Abstract
La presente invención proporciona un proceso mejorado para la preparación del ácido (Z)-3-(3-(3,5-bis(trifluorometil)fenil)-1H-1,2,4-triazol-1-il)acrílico (denominado compuesto de la Fórmula estructural <strong>III</strong>), que es un producto intermedio clave útil para la síntesis de selinexor ((Z)-3-(3-(3,5-bis(trifluorometil)fenil)-1H-1,2,4-triazol-1-il)-N'-(pirazin-2-il)acrilohidrazida). El proceso comprende la reacción del compuesto de la Fórmula estructural <strong>I</strong> (tal como se describe en el presente documento) con el compuesto de la Fórmula estructural <strong>II</strong> (tal como se describe en el presente documento) en presencia de un catalizador, una base orgánica y un disolvente que contiene éter. La posterior hidrólisis del compuesto formado de la Fórmula estructural <strong>IIIa</strong> (tal como se describe en el presente documento) se realiza sin el aislamiento del compuesto de la Fórmula estructural <strong>IIIa</strong>, proporcionando el compuesto de la Fórmula estructural <strong>III</strong> con un alto rendimiento y estereoselectividad.
