背景技术

同位素变体

已发表的工作通过在二硫键的任一侧引入空间位阻,建立了修饰二硫键对还原的敏感性的潜力(Kellogg等人(2011),Bioconjugate Chemistry,22,717)。更大程度的空间位阻会降低通过细胞内谷胱甘肽以及细胞外(系统性)还原剂的还原率,从而降低了细胞内和细胞外毒素释放的容易程度。因此,可以通过仔细选择所述二硫键任一侧的阻碍程度,来选择循环中二硫化物稳定性(其将毒素的不期望的副作用最小化)相对于细胞内环境中的有效释放(其将治疗效果最大化)的优化。

合成
本发明的肽可以通过标准技术合成制造,然后与分子支架在体外反应。进行此操作时,可以使用标准化学方法。这使得能够快速大规模地制备可溶性材料,以用于进一步的下游实验或验证。可以使用如Timmerman等人(同上)中公开的常规化学方法来完成这样的方法。

药物组合物
根据本发明的一个进一步的方面,提供了一种药物组合物,其包含如本文所定义的肽配体或药物偶联物,与一种或多种药学上可接受的赋形剂组合。

治疗用途
本发明的双环肽作为CAIX结合剂具有特殊的用途。
各种形式的碳酸酐酶(CA)催化二氧化碳的水合作用,以生成碳酸氢根阴离子(HCO3-)和质子。由CA催化的反应底物调节许多生理过程,包括CO2、质子和碳酸氢根阴离子的形成和运输,如呼吸、pH水平的维持、骨骼发育和其他过程。在人有机体中鉴定了12种具有催化活性的CA同工酶,其细胞定位和其在各种组织中的表达不同。
通过小分子抑制剂对人碳酸酐酶(hCA)活性的临床调节被证明是治疗多种人类疾病的可靠方法,并且几十年来其一直是高血压、青光眼、甲状腺机能亢进和低血糖症的疗法的主要组成部分(Supuran(2008)Nat.Rev.Drug Discov.7,68)。碳酸酐酶的经典抑制剂结合至CA的活性位点,是芳香族或杂芳香族磺酰胺类。
人碳酸酐酶IX(hCAIX)是一种与细胞外膜结合的同种型(其催化结构域位于细胞外空间)。在生理条件下,hCAIX仅在胃肠道的特定组织中表达。在体外和体内癌细胞缺氧期间均显示其过表达。在宫颈、卵巢、肾、食道、肺、乳腺和脑的癌症中检测到hCAIX的表达。在肿瘤中,hCAIX是一种对维持细胞内pH值在正常水平上至关重要的分子,其表达为缺氧的肿瘤细胞提供了在酸性条件下生长的优势(Chiche等人(2009)Cancer Res 69,358)。因此,hCAIX酶是开发用作具有新作用机制的抗癌疗法的特异性抑制剂的便捷靶标(Neri和Supuran(2011)Nature Reviews 10,767)。
根据本发明的方法选择的多肽配体可以用于体内治疗和预防应用、体外和体内诊断应用、体外试验和试剂应用等。具有所选择的特异性水平的配体可以用于涉及在期望交叉反应性的非人动物中进行测试的应用中,或在需要小心控制与同源物或旁系同源物的交叉反应性的诊断应用中。在某些应用如疫苗应用中,可以利用对预定范围的抗原引起免疫反应的能力来调整疫苗适应特定的疾病和病原体。
对哺乳动物的施用优选具有至少90%至95%同质性的基本纯的肽配体,对于药物用途,特别是当所述哺乳动物是人时,最优选98%至99%或更高的同质性。所选择的多肽一旦部分纯化或纯化至所期望的同质性,就可以用于诊断或治疗(包括体外)或用于开发和进行试验步骤、免疫荧光染色等(Lefkovite和Pernis(1979和1981),ImmunologicalMethods,Volumes I and II,Academic Press,NY)。

实施例
材料与方法
肽的合成
肽的合成基于Fmoc化学,使用Peptide Instruments生产的Symphony肽合成仪和MultiSynTech生产的Syro II合成仪。使用标准的Fmoc-氨基酸(Sigma,Merck),其带有适当的侧链保护基团:在每种情况下均使用适用的标准偶联条件,然后使用标准方法进行脱保护。使用HPLC纯化肽,并在分离后将其用1,3,5-三丙烯酰基六氢-1,3,5-三嗪(TATA,Sigma)修饰。为此,将线性肽用50:50MeCN:H2O稀释至约35mL,加入约500μL的100mM TATA的乙腈溶液,然后用5mL的1M NH4HCO3的H2O溶液引发反应。使反应在室温进行约30至60分钟,并且一旦反应完成即冻干(通过MALDI判断)。完成后,在室温将1mL的1M L-半胱氨酸盐酸盐一水合物(Sigma)的H2O溶液加入反应中约60分钟以淬灭任何过量的TATA。
冻干后,如上纯化修饰的肽,同时用Gemini C18柱(Phenomenex)替换Luna C8,并将酸改为0.1%三氟乙酸。合并包含正确的TATA修饰材料的纯级分,冻干并在-20℃进行保存。
除非另有说明,所有氨基酸均以L-构型使用。
生物数据
CAIX竞争结合试验
使用类似于Dubois等人(2011)Radiotherapy and Oncology 99(3),424-43中描述的竞争荧光偏振试验,使用ACAECWIDGWVPCA-Sar6-K(Fl)((SEQ ID NO:18)-Sar6-K(Fl))作为荧光配体,测定本发明的肽对人CAIX的亲和力(Ki)。
CAIX酶抑制试验
