Abstract
The invention relates to a traditional Chinese medicine composition for treating children's chronic cough and application thereof, wherein the traditional Chinese medicine composition is prepared from the following raw material medicines in parts by weight: 6 parts of asarum, 5 parts of euphorbia kansui, 3 parts of fried white mustard seed, 3 parts of ephedra, 3 parts of myrobalan, 3 parts of astragalus, 3 parts of bighead atractylodes rhizome, 3 parts of salvia miltiorrhiza, 3 parts of each of radix adenophorae, 2 parts of earthworm, 2 parts of cortex dictamni and 3 parts of cinnamon. The invention also comprises application of the traditional Chinese medicine composition in preparing a patch for treating infantile chronic cough. Its advantages are: in the formula, asarum and euphorbia kansui are used as monarch drugs, semen sinapis alba and ephedra are used as ministerial drugs, myrobalan, astragalus mongholicus, bighead atractylodes rhizome, red-rooted salvia root, radix adenophorae, earthworm and cortex dictamni are used as adjuvant drugs, cinnamon is used as conductant drug, the drugs act synergistically, the traditional Chinese medicine composition is prepared into an acupoint application, and the odor of the traditional Chinese medicine composition is utilized to directly reach channels through skin acupoints and is absorbed in the body, and the traditional Chinese medicine composition is dissolved in body fluid to achieve double effects of medicine effect and acupoint.
Description
Technical Field
The invention relates to the technical field of traditional Chinese medicines, in particular to a traditional Chinese medicine composition for treating children's chronic cough and application thereof.
Background
Cough and asthma is a common disease and a plurality of diseases of children clinically, the modern medicine considers that the exact pathogenesis is not completely clear, the pathogenesis is the result of the comprehensive action of a plurality of factors, and the low immune function is considered to be one of the important causes of the disease.
The clinical manifestations are mostly:

The oral administration, western medicine and injection of Chinese medicine are not easy to be accepted by children, but the effect is not ideal enough for the lack of targeted treatment of individual constitutions. The medicament can be used for preventing and treating recurrent respiratory tract infection by a susceptible formula, and avoids toxic and side effects caused by long-term administration of western medicines. Therefore, a traditional Chinese medicine preparation which has small toxic and side effect and quick curative effect and is suitable for the mouth feel of children and used for preventing and treating recurrent respiratory tract infection is urgently needed.
Chinese patent application CN201610943919.3 discloses a traditional Chinese medicine for treating children's cough caused by exogenous pathogens and a preparation method thereof, which comprises 10 to 15 weight parts of fried white mustard seed, 10 to 12 weight parts of blackberrykiky rhizome, 8 to 10 weight parts of corydalis tuber, 6 to 8 weight parts of asarum and 6 to 8 weight parts of liquorice, and overcomes many defects of western medicines. However, the traditional Chinese medicine composition for treating infantile chronic cough and the application thereof are not reported at present.
Disclosure of Invention
The invention aims to provide a traditional Chinese medicine composition for treating children's chronic cough and application thereof, aiming at the defects of the prior art.
In order to achieve the purpose, the invention adopts the technical scheme that:
firstly, the invention provides a traditional Chinese medicine composition for treating infantile chronic cough, which comprises the following raw material medicines in parts by weight: 4-8 parts of asarum, 3-7 parts of euphorbia kansui, 1-5 parts of fried white mustard seed, 1-5 parts of ephedra, 1-5 parts of myrobalan, 1-5 parts of astragalus, 1-5 parts of bighead atractylodes rhizome, 1-5 parts of salvia miltiorrhiza, 1-5 parts of adenophora tetraphylla, 1-5 parts of radix glehniae, 1-3 parts of earthworm, 1-3 parts of cortex dictamni and 1-5 parts of cinnamon.

Further, the traditional Chinese medicine composition is prepared into a clinically acceptable pharmaceutical preparation according to a conventional traditional Chinese medicine preparation method.

Secondly, the invention provides the application of the traditional Chinese medicine composition in preparing a medicine for treating the infantile chronic cough.
Further, the invention provides application of the traditional Chinese medicine composition in preparing a medicine for treating qi-yin deficiency type children chronic cough.
Finally, the invention provides a medicine for treating children's chronic cough, which is prepared from the traditional Chinese medicine composition, and the preparation method of the oral preparation comprises the following steps:
(1) taking the raw materials according to the weight part ratio, adding water, and decocting over strong fire for 0.5-1.5 hours;
(2) decocting with slow fire for 20-40 min;
(3) and (4) leaching out juice, precipitating, taking clear liquid, and bottling for later use.
Further, the invention provides an acupoint application for treating infantile chronic cough, which is prepared from the traditional Chinese medicine composition and ginger juice, and the preparation method of the acupoint application comprises the following steps:

The preparation method comprises the following steps: taking the raw materials according to the weight part ratio of any one of the embodiments 1-5 respectively, adding water, decocting with strong fire for 0.5-1.5 hours, decocting with slow fire for 20-40 minutes, draining juice, filtering, concentrating the filtrate, and cooling to room temperature; concentrating the supernatant, adding water, stirring, standing, and concentrating the supernatant into fluid extract. Example 10 pharmacological toxicity test
1. Acute toxicity test
Acute toxicity experiments using mice show that: the traditional Chinese medicine preparation capsule of the invention is orally taken by mice, under the dosage of 332.5g crude drugs/kg, the mice have slight activity reduction after administration, the mice recover to normal after about 1 hour, no animal dies after continuous observation for 7 days after administration, the general condition, diet, water intake, urination and weight increase are all normal, and the experimental result shows that: the maximum dosage of the traditional Chinese medicine preparation capsule of the invention for oral gavage of mice is 332.5g crude drug/kg/d (LD50 is more than 332.5g crude drug/kg). The total daily clinical medication amount of the traditional Chinese medicine preparation is 0.1g crude drugs/kg/d; according to the weight, the tolerance of the traditional Chinese medicine preparation for intragastric administration of the mice is 3325 times of that of clinical patients. The medicine has low acute toxicity and safe clinical medication.
2. Long term acute toxicity test
SD rats are selected, pills of the traditional Chinese medicine preparation of the invention with different concentrations (18.0, 6.0 and 2.0g crude drug/kg) are administered, the stomach is drenched once a day for 90 days continuously, 1/2 animals (half of a male and a female) are killed in each group 24 hours after the last administration, and the rest 1/2 animals are killed after 2 weeks of continuous observation. The animals were observed for changes in appearance, general behavior, food intake, and body weight during the test period, and examined for indices of hematology (RBC, HB, reticulocyte, PLT, CT, WBC, and classification) and blood biochemistry (AST, ALT, ALP, Glu, BUN, Crea, TP, T.BIL, ALB, CHOL), urine biochemistry, organ coefficients, histopathology, etc. 90 days after administration and 2 weeks after discontinuation. The test result shows that: the traditional Chinese medicine preparation has good general state in high, medium and low dose animals, and has no abnormal change in appearance physical signs, behavior activity, food intake and weight growth; the three dose groups and the control group are in normal range of hematology examination, blood biochemistry examination and urine biochemistry examination, and have no obvious difference among the groups; no obvious abnormality is found in histopathological examination of main organs of each group. The above index was not changed after 2 weeks of withdrawal. The test dosage is 180, 60 and 20 times of the clinical dosage, and according to the test result, the continuous administration of the traditional Chinese medicine preparation for 90 days at high, medium and low dosages (18.0, 6.0 and 2.0g crude drug/kg) has no obvious influence on rats, has no clear toxic target organs and sensitive indexes, has no delayed toxic reaction in the observation of the recovery period, and prompts that the clinical application dosage of the traditional Chinese medicine preparation has higher safety.
3.
