Abstract

The invention comprises thiadiazoles of the general formula <FORM:0815188/IV (b)/1> and their mineral acid addition salts (wherein R is a 2-thienyl, o-chlorophenyl or o-tolyl radical), and their preparation by cyclising thiosemicarbazones of the formula R-CH = N-NHCSNH2 (prepared from R.CHO and thiosemicarbazide) with aqueous ferric chloride. The thiadiazoles may also be prepared by cyclising N1-acyl-thiosemicarbazides of the formula R-CONHNHCSNH2 (prepared from R.COCl and thiosemicarbazide) with concentrated sulphuric acid. Examples describe the preparation of 2-amino-5-(2-thienyl) - 1,3,4 - thiadiazole, 2 - amino - 5 - (o-chlorophenyl) - 1,3,4 - thiadiazole and 2-amino - 5 - (o - tolyl) - 1,3,4 - thiadiazole (and their hydrochlorides) by heating 2-thiophene-aldehyde thiosemicarbazone, o-chlorobenzaldehyde thiosemicarbazone and o-tolualdehyde thiosemicarbazone, respectively, with aqueous ferric chloride at 80 DEG C. 2 - Thiophene - aldehyde - thiosemicarbazone is prepared by reacting ethanolic 2-thiophene-aldehyde with a solution of thiosemicarbazide in dilute hydrochloric acid.ALSO:Tranquillizing compositions are prepared from 5 - substituted - 2 - amino - 1,3,4 - thiadiazoles of the general formula <FORM:0815188/VI/1> or pharmacologically acceptable acid addition salts thereof (wherein R is 2-thienyl, o-chlorophenyl or o-tolyl), and a solid phrmaceutical carrier. Examples describe the preparation of tablets containing 2-amino-5-(2-thienyl)-1,3,4-thiadiazole, cornstarch and magnesium stearate; 2 - amino - 5 - (o - chlorophenyl) - 1,3,4 - thiadi - azole, lactose, talc, and magnesium stearate; and 2 - amino - 5 - (o - tolyl) - 1,3,4 - thiadiazole, lactose, cornstarch, magnesium stearate, talc and a sugar coating.
