本发明公开了一种减缓反刍动物胃肠道炎症的营养素功能包的制备及饲喂方法。制备步骤是:称取一定量芦丁,采用DMSO溶解;再称取葡萄籽原花青素、谷氨酰胺和复合菌培养物,按照芦丁∶葡萄籽原花青素;谷氨酰胺∶复合菌培养物=1∶8~10∶19~21∶18~22质量比例混合,搅拌均匀,65℃风干,研磨制成。称取量按150mg.kg‑1BW计算,分装于空心明胶胶囊中,每日于晨饲前一次性灌服,以保证其被全部摄入,本功能包持续饲喂时间不少于30天,每日1次。本功能包是针对反刍动物胃肠道炎症研发的具有多靶点调控功能的营养活性物质组学产品。产品中的4种营养活性组分按不同比例组合后其整体发挥着减轻或修复反刍动物胃肠道损伤,抑制胃肠道促炎因子分泌,减缓炎症持续发生的作用。

背景技术
肠道营养决定肠道健康,肠道健康决定整体健康,可见肠道健康在机体健康中起着至关重要的作用。
胃肠道是营养物质消化、吸收利用的主要器官,也是动物响应应激反应的重要始动器官。反刍动物长期摄入高谷物日粮,会引起胃肠道内环境紊乱,发酵异常,产生大量脂多糖、组织胺和生物胺等异常代谢产物,这些产物在胃肠道内不断积累,并通过胃肠道壁的不断吸收进入血液循环,从而引起局部和全身性炎症反应,疾病易感性增加。近年来针对高谷物日粮诱发反刍动物发生亚急性瘤胃酸中毒(subacute ruminal acidosis,SARA)的大量研究表明:反刍动物诱发SARA,可破坏其胃肠道屏障完整性,促发炎症因子持续释放,导致机体处于炎性反应状态,继发蹄叶炎、子宫炎、腹泻等多种疾病。因长期饲喂高谷物日粮对反刍动物胃肠道健康的影响十分复杂,对机体的影响也涉及多个器官(胃肠道、肝脏、蹄部等)、多个层面(如消化、代谢、免疫、氧化、炎症、应激等),导致机体产生一系列健康问题。因此,如果用单一靶点、单指向的调控剂进行防控和治疗会存在很大难度,且效果也不理想。在“饲料”无抗大背景下,研发安全、高效、绿色、由多种营养活性物质组成具有多作用点的复合营养素功能包,形成一种多靶点、多指向综合作用机制来防控和减缓反刍动物胃肠道炎症的持续发生,提高动物的福利指数具有重要的理论意义、应用价值和光明的前景。

本发明要解决的技术问题是提供一种减轻或调节高谷物日粮条件下反刍动物胃肠道炎症的由多种营养活性物质组成的营养素功能包,这种营养素功能包可有效维护胃肠道健康。

一种减缓反刍动物胃肠道炎症的营养素功能包的制备及饲喂方法是:
1、称取一定量芦丁,采用DMSO溶解;再称取葡萄籽原花青素、谷氨酰胺和复合菌培养物,按照芦丁∶葡萄籽原花青素∶谷氨酰胺∶复合菌培养物=1∶8~10∶19~21∶18~22质量比例混合,搅拌均匀,65℃风干,研磨,制成棕红色粉末状制剂;
2、称取一定量该制剂,称取量按150mg·kg-1BW计算,分装于空心明胶胶囊中,每日于晨饲时口服饲喂给反刍动物,以保证其被全部摄入;
3、反刍动物采食高谷物日粮条件下,本功能包每日饲喂1次,持续饲喂时间至少30天。

选取体况健康,处于泌乳期奶山羊12只,按体重(60±3.36kg)、泌乳量相近原则随机分为饲喂高谷物的对照组(CON,80%谷物,n=6),饲喂高谷物的处理组(HG+CNP,80%谷物,n=6),预饲期15天,日粮精粗比从50∶50逐渐过渡到80∶20,正饲期60天。正饲期开始处理组在开始饲喂高谷物日粮时,同期饲喂本发明的复合营养素功能包,其制备方法操作步骤是:称取一定量芦丁,采用DMSO溶解;再称取葡萄籽原花青素、谷氨酰胺和复合菌培养物,按照芦丁∶葡萄籽原花青素;谷氨酰胺∶复合菌培养物=1∶9∶20∶20质量比例混合,搅拌均匀,65℃风干,研磨至粉末状,称取约9.0g该粉末制剂分装至明胶胶囊。本发明饲喂量按150mg·kg-1BW计算,试验期每周根据体重调整一次饲喂量,每日于晨饲前逐只一次性用水灌服,以保证完全摄入。试验期结束颈静脉采集血液,禁食12后,屠宰奶山羊,采集新鲜的瘤胃、十二指肠和空肠组织采用酶联免疫吸附(ELISA)法测定血清和胃肠道组织TNF-α、IFN-γ、IL-1β、IL-2、IL-4、IL-6和IL-10含量。
试验结果表明:与对照组相比,处理组显著降低了血清淀粉样蛋白A(SAA)和LPS结合蛋白(LBP)水平(p<0.05),降低了TNF-α、IFN-γ、IL-1β和IL-10含量,对血清IL-6含量无影响;同时显著降低了瘤胃、十二指肠和空肠黏膜TNF-α、IL-6和IL-1β含量(p<0.05),增加了IL-2和IL-4含量,显著提高了胃肠道黏膜NF-κB和Nrf2活性(p<0.05),减轻了胃肠道的炎性反应。这些结果表明本发明可以显著降低高谷物日粮条件下奶山羊胃肠道黏膜NF-κB和Nrf2活性,抑制促炎因子TNF-α、IFN-γ、IL-1β和IL-10等的释放,有效抑制胃肠道炎症,对高谷物日粮所致的全身炎症也有缓解的潜力。
实施例2:
在某奶牛场,选取pH值在5.2~5.5之间的泌乳期奶牛20头(BW:620±8.20kg),随机分为两组,一组为SARA组,一组为处理组,每组10头。SARA组与处理组饲养管理均相同,只是处理组每日于晨饲前一次性用水灌服本发明。本发明制备方法操作步骤是:称取一定量芦丁,采用DMSO溶解;再称取葡萄籽原花青素、谷氨酰胺和复合菌培养物,按照芦丁∶葡萄籽原花青素;谷氨酰胺∶复合菌培养物=1∶9∶20∶20质量比例混合,搅拌均匀,65℃风干,研磨至粉末状,称取约93g该粉末制剂分装至明胶胶囊。本发明饲喂量按150mg·kg-1BW计算,连续灌服30天。试验结束颈静脉采集血液用于检测炎症因子(TNF-α、IFN-γ、IL-1β、IL-6)含量、肠道通透性标记物LPS和DAO活性;同时采集新鲜的瘤胃、十二指肠和空肠组织,采用荧光定量PCR方法分析和检测瘤胃、十二指肠和空肠组织炎症因子和NO信号通路关键基因表达量。试验结果表明:(1)与SARA组相比,处理组显著降低了血清TNF-α、IFN-γ、IL-1β和IL-6含量(p<0.05),这表明处理组有效减轻了由SARA引起的机体炎症反应;(2)处理组降低了由SARA所致上升的瘤胃上皮电生理参数Isc、Gt值和分子标记物辣根过物氧化镁(HRP)流速,显著降低了血液LPS和DAO含量(p<0.05),这表明处理组降低了由SARA所致上升的瘤胃和肠道上皮细胞通透性,改善了胃肠道屏障功能;(3)处理组降低了由SARA所致上升的瘤胃黏膜IL-1β、IL-6、iNOS和NO mRNA表达水平(p<0.05),降低了十二指肠和空肠上皮TNF-α、IFN-γ和NO mRNA表达水平,显著增加了瘤胃和十二指肠sIgA表达水平(p<0.05),这表明处理组能抑制SARA所致的瘤胃、十二指肠和空肠上皮促炎因子的表达,缓解炎性反应持续发生,并改善了胃肠道免疫屏障功能。以上结果表明:高谷物日粮引发的SARA可损伤奶牛瘤胃、十二指肠和空肠黏膜屏障,产生炎症反应;本发明能通过抑制NO信号通路来减少促炎因子的释放,有效缓解和调节SARA所致奶牛胃肠道黏膜的炎性反应,减轻SARA对胃肠道的损伤,一定程度上能修复胃肠道黏膜屏障功能。
Claims (2)
1.一种减缓反刍动物胃肠道炎症的营养素功能包,其特征是:原料由芦丁、葡萄籽原花青素、谷氨酰胺和复合菌培养物组成。
2.一种减缓反刍动物胃肠道炎症的营养素功能包的制备及饲喂方法,其特征是:具体制备方法操作步骤是:
①称取一定量芦丁,采用DMSO溶解;再称取葡萄籽原花青素、谷氨酰胺和复合菌培养物,按照芦丁:葡萄籽原花青素;谷氨酰胺∶复合菌培养物=1∶8~10∶19~21∶18~22质量比例混合,搅拌均匀,65℃风干,研磨,制成棕红色粉末状制剂;
②称取该制剂,称取量按150mg·kg-1BW计算,分装于空心明胶胶囊中,每日于晨饲前一次性灌服,以保证其被全部摄入;
③本功能包持续饲喂时间不少于30天,每日1次。
