Abstract
Παρέχονται τα ολιγονουκλεοτιδικά ανάλογα που ενσωματώνουν 5-αζακυτοσίνη στην ολιγονουκλεοτιδική αλληλουχία, π.χ., με τη μορφή της 5-αζα-2'-δεοξυκυτιδίνης (Δεσιταβίνη) ή 5-αζα-κυτιδίνης. Ειδικότερα, τα ολιγονουκλεοτιδικά ανάλογα που είναι πλούσια σε νησίδια δεσιταβίνης-δεοξυγουανοσίνης (DpG και GpD) παρέχονται για την στόχευση των νησιδίων CpG στο ανθρώπινο γονιδίωμα, ιδιαίτερα στις περιοχές των υποκινητών των γονιδίων που είναι ευάλωτες σε παρεκκλίνουσα υπερμεθυλίωση. Τέτοια ανάλογα μπορούν να χρησιμοποιηθούν για τη φαρμακοτεχνική μορφή της μεθυλίωσης του DNA, όπως την αποτελεσματική αναστολή της μεθυλίωσης της κυτοσίνης στη θέση C-5. Παρέχονται οι μέθοδοι για τη σύνθεση αυτών των ολιγονουκλεοτιδικών αναλόγων και για τη ρύθμιση της μεθυλίωσης των νουκλεϊκών οξέων.
