Abstract

of formula (I) and their acid addn. salts are new: (where Z = 1-5 C alkyl, 6-14C mono- or poly-cyclic aryl opt. contg. 1-3 same or different halo, (halo)alkyl, (halo)alkoxy, (halo)alkylthio, OH, SH, or nitro substnts., all alkyl 1-4C, or is a 5-7 membered heteroaryl e.g., furyl, thienyl or pyridyl opt. substd. as for aryl; R = H, 1-5 C alkyl, opt. with 1-3 substnts. from NR6R7 or OH; R6 and R7 are H, 1-4C alkyl or NR6R7 is a 5-7 membered ring, opt. contg. another heteroatom, e.g., O, S or N and opt. substd. by one or more 1-5C alkyl (itself opt. substd. by >=1 OH, alkoxy, opt. substd. mono- or poly-cyclic aryl or opt. substd. heterocyclyl) or mono- or poly-cyclic aryl, opt. substd. by halo, (halo)alkyl, (halo)alkoxy, (halo)alkylthio, the amine gps. may be as their acid addn. or quat. ammonium salts; R1,R2 and R3 are H, 1-4C alkyl, 3-9C cycloalkyl or 6-10C aryl all opt. substd., R4 and R5 are both H or together complete a double bond; all alkyl gps. may be branched and/or unsatd.). (I) are useful in human and veterinary medicine as analgesics and psychotropic agents, as anti-anxiety agents they are practically free from sedative and muscle-relaxant side effects. Acute LD50 (p.o., mouse) is e.g., 1400 mg./kg. for 1-(2- 4-(m-chlorophenyl)piperazino ethyl)-3-phenyl-4,5,6,7-tetrah- ydro-1H- 1,2 diazepin-7-one and ED50 in the hot-plate test for analgesia is 42 mg./kg. The daily dose is 30-300 mg. (p.o.), 20-400 mg. (rectally) or 4-40 mg. (by injection).
