Abstract
1-Hydroxy-N-(2-O-acetamidophenyl-ethyl) - 2 -naphthamide is prepared by acetylating b -phenethylamine with acetic anhydride, nitrating the product and separating the o- and p-nitro compounds, de-acylating the o-nitro derivative to give the amine and reacting this with phenyl 1-hydroxy-2-naphthoate to give 1-hydroxy-N-(2-O-nitrophenyl-ethyl)-2-naphthamide, reducing the nitro group to an amine and acylating the product with acetic anhydride. An alternative preparation is to prepare ethyl o-nitrophenyl-pyruvate by reacting diethyl oxalate with o-nitrotoluene and hydrolyse this to the free acid, which is then reacted with hydroxylamine hydrochloride, made basic with caustic soda and heated with sulphuric acid to give o-nitrophenyl-acetonitrile. The nitro group is then hydrogenated and the amino group acylated to give o-acetamidophenyl-acetonitrile. Hydrogenation using a nickel catalyst then gives o-acetamidophenyl-ethylamine which is condensed with phenyl 1-hydroxy-2-naphthoate to give the required product.
