Abstract

A vector formed of a modified DNA of bacteriophage lambda , comprises in combination: - mutations, particularly in the Q and S genes, having the effect of preventing or retarding the expression of the late genes, particularly of regulating the production of proteins necessary for the encapsidation of this modified DNA in a bacterium notably E. coli, provided with suppressors of said mutations; - a DNA fragment of bacterial origin comprising a part at least of the Z gene of E. coli and a promotor of a bacterial operon, inserted in a non-essential part of the genome of the phage and a site of cleavage by an endonuclease, particularly EcoRI, in the above- said portion of the Z gene, to the exclusion of any other site of cleavage by the same endonuclease in the above-mentioned modified DNA. This vector enables the insertion in the site of said endonuclease of the gene of a determined protein, and the expression and production of a hybrid protein comprising said determined protein, by bacteria infected with said vector. The mutations in the Q and S genes delay the bacterial lysis and favor the accumulation of numerous copies of the hybrid protein in said bacteria.
