Abstract
The invention comprises Raugustine of the formula <FORM:0833149/IV (b)/1> its esters, and salts thereof, together with a process for isolating weakly basic alkaloids of the raunescine series wherein an aqueous solution of a weak acid containing alkaloids of the raunescine group in admixture with other alkaloids from Rauwolfia plants is extracted with benzene (or other organic solvent having similar extraction properties for the alkaloids) to remove the reserpine alkaloids and the greater part of the ajmalicine alkaloids, the residual aqueous solution is extracted with ethylene chloride (or other organic solvent of similar action) to obtain the raunescine alkaloids, which are isolated if desired in pure form by adsorption, elution or crystallization of the mixture so obtained. Detailed examples illustrate the extraction of raugustine, raunescine, isoraunescine, reserpiline and pseudoreserpine from various Rauwolfia species, and the preparation of 18-O-acetyl-raugustine and raugustine nitrate. The conversion of raugustine to 17-acetyl-pseudorescopine by treatment with pyridine hydrochloride, then sodium methylate, acetylation to methyl 17-acetyl pseudoreserpic acid ester and esterification with trimethoxybenzoyl chloride is described.
