アリールブロミド(1.0当量)、Cs2CO3(1.5当量)、及びアミン(1.2当量)をトルエンに溶解した。アミンを塩として用いた場合、更なる当量のCs2CO3を用いた。更に、臭化アリールを塩(イソキノリンのHCl−又はTFA−塩)として使用した場合は、更に1当量のCs2CO3を追加して使用した。溶液はアルゴンで脱気し、フラッシングした。その後、Pd(OAc)2(0.03当量)及びBINAP(0.045当量)を加え、反応が完了するか、又はこれ以上向上が達成できなくなるまで、この溶液を100℃で加熱した。生成物を単離するため、この溶液を室温に冷却し、濾過し、濾液を蒸発させた。残留物を水に溶解し、酢酸エチルで抽出した。有機層を分離し、硫酸マグネシウムで乾燥し、溶媒を減圧下で除去した。粗生成物を分取型HPLCにより精製した。一般法Aに従って、以下の化合物を合成した(表1):
Boc−保護化合物をメタノールに溶解し、同容積の2NのHClを加えた。完全な変換が確認されるまで、この溶液を室温で攪拌した。減圧下で溶媒を除去し、残留物をH2Oに溶解した。最後に凍結乾燥を行って所望の生成物を塩酸塩として得た。一般手順Aに従って、以下の化合物を塩酸塩として合成した(表2)。
PMB−及びBoc−保護化合物の場合、出発物質をトリフルオロ酢酸に溶解し、室温で1時間攪拌し、次いでマイクロ波中140℃で3時間加熱した。単離のため、この溶媒を減圧下で除去し、残留物を分取型HPLCにより精製した。生成物画分を合わせて蒸発させ、所望の生成物をトリフルオロ酢酸塩として得た。いくつかのケースでは、この生成物を、2NのHClに溶解し、蒸発させた。水溶液を最後に凍結乾燥した後、これらの所望の生成物を塩酸塩として単離した。

Rhoキナーゼ阻害を測定するために、IC50値を以下のプロトコールに従って測定した。
活性ヒト組換えROCK II(N−末端His6標識組換えヒトROCK−II残基11−552)は、Upsate Ltd.社(Dundee、英国)から購入した。ペプチド基質、フルオレセイン−AKRRRLSSLRA−COOHはJPT Peptide Technologies社(Berlin、ドイツ)から入手した。アデノシン−5′−三リン酸(ATP)、ウシ血清アルブミン(BSA)、ジメチルスルホキシド(DMSO)、4−(2−ヒドロキシエチル)ピペラジン−1−エタンスルホン酸(Hepes)、Brij−35及びジチオスレイトール(DTT)は、Sigma‐Aldrich社(Munich、ドイツ)から購入した。トリス(ヒドロキシメチル)−アミノメタン(Tris)、塩化マグネシウム、NaOH、1MのHCl及びEDTAはMerck Biosciences社(Darmstadt、ドイツ)から入手した。「完全」プロテアーゼ阻害剤は、Roche Diagnostics社(Mannheim、ドイツ)からのものであった。
4、5mM MgCl2、2mMのDTT及び0.02%(w/v)BSA)中で100ng/mlの濃度に希釈した。ペプチド基質及びATPを緩衝液2中で、それぞれ3μM及び120μMの濃度に希釈した。2μlの化合物を、384ウェルの小容量マイクロタイタープレート(Greiner社(Bio‐One、Frickenhausen、ドイツ))中で2μlの希釈酵素と混合し、そしてペプチド基質とATPを含有する2μlの溶液の添加によって酵素反応を開始した。32℃で60分インキュベーションした後、100mMのHepes−NaOH、pH7.4、0.015%(v/v)Brij−35、45mMのEDTA及び0.227%のチップコーティング試薬1(Caliper Lifescience Inc社(Hopkinton、MA))を含有する溶液(20μl)の添加により反応を停止した。次いで基質ペプチドのリン酸化を、基本的にPommereau et al (J.Biomol. Screening 9 (5), 409-416, 2004)によって記述された通りCaliper 3000装置で検出した。分離条件は以下の通りである:圧力−1.3psi、上流電圧−1562V、下流電圧−500V、試料シップ時間−200ms。陽性対照(化合物の代わりに緩衝液1)及び陰性対照(化合物の代わりに緩衝液1及びROCK IIの代わりに緩衝液2)を各プレートで並列に作動させた。
