Abstract
1503828 Enumerating bacteria STRATHCLYDE UNIVERSITY OF 22 June 1976 25802/76 Heading C6F A method of staining micro-organisms to facilitate counting comprises the steps of providing a suspension of micro-organisms in a liquid medium, reacting the micro-organisms in the suspended state with phosphate ions whereby phosphate ions react at active sites on the micro-organisms to produce phosphate derivatives of the micro-organisms having at said sites polydentate phosphate groups, and treating the phosphate derivatives with a phosphate-reactive fluorochrome dye (specified) which thereby combines chemically with the microorganisms via the intermediate phosphate groups. Preferably the phosphate derivatives are treated with the fluorochrome dye in the suspended state in liquid medium to produce a liquid specimen for microscopic examination in the liquid state. Alternatively, the phosphate derivatives are applied to a carrier substrate and fixed thereto using a liquid fixative, and the fixed sample is brought into contact with the fluorochrome dye to be stained, the stained sample being dried to provide on the substrate a dry specimen for microscopic examination in the dry state. The micro-organisms may be subjected to further chemical modifications e.g. methylation, esterification, hydrolysis, oxidation or reaction with sulphur dioxide, whereby to increase the number of active sites available. They may also be treated with an optical brightening agent. Many detailed Examples are provided.
