方法を開示する。
本発明の化合物は、網膜の疾患を治療するために使用されてもよい。出願人による「網膜の疾患」とは、酸化ストレスによって生じる、または悪化される、網膜またはそれを囲繞する組織の任意の病気を意味する。これらは、黄斑変性、糖尿病性網膜症、脈絡膜新生血管膜、黄斑浮腫(類嚢胞黄斑浮腫および黄斑の腫れとも称される)、網膜上膜(黄斑ひだ)、黄斑円孔、網膜色素変性、黄斑変性(スターガルト病若年性黄斑変性、ベスト卵黄状ジストロフィ、錐体ジストロフィ、および網膜色素上皮のパターンジストロフィ等)、網膜剥離、網膜の外傷、それらと関連付けられた網膜腫瘍および網膜疾患、網膜色素上皮の先天性肥大、急性後部多発性斑状網膜色素上皮症、ならびに急性網膜色素上皮炎を含む。
本発明の方法は、クロザピン、オランザピン、またはオランザピンのある代謝産物を治療の必要がある患者へ投与することによって、網膜の疾患を治療する。本化合物は、よく知られている。
本発明の化合物は、経口、経皮(例えば、パッチの使用を通して)、経鼻、舌下、直腸、または非経口経路のいずれかを介して投与されてもよい。ある好ましい実施形態では、化合物は、眼内への注入によって送達される。
硝子体内注射(IVT)のために、動物は、青色光暴露の1時間前にオランザピン(以下で示される構造を有する)を受けた。局所投与のために、薬物は、24時間(BID)および青色光暴露の1時間前(QI)に与えられた。水は、並行対照溶媒IP/IVT注入または局所投与のために使用された。ラットは、4時間、約6〜7kのルクス強度を伴う青色光に暴露される前、24時間、暗順応させた。青色光暴露の直後、通常の室内灯に戻す前、ラットは、さらに3日間、再び暗順応させた(12時間点灯/12時間暗闇)。光干渉断層撮影(OCT)が、青色光暴露後、7〜10日において、青色光によって生じた網膜の厚さの変化を評価するために使用された。エラーバーは、SEMである(図3)。
Claims (6)
- 酸化ストレスによって生じる、または悪化される、網膜障害を治療するための方法であって、
から成る群から選択される化合物を、それを必要とする患者に投与することを含む、方法。 - 前記網膜障害は、湿潤および乾燥型加齢黄斑変性、網膜色素変性、スターガルト病錐体ジストロフィおよび網膜色素上皮のパターンジストロフィ、黄斑浮腫、網膜剥離、網膜の外傷、腫瘍と関連付けられた網膜腫瘍および網膜疾患、網膜色素上皮の先天性肥大、急性後部多発性斑状網膜色素上皮症、ならびに急性網膜色素上皮炎から成る群から選択される、請求項1に記載の方法。
- 前記網膜障害は、湿潤および乾燥型加齢黄斑変性、網膜色素変性、スターガルト病錐体ジストロフィおよび網膜色素上皮のパターンジストロフィ、網膜色素上皮の先天性肥大、急性後部多発性斑状網膜色素上皮症、ならびに急性網膜色素上皮炎から成る群から選択される、請求項2に記載の方法。
- 前記化合物は、経口投与される、請求項1に記載の方法。
- 前記化合物は、眼内にそれを注入することによって投与される、請求項1に記載の方法。
- 前記化合物は、眼に局所的に投与される、請求項1に記載の方法。
