Abstract
1421955 Insolubilized enzymes ABM INDUSTRIAL PRODUCTS Ltd 13 May 1974 [24 May 1973] 24896/73 Heading C3H [Also in Division C6] Conversion of residual carbohydrates (dextrins) in wort or beer to glucose is achieved using amyloglucosidase in insolubilized form, e.g. in a static or fluidized bed. The insolubilized enzyme may also include glucose oxidase. Insolubilization may be achieved by entraining or encapsulating the enzyme within an insoluble supporting material, by adsorbing it on to a solid surface, or chemically by bonding free amyloglucosidase to silica or glass by means of titanium salts, e.g. the trichloride, or salts of zirconium, vanadium, iron or tin, bonding to filter aid, e.g. kieselguhr by means of a diazo compound, impregnating cellulose with a dialysed enzyme solution, entraining the enzyme within cellulose acetate fibres by extending the enzyme with the cellulose acetate in the fibre production, or, preferably, by bringing the free enzyme and a gelling protein into a particulate form and reacting with a polyfunctional protein reagent. The insolubilized enzyme employed according to the examples consisted of substantially spherical particles or beads prepared by reacting free amyloglucosidase derived, e.g. from Aspergillus awamori and gelatine in particulate form with glutaraldehyde. The particles may be ground to size 60-120 BSS mesh.
