Abstract
本发明公开了一种水稻抗氧化能力相关基因BRHIS1及其应用。所述BRHIS1基因的核苷酸序列如SEQ ID NO:1所示,其编码的蛋白的氨基酸序列如SEQ ID NO:2所示。本发明提供了的水稻调控氧化胁迫基因BRHIS1,是一个新的水稻响应环境胁迫基因,该基因可以调控水稻耐氧化胁迫能力。所述BRHIS1基因对培育耐氧化胁迫植物新品种,特别是培育耐氧化胁迫水稻新品种具有重要意义。为水稻育种及发育机制研究提供了新的基因资源,可以在调控水稻耐氧化胁迫方面得到应用。
Description
技术领域
本发明涉及植物生物技术领域,更具体地,涉及一种水稻抗氧化能力相关基因BRHIS1及其应用。
背景技术
水稻在农业生产过程中会遇到多种不良环境胁迫,影响水稻自身生理环境的变化,最终造成农作物减产。如冷、热、干旱和水淹等胁迫,造成水稻过氧化物酶活性升高,进而造成水稻生长减缓甚至死亡。
水稻耐氧化胁迫是一个数量性状,存在多个调控位点,例如:专利CN101838653A公开了一种水稻分泌型硫氧还蛋白基因,该编码调控水稻体内氧化还原水平相关蛋白,通过抗氧化功能可以降低正常及盐胁迫下水稻体内氧化还原水平,具有延缓水稻衰老的效果;但现在已知的调控水稻耐氧化胁迫性的基因依然还很少。因此,还需要提供更多的与水稻抗氧化能力相关基因,为育种提供更多的选择。
发明内容
本发明的目的在于克服现有技术中存在的上述缺陷和不足,提供一种水稻抗氧化能力相关基因BRHIS1。
本发明的第二个目的在于提供水稻抗氧化能力相关蛋白BRHIS1。
本发明的第三个目的在于提供所述水稻稻抗氧化能力相关基因BRHIS1的应用。
本发明的上述目的是通过以下技术方案给予实现的:
一种水稻抗氧化能力相关基因BRHIS1,所述基因的核苷酸序列如SEQ ID NO:1所示。
一种水稻抗氧化能力相关蛋白BRHIS1,所述蛋白的氨基酸序列如SEQ ID NO:2所示。
本发明还提供含有上述BRHIS1基因的表达载体。
优选地,所述表达载体为含有上述BRHIS1基因的超表达载体。
本发明还提供含有上述表达载体的重组细胞。
本发明还提供一种用于扩增、检测上述水稻抗氧化能力相关基因BRHIS1的引物组,所述引物对包括上游引物和下游引物,其序列分别如SEQ ID NO:3、SEQ ID NO:4所示。
本发明从水稻粳稻品种中花11的cDNA中扩增出的核苷酸序列SEQ ID NO:1并克隆到植物超表达转化载体,通过农杆菌粳稻转化到水稻中花11,所获得的转基因材料具有提高中花11抗氧化胁迫的能力。
因此,本发明请求保护所述BRHIS1基因在提高植物抗氧化能力中的应用。
本发明还提供所述BRHIS1在培育耐氧化胁迫的植物新品种中的应用。
一种提高植物耐氧化胁迫的方法,所述方法包括用上述表达载体转化植物的细胞或组织,并且将转化的植物细胞或组织培育成植株。
当本发明的BRHIS1基因被用于改变植物耐氧化性状时,包括以下步骤:(1)将本发明的水稻耐氧化基因BRHIS1克隆到植物转化载体中;(2)将所构建的植物转化载体转化可再生水稻组织(或器官)并使本发明的水稻耐氧化基因在转化的组织中表达;(3)将被转化的组织(或器官)培养成植株。
优选地,所述转化通过农杆菌介导法或基因枪法进行。
优选地,所述植物为禾本科植物。
进一步优选地,所述禾本科植物为水稻。
与现有技术相比,本发明具有以下有益效果:
本发明提供了的水稻调控氧化胁迫基因BRHIS1,是一个新的水稻响应环境胁迫基因,该基因可以调控水稻耐氧化胁迫能力。所述BRHIS1基因对培育耐氧化胁迫植物新品种,特别是培育耐氧化胁迫水稻新品种具有重要意义。为水稻育种及发育机制研究提供了新的基因资源,可以在调控水稻耐氧化胁迫方面得到应用。

具体实施方式
以下结合说明书附图和具体实施例来进一步说明本发明,但实施例并不对本发明做任何形式的限定。除非特别说明,本发明采用的试剂、方法和设备为本技术领域常规试剂、方法和设备。
除非特别说明,以下实施例所用试剂和材料均为市购。
实施例1 pUbi::BRHIS1超表达载体构建
本发明通过NCBI(http://www.ncbi.nlm.nih.gov/)网站中的BLAST工具进行BRHIS1核酸序列与氨基酸序列的同源性分析BRHIS1基因的结构特征,通过与水稻粳稻品种日本晴(Oryz asativa L sp.Japanic)和水稻籼稻品种中花11(Oryz asativa Lsp.Indica)基因组序列比对,选择编码区起始密码子至终止子共3200bp序列,通过引物对F:5′-ATGGCTGCCTCGGTGGAGGCG-3′(SEQ ID NO:3)和R:5′-TTAGAATGCCGAGCGCCCTTTTG-3′(SEQ ID NO.4)从水稻品种中花11cDNA中扩增出的核苷酸序列SEQ ID NO:1并克隆到植物超表达转化载体中,挑取已合成的含有BRHIS1的目的片段的克隆,于5mL含Kna抗生素的LB液体培养基中,37℃摇床中培养过夜。再提取BRHIS1超表达载体质粒。将提取的质粒用Sanger法测序鉴定。BRHIS1超表达载体中如图1所示。
实施例2 BRHIS1超表达载体的遗传转化
(1)农杆菌感受态细胞的制备
取保存于-80℃的根癌农杆菌EHA105,将其在含有利福平(50μg/mL)的YEB平板上划线,置于28℃培养2天。从YEB平板上挑取农杆菌的单菌落,将其接种于5mL的YEB液体培养基中,置于摇床200rpm,28℃振荡培养过夜。取过夜培养的菌液2mL,接种于50ml的YEB液体培养基中,同样处于200rpm,28℃振荡培养,直到OD600达到0.5。将菌液移至无菌离心管中,冰浴30min,再5000rpm,离心10min,弃上清,收集菌体。再将收集的菌体悬浮于10mL,0.15mol/L预氧化的NaCl中,5000rpm,离心10min,弃去上清。将收集的菌体重悬浮于1mL,20mmol/L预氧化的CaCl2溶液中。将农杆菌悬浮液分装于1.5mL无菌离心管中,每管200uL置于液氮中速冻1min,-80℃冰箱保存以备用。
(2)表达载体转入农杆菌感受态细胞
将农杆菌EHA105感受态细胞置于冰上,取1uL含有目的基因的重组质粒载体加入到其中,充分混匀后,在冰上放置30min。再将其置于液氮中快速氧化冻1min,然后迅速转至水浴锅中,37℃水浴3min,待其融化。向其中加入1mLYEB液体培养基,充分混匀后,放入摇床中,220rpm,28℃恢复培养3h。将恢复培养后的菌液在5000rpm,离心3min,收集菌体,保留200uL菌液。将菌液均匀涂布于含卡那霉素(50μg/mL),利福平(50μg/mL)的YEB固体培养基上,晾干后封口。将平板放在培养箱中,28℃条件下培养2天。待长出农杆菌单菌落后,挑取进行菌落PCR检测。
(3)农杆菌菌落PCR鉴定
挑取农杆菌单菌落,用菌落PCR方法检测是否正确转入根癌农杆菌EHA105中。
实施例3 农杆菌介导的水稻转化
(1)水稻愈伤组织的诱导培养
选取饱满,无霉变的中花11成熟种子,采用糙米机除去种子的颖壳,注意保持成熟胚的完整性。首先用70%乙醇浸泡消毒2min,再浸泡于含有0.1%Tween20的30%次氯酸钠溶液中,置于摇床中,100rpm,振荡消毒30min。于超净工作台中,利用无菌水洗涤消毒后的种子5次。将其转移至铺有无菌滤纸的培养皿中,吸干水分后,再将成熟胚愈伤接种于诱导培养基中,处于持续光照(100μmol·m-2·s-1),32℃条件下培养。
(2)农杆菌的培养
将鉴定为阳性的含有目的基因表达载体的农杆菌,于含有kana和Rif的YEB培养基上划线,28℃培养2天。将收集到的菌体,用适量添加有AS(100μmol/L)的AAM培养基洗涤。洗涤完成后,悬浮于添加有AS(100μmol/L)的AAM培养基中,剧烈摇动,并且调整菌液浓度为OD600值在0.3-0.5之间。
(3)水稻愈伤组织与农杆菌的共培养
挑取诱导7天,且生长状态良好的愈伤组织,放入50mL灭菌的离心管中。将30ml农杆菌浸泡液加入其中,浸泡2min,期间要不断晃动离心管,使菌液与愈伤组织充分接触。再将浸染完成后的愈伤组织置于无菌滤纸上,吸干菌液。同时,事先在共培养基平板上铺上1张无菌滤纸,并用1mL的AAM培养基浸湿,除去上面的气泡。把吸干菌液后的愈伤组织置于铺有滤纸的共培养基上,置于25℃黑暗环境下,培养3天。通常一个表达载体需要100粒左右的愈伤。将共培养3天后的愈伤组织放入50mL的灭菌离心管中。用无菌水洗涤4-5次,直至洗涤液变清澈透明为止。再加入30mL灭菌的羧苄青霉素溶液(500mg/L),置于摇床上,100rpm,振荡洗涤15min。洗涤完成后,倒掉洗涤液,将愈伤组织置于无菌滤纸上,吸干洗涤液。
(4)抗性愈伤组织的筛选
将共培养后,农杆菌洗脱的愈伤组织置于含有羧苄青霉素和潮霉素的筛选培养基上,处于32℃,持续光照下培养,筛选14天,每7天继代一次。筛选2-3个循环,愈伤组织一般在10天左右褐化,在褐化的愈伤组织中重新生长出抗性愈伤组织。
(5)抗性愈伤组织的分化
将含抗性的颜色鲜黄色的愈伤组织转移至不含筛选剂的分化培养基上,置于28℃,持续光照环境下培养,分化14天,每7天继代一次。一般经过15-25天左右,抗性愈伤出现绿芽,进而长出幼芽。直到30-40天后继续分化出小苗。
(6)转基因植株的生根和移栽
将转基因再生植株幼苗移栽于生根培养基中,置于32℃,持续光照条件下,培养7天。待幼苗生根长成后,选取根系发达的小苗,用温水洗净根上的培养基后,在温室内移至土中栽种。
(7)转基因植株的鉴定
提取T0代转BRHIS1水稻株系基因组DNA,使用潮霉素检测引物HPT5:5′-CTGAACTCACCGCGACGTC-3′和HPT3:5′-ATCAGCAATCGCGCATATG-3′得到PCR阳性转基因株系OX-7和OX-13。提取以上材料总RNA,反转录得到cDNA作为模板,用引物BRHIS1 RTF:5′-ATGGTTGAGGAGAGCAAG-3′(SEQ ID NO:5)和BRHIS1 RTR:5′-ATCAAGCCTATGGCCAGC-3′(SEQID NO:6)进行PCR扩增、测序,检测各材料中BRHIS1基因是否在中花11材料中增加表达水平,结果如图2所示,表明成功获得了BRHIS1超表达植株。
实施例4 BRHIS1转基因水稻响应氧化胁迫的表型分析
选取中花11和T2代纯合转基因突变体株系成熟种子播种,每种株系连续播种两行;播种后先于自来水中催芽,幼苗于光照培养室(28℃,12h光照/12h黑暗)中培育20d,待水稻幼苗生长至两叶一心期后移至0-800PPM浓度梯度甲基磺酸甲酯(MethylMethanesulfonate MMS)培养液,28℃室温条件生长5d,进行表型观察及分析。经过0-800PPM浓度梯度的MMS培养液处理后,野生型中花11和转基因株系BRHIS1-2的表型呈现明显的差异(图3),野生型中花11受影响严重,叶片萎焉,大部分株系不能恢复生长;而BRHIS1超表达转基因材料虽有不同程度的叶片干枯变黄,但植株高度高于中花11,表明BRHIS1基因可用于提高水稻抗氧化能力,BRHIS1基因可用于培育耐氧化胁迫的水稻新品种。

1.水稻抗氧化能力相关基因BRHIS1在提高植物抗氧化能力中的应用,其特征在于,所述基因的核苷酸序列如SEQ ID NO.1所示。
2.水稻抗氧化能力相关基因BRHIS1在培育耐氧化胁迫的植物新品种中的应用,其特征在于,所述基因的核苷酸序列如SEQ ID NO.1所示。
3.一种培育耐氧化胁迫的植物新品种的方法,其特征在于,将含SEQ ID NO.1所示基因的表达载体转化植物的细胞或组织中进行超表达,并且将转化的植物细胞或组织培育成植株。
4.根据权利要求1或2所述应用,其特征在于,所述植物为禾本科植物。
5.根据权利要求4所述应用,其特征在于,所述禾本科植物为水稻。
