- 【特許請求の範囲】 1.非毒性でありかつ1ケ月未満に生分解可能である生物適合性コラーゲン材 料であって、該材料が、コラーゲンと、該コラーゲンに対して化学的に非反応性 である少くとも1の親水性高分子添加物と、を含み、ここで該コラーゲンが、そ のヘリックス構造を少くとも部分的に失っておりかつ架橋されていることを特徴 とするコラーゲン材料。 2.1週間未満に生分解可能であることを特徴とする請求項1に記載のコラー ゲン材料。 3.前記コラーゲンが、37℃超に加熱することにより改変されていることを特 徴とする請求項1又は2のいずれかに記載のコラーゲン材料。 4.酸化的開裂により改変されており、かつ前記コラーゲンに対して化学的に 非反応性である少くとも1の親水性高分子添加物の存在下で架橋されているコラ ーゲンを含むことを特徴とする請求項1〜3のいずれか一に記載のコラーゲン材 料。 5.前記コラーゲンが、過ヨウ素酸又はその塩のうちの1つを用いて酸化的開 裂により改変されていることを特徴とする請求項4に記載のコラーゲン材料。 6.前記コラーゲンに対して化学的に非反応性である親水性高分子添加物の存 在下でβ照射又はγ照射により架橋されていることを特徴とする請求項1〜3の いずれか一に記載のコラーゲン材料。 7.前記親水性高分子添加物が、3000ダルトンより大きい分子量を有すること を特徴とする先の請求項のいずれか一に記載のコラーゲン材料。 8.前記親水性添加物が、3000〜20,000ダルトンの間の分子量を 有する親水性ポリマーであることを特徴とする先の請求項のいずれか一に記載の コラーゲン材料。 9.前記親水性添加物がポリエチレングリコールであることを特徴とする先の 請求項のいずれか一に記載のコラーゲン材料。 10.前記親水性添加物が、デンプン、デキストラン及びセルロースから好まし くは選択されるポリサッカライドであることを特徴とする請求項1〜7のいずれ か一に記載のコラーゲン材料。 11.コラーゲン及び少くとも1の親水性高分子添加物を、1/1〜9/1、好 ましくは2/1〜4/1、より好ましくは3/1のコラーゲン/親水性添加物比 に従って含むことを特徴とする先の請求項のいずれか一に記載のコラーゲン材料 。 12.グリセロールを更に含むことを特徴とする先の請求項のいずれか一に記載 のコラーゲン材料。 13.フィルムの形態で存在することを特徴とする先の請求項のいずれか一に記 載のコラーゲン材料。 14.潜在的に接着性であることを特徴とする請求項1〜13のいずれか一に記載 のコラーゲン材料。 15.前記材料がβ照射にかけられることを特徴とする請求項1〜5及び7〜14 のいずれか一に記載のコラーゲン材料。 16.前記材料が、25キログレイ超又はそれに等しい投与量のβ照射にかけられ ることを特徴とする請求項15に記載のコラーゲン材料。 17.非毒性、潜在的に接着性かつ1ケ月未満、好ましくは1週間未満で生分解 可能である生物適合性コラーゲン材料を得るための方法であって、 a)コラーゲン溶液を調製するステップと; b)該溶液を処理してコラーゲンを少くとも部分的にそのヘリッ クス構造を失わせるステップと; c)生じたコラーゲン溶液を、存在するコラーゲンに対して化学的に非反応性 である少くとも1の親水性高分子添加物を含む溶液と混合するステップと; d)該溶液を架橋して要求されるコラーゲン材料を得るステップと、 を含むことを特徴とする方法。 18.ステップa)において、酸化的開裂により改変されたコラーゲンの溶液が 調製されることを特徴とする請求項17に記載の方法。 19.ステップa)において、5〜50g/lの濃度を有するコラーゲンの酸溶液 が調製され、次に環境温度において、1〜10-5Mの濃度の過ヨウ素酸又はその塩 のうちの1つの溶液と混合されることを特徴とする請求項18に記載の方法。 20.ステップb)において、コラーゲンの溶液が、37℃超の温度で加熱される ことにより処理されることを特徴とする請求項17〜19のいずれか一に記載の方法 。 21.ステップb)において、コラーゲンの溶液が、40〜50℃の間の温度に加熱 されることを特徴とする請求項20に記載の方法。 22.ステップc)において、前記親水性添加物が、3000ダルトンより大きい分 子量を有することを特徴とする請求項17〜21のいずれか一に記載の方法。 23.ステップc)において、前記親水性添加物が、3000〜20,000ダルトンの間 の分子量を有する親水性ポリマーであることを特徴とする請求項17〜22のいずれ か一に記載の方法。 24.ステップc)において、前記親水性添加物が、ポリエチレングリコールで あることを特徴とする請求項17〜23のいずれか一に記載の方法。 25.ステップc)において、前記親水性添加物が、デンプン、デキストラン及 びセルロースから好ましくは選択されるポリサッカライドであることを特徴とす る請求項17〜24のいずれか一に記載の方法。 26.ステップc)において、親水性添加物の濃度が、コラーゲンの濃度より2 〜10倍低いことを特徴とする請求項17〜25のいずれか一に記載の方法。 27.ステップc)において、グリセロールが更に添加されることを特徴とする 請求項17〜26のいずれか一に記載の方法。 28.グリセロールの濃度が3〜8g/lであることを特徴とする請求項27に記 載の方法。 29.ステップd)において、前記混合物が中性pHに中和されることを特徴とす る請求項18〜28のいずれか一に記載の方法。 30.ステップd)において、前記混合物が、β又はγ放射により架橋されるこ とを特徴とする請求項17及び20〜28のいずれか一に記載の方法。 31.ステップd)から得られたコラーゲン材料が、β放射での照射にかけられ ることを特徴とする請求項17〜29のいずれか一に記載の方法。 32.前記コラーゲン材料が、25キログレイ超又はそれに等しい投与量でのβ照 射にかけられることを特徴とする請求項31に記載の方法。 33.ヘリックス構造を少くとも部分的に失い、かつ酸化的開裂により改変され ているコラーゲンと、少くとも1の親水性添加物と、適切なら、グリセロールと 、を含む溶液からフィルムが形成されることを特徴とする請求項17〜32のいずれ か一に記載の方法。 34.前記溶液の薄層が、実質的に平らである不活性支持体に実質 的に均一に適用され、前記コラーゲンが架橋され、そして次に形成されたフィル ムが前記支持体から分離されることを特徴とする請求項33に記載の方法。 35.前記溶液が、2〜6%のコラーゲンと、0.6〜2%の親水性添加物と、0.3 〜0.8%のグリセロールと、を含み、前記不活性支持体に適用される薄層が、0.0 5〜0.3g/cm2の密度を有することを特徴とする請求項33又は34のいずれかに記 載の方法。 36.前記架橋されたコラーゲンが、照射により滅菌するステップの前に、湿っ た環境での37℃でのインキュベーションにかけられ、得られる材料が2〜4週間 以内に生分解可能であることを特徴とする請求項12〜24のいずれか一に記載の方 法。
