Abstract

1480089 Diluent composition for diagnostic tests E I DU PONT DE NEMOURS & CO 28 Nov 1974 [29 Nov 1973] 51606/74 Heading G1B [ Also in Division C3] A liquid composition suitable for use as a diluent for a biologically active material e.g. in the diagnostic measurement of enzyme activity, comprises 4À5 to 8À0 g/dl of a bovine albumin 95- 145 meq/l of sodium ions, 90-112 meq/l of chloride ions, 1-4 mg/dl of magnesium ions, 7- 12 mg/dl of calcium ions, 3-5À5 meq/l of potassium ions, up to 4 mg/l of zinc ions, 2-6 mg/dl of phosphorus as phosphate ions and up to 4 mg/dl of sulphur as sulphate ions, in de-ionized water. The pH is typically 6À5-7À5, osmolarity 190-300 milliosmoles, viscosity 1À2-2À0 centipoise and ionic strength 0À11-0À14 Á. The composition may be stored in the form of porous spheres of diameter not exceeding 1 mm and pore volume of at least 90%, which spheres are produced by spraying the liquid composition into boiling dichlorodifluoromethane at -25‹C and freeze-drying the frozen droplets. The porous spheres are dissolved in de-ionized water prior to use. Examples are directed to the comparison of acid or alkaline phosphatase and creatine phosphokinase activities when measured in undiluted human serum and human serum diluted with distilled water, saline solution or the liquid composition of the invention.
