Abstract
【解決手段】分子構造中の側鎖脂肪酸に少なくとも1つの水酸基を有する新規マンノシルエリスリトールリピッドを製造するために、マンノシルエリスリトールリピッドを生産する能力を有する微生物を、少なくとも1つの水酸基を有する脂肪酸、脂肪酸誘導体または植物油を含む培地にて培養する。

にて示されるマンノシルエリスリトールリピッドであることを特徴としている。
下記の一般式(1)にて示されるマンノシルエリスリトールリピッド、つまり、
にて示されるマンノシルエリスリトールリピッドの製造方法であって、マンノシルエリスリトールリピッドを生産する能力を有する微生物を、少なくとも1つの水酸基を有する脂肪酸、脂肪酸誘導体または植物油を含む培地にて培養することを特徴としている。
従来型のMELは、MEL生産菌の培養によって得られ、下記の一般式(2)に示すように、その化学構造は、4−O−β−D−マンノピラノシル−meso−エリスリトールまたは1−O−β−D−マンノピラノシル−meso−エリスリトールをその基本構造とするものである。
本発明のMELの製造方法は、マンノシルエリスリトールリピッドを生産する能力を有する微生物を用いることを特徴としている。なお、本明細書において「マンノシルエリスリトールリピッドを生産する能力を有する微生物」とは、従来型のMEL(MEL−A、MEL−B、MEL−CおよびMEL−D)の少なくとも1つを生産することができる微生物が意図される。
本発明のマンノシルエリスリトールリピッドの製造方法は、マンノシルエリスリトールリピッドを生産する能力を有する微生物を培養することによって、当該微生物に新規マンノシルエリスリトールリピッドを生産させる方法である。
本発明のマンノシルエリスリトールリピッドの製造方法では、上記微生物が、少なくとも1つの水酸基を有する脂肪酸、脂肪酸誘導体または植物油を含む培地にて培養される。
本発明のマンノシルエリスリトールリピッドの分子構造は、基本的には上記一般式(1)における置換基R1の脂肪族アシル基の炭素数、あるいは二重結合および水酸基の有無などが異なる各種化合物の混合物の形態にて得られる。したがって、本発明のマンノシルエリスリトールリピッドの製造方法では、微生物を培養した後、マンノシルエリスリトールリピッドを精製することが好ましい。
本発明のマンノシルエリスリトールリピッドの製造方法によって得られる新規MELの構造解析は従来公知の方法によって行うことができ、特に限定されるものではない。
保存培地(酵母エキス 5g/L、大豆エキス 5g/L、NaNO3 5g/L、グルコース 40g/L、寒天30g/L)に保存しておいたPseudozyma tsukubaensis NBRC 1940株を、2mLの液体培地(グルコース 40g/L、酵母エキス 5g/L、大豆エキス 5g/L、NaNO3 5g/L)が入った試験管に、1白金耳接種した。
培養終了後、菌体培養液に対して等量の酢酸エチルを加え、上下に撹拌した。撹拌後、菌体培養液と酢酸エチルとの混合溶液を静置させ、酢酸エチル層と菌体培養液層とを分離させた。分離後、酢酸エチル層を回収し、当該酢酸エチル層に対して等量の飽和食塩水を加えた。その後、酢酸エチル層と飽和食塩水との混合溶液を上下に撹拌させ、その後、当該混合溶液を静置させて、酢酸エチル層と飽和食塩水層とを分離させた。そして、分離後、酢酸エチル層を回収した。
上述した方法によって得たMELの構造と従来型のMELの構造とを、薄層クロマトグラフィーによって比較した。
10mgの新規MEL(MEL−B(CL))を1mlの5%塩酸メタノールに溶解し、95℃にて30分間反応させた。その後、当該反応液に1mLの水を加えて、反応を止めた。
新規MEL(MEL−B(CL))の界面活性能について検討した。表面張力の測定には、協和界面科学社製のdrop master DM500を用い、添付のプロトコールにしたがって、様々な濃度のMEL−B(CL)の表面張力を測定した。
ヒマシ油を用いて微生物を培養することによって得られた新規MEL−B(MEL−B(CL))の構造を更に詳細に検討するために、1H NMR測定、13C NMR測定、および二次元NMR測定によって、MEL−B(CL)の構造解析を行った。
保存培地(麦芽エキス 3g/L、酵母エキス 3g/L、ペプトン 5g/L、グルコース 10g/L、寒天 30g/L)に保存しておいたPseudozyma antarcticaKM-34(FERMP-20730)株を、2mlの液体培地(グルコース 20g/L、酵母エキス 1g/L、硝酸ナトリウム 1g/L、リン酸2水素カリウム 0.5g/L、硫酸マグネシウム 0.5g/L)が入った試験管に1白金耳接種した。
Pseudozyma parantarctica JCM 11752株を用いて、上述したPseudozyma antarcticaKM-34(FERMP-20730)株の場合と同じ組成の培地を用い、34℃の温度にてジャー・ファーメンターを用いてPseudozyma parantarcticaの培養を行った。なお、以下に具体的な手順について説明する。
Claims (6)
- 下記の一般式(1)にて示されるマンノシルエリスリトールリピッド。
- 下記の一般式(1)にて示されるマンノシルエリスリトールリピッドの製造方法であって、
マンノシルエリスリトールリピッドを生産する能力を有する微生物を、少なくとも1つの水酸基を有する脂肪酸、脂肪酸誘導体または植物油を含む培地にて培養することを特徴とするマンノシルエリスリトールリピッドの製造方法。
- 前記脂肪酸が、リシノール酸であることを特徴とする請求項2に記載のマンノシルエリスリトールリピッドの製造方法。
- 前記植物油が、ヒマシ油であることを特徴とする請求項2に記載のマンノシルエリスリトールリピッドの製造方法。
- 前記微生物は、シュードザイマ属に属する微生物であることを特徴とする請求項2〜4の何れか1項に記載のマンノシルエリスリトールリピッドの製造方法。
- 前記シュードザイマ属に属する微生物は、Pseudozyma tsukubaensis、Pseudozyma antarctica、Pseudozyma aphidis、Candida sp. SY16、Pseudozyma parantarctica、Pseudozyma rugulosa、Pseudozyma fusiformata、Pseudozyma hubeiensis、Pseudozyma shanxiensis、Pseudozyma graminicola、またはPseudozyma crassaであることを特徴とする請求項5に記載のマンノシルエリスリトールリピッドの製造方法。
