Description
Technical Field
The invention relates to the technical field of health products, in particular to stichopus japonicus and abalone oligopeptide powder and a preparation method thereof.
Background
At present, the research and industrialization of domestic peptides are mostly in the research stage. The lower the molecular weight the better the product, the stronger the functionality. And are classified into large polypeptides, small molecule peptides, oligopeptides, and short peptides. However, it has been clear over many years that the function of a peptide product depends primarily on the amino acid sequence and structure, i.e., the potency, of the peptide from which it is composed.
The application of marine polysaccharides in cancer and bone treatment in the American and Japanese polysaccharide products is relatively wide at present, but domestic polysaccharide products still stay in a relatively crude processing stage, most of mixed polysaccharides have insufficient precision, and a general technical bottleneck mainly exists in extraction, screening and other links.
In China, a plurality of enterprises and factories are used for processing aquatic products, and in the production of products, the boiled liquid of marine food materials (sea cucumbers, abalones and shellfish) is mostly discarded. Through research, a large amount of active substances such as water-soluble polysaccharide, polypeptide and the like exist in the boiled liquid, and the boiled liquid is discarded to cause great waste, so that the waste is changed into valuable. The quantity is very large, and the quality of the final product is even better than that of the bulk preparation of the raw materials, so that the solution is realized, and the social benefit and the economic benefit are obvious.
Disclosure of Invention
Aiming at the technical defects, the invention aims to provide sea cucumber and abalone oligopeptide powder and a preparation method thereof.
In order to solve the technical problems, the invention adopts the following technical scheme:
the invention provides sea cucumber-abalone oligopeptide powder, which comprises sea cucumber oligopeptide powder, abalone oligopeptide powder, cod collagen peptide powder, oyster oligopeptide powder, sea mussel oligopeptide powder, tussah pupa oligopeptide powder and crude polysaccharide; wherein the total amount of the sea cucumber oligopeptide powder, the abalone oligopeptide powder, the cod collagen peptide powder, the oyster oligopeptide powder, the sea mussel oligopeptide powder and the tussah pupa oligopeptide powder accounts for not less than 90 percent of the total amount of the sea cucumber oligopeptide powder, the abalone oligopeptide powder, the cod collagen peptide powder, the oyster oligopeptide powder, the sea mussel oligopeptide powder, the tussah pupa oligopeptide powder and the crude polysaccharide.
The invention also provides a preparation method of the sea cucumber and abalone oligopeptide powder, which comprises the following steps:
s1, preparing raw materials, wherein the raw materials comprise sea cucumber oligopeptide powder, abalone oligopeptide powder, cod collagen peptide powder, oyster oligopeptide powder, marine mussel oligopeptide powder, tussah pupa oligopeptide powder and crude polysaccharide;
s2, weighing raw materials, and feeding and mixing the raw materials through a three-dimensional mixer for 60 minutes to obtain a mixture;
s3, filtering the mixture by using a 80-mesh sieve to obtain the sea cucumber and abalone oligopeptide powder.
Preferably, the preparation method of the sea cucumber oligopeptide powder comprises the following steps:
collecting sea cucumber cooking liquor, soaking and foaming water, filtering out sea cucumber tissue fragments, silt and other particulate substances in the water by using a cartridge filter, concentrating and desalting filtrate by using an organic membrane with the aperture of 1-2-ten-thousand Da to obtain inorganic salt concentrated solution, separating silt from particulate solids by using a cyclone separator, and subjecting the sea cucumber fragments to enzymolysis;
mixing the sea cucumber tissue fragments with the concentrated solution, adjusting the solid content of the feed liquid to 10-20%, adjusting the temperature of the feed liquid to 40-55 ℃, adjusting the pH of the feed liquid to 2.5-9.0, and adding special compound protease, wherein the proportion of the compound protease is 2709 alkaline protease, 20 ten thousand mu/g, and 10-40%; 30% -60% of As1398 neutral protease with the concentration of 10 ten thousand mu/g; 10-40% of trypsin, 1.0-10% of flavor enzyme and 1.0-5.0% of total protein, stirring for enzymolysis for 3-6h, adjusting the pH of the enzymolysis solution to 5.0-7.0, heating to 70-75 ℃ for enzyme deactivation, removing granular impurities and lipids by using a tubular centrifuge, cooling the filtrate to 10-30 ℃ through a heat exchanger, separating through an organic membrane with the molecular weight cutoff of 5000-20000 Da, and taking the permeate as oligopeptide;
heating the permeate to 50-80 ℃, adding beta-cyclodextrin, wherein the total protein amount of the beta-cyclodextrin is 1-10%, stirring for 20-60 min, passing the fishy smell-removed liquid through a granular activated carbon column for further fishy smell removal, smell removal and decoloration, passing the column-passed liquid through a 50-100 nm ceramic membrane or cotton cake filter for decarburizing to obtain a fine filtrate, further concentrating the fine filtrate through an organic membrane with the pore diameter of 50Da-100Da until the solid content is 17-18%, then carrying out reduced pressure evaporation concentration until the solid content is more than or equal to 40%, and carrying out spray drying to obtain the sea cucumber oligopeptide powder.

The invention has the beneficial effects that: the sea cucumber protein contains more than 18 kinds of amino acids, and the composition ratio of the amino acids is better. The sea cucumber contains 0.4mg/100g of vanadium, and the physiological function of the vanadium is to transmit iron in a body to the liver, so that the iron can be fully absorbed to form hemoglobin. Contains chondroitin sulfate, Stichopus japonicus mucopolysaccharide, etc. Chondroitin sulfate can delay human body aging, and Stichopus japonicus mucopolysaccharide has broad-spectrum anticancer effect, and can prevent acute radiation injury and promote hemopoiesis recovery.
The small molecular oligopeptide is absorbed more quickly and easily by organisms than single amino acid, and has more important biological activity in the aspect of human metabolism. Therefore, the sea cucumber oligopeptide after enzymolysis has higher biological value and more important physiological functions. The sea cucumber peptide is degraded by enzyme, has no antigenicity, and does not cause any anaphylactic reaction after being eaten, thus having high safety. Improving immunity and resisting fatigue; preventing tumor and cancer; regulating blood pressure and blood lipid, and has adjuvant treatment effect on diabetes; delaying aging, helping sleep, and delaying the appearance of senile plaque; tonifying kidney and strengthening body; radioprotective enhances the primary function of phagocytic activity of leukocytes.
