Detailed Description
The following detailed description of specific embodiments of the present invention is provided in conjunction with examples, but it should be understood that the scope of the present invention is not limited to the specific embodiments.
Indoor bioassay 1: indoor insecticidal toxicity determination method of acetamiprid and bisultap on spodoptera frugiperda
Test subjects: the Spodoptera frugiperda collected in the field is bred and propagated indoors by using corn leaves, and F1 generation egg masses and hatched 2-instar larvae bred by using corn leaves are used as experimental insect sources.
The test method comprises the following steps: the insecticidal activity of the medicament is determined by adopting a leaf soaking method. Preparing a series of drug solutions to be tested with concentration, cutting corn leaves into 5cm leaves, soaking the leaves in the drug solution for 10s, naturally drying the leaves, and placing the leaves in a circular plastic box. And selecting 2-instar larvae, performing starvation treatment for 4 hours, and inoculating the treated leaves. Clear water was used as control. Larvae were treated 20 at a time, 3 replicates. And checking the survival state of the larvae 48h after treatment, judging the larvae to die by no reaction when the brush lightly touches the body surface of the larvae, recording the death number and survival number, and calculating and correcting the death rate.
The treatment-corrected mortality was converted to a probability value, the concentration was converted to a logarithmic value, and the virulence equation and the inhibitory median concentration LC50 were calculated by the least squares method. Calculating the virulence index and the cotoxicity coefficient (CTC) of the medicament according to the Sun Yunpei method.
When the CTC is less than or equal to 80, the composition shows antagonism, when the CTC is more than 80 and less than 120, the composition shows additive action, and when the CTC is more than or equal to 120, the composition shows synergistic action.
Measured virulence index (ATI) ═ standard medicament LC 50/test medicament LC50 × 100
Theoretical virulence index (TTI) ═ A agent virulence index x percent of A in the mixture + B agent virulence index x percent of B in the mixture
Co-toxicity coefficient (CTC) × 100 [ measured toxicity index (ATI) of the mixture)/Theoretical Toxicity Index (TTI) of the mixture ].
The results of the virulence determination are shown in table 1 below.
TABLE 1 acetamiprid and dimehypo indoor insecticidal virulence determination for Spodoptera frugiperda
As shown in Table 1, the weight ratio of the acetamiprid to the dimehypo in the insecticidal composition is in the range of 1: 30-30: 1, the composition can be used for preventing and controlling Spodoptera frugiperda, and has excellent insecticidal activity. The preferred weight ratio of the acetamiprid to the dimehypo is 1: 1 and 1: 15.
Indoor bioassay 2: indoor ovicidal toxicity determination of acetamiprid and dimehypo against spodoptera frugiperda
The test method comprises the following steps: the ovicidal activity of the agent was determined by egg immersion. Selecting light green egg masses which are originally laid in the current day, dispersing the egg masses into single egg grains by using a brush, and uniformly adhering the egg grains on a transparent adhesive tape to prepare egg cards; soaking the egg card in the liquid medicine for 10s, sucking the excessive liquid medicine by using filter paper, drying the liquid medicine, and then putting the liquid medicine into a 50mL centrifuge tube. Clear water was used as control. Each 20 eggs were treated, 3 replicates. After the treatment 1d, the number of hatched and unhatched eggs was checked using a split dissecting microscope, and the hatching inhibition rate was calculated and corrected. The co-toxicity coefficient calculation method is the same as above.
TABLE 2 indoor ovicidal toxicity assay for acetamiprid and dimehypo against Spodoptera frugiperda
As shown in Table 2, the weight ratio of the acetamiprid to the dimehypo in the insecticidal composition is in the range of 1: 30-30: 1, the composition can be used for preventing and controlling Spodoptera frugiperda, and the weight ratio of the acetamiprid to the dimehypo is preferably in the range of 30: 1-1: 15.
It is found from tables 1 and 2 that when the weight ratio of the acetamiprid to the dimehypo is 1: 1 and 1: 15, the composition has excellent insecticidal and ovicidal effects on spodoptera frugiperda, and shows remarkable synergistic effects.
Determination of field drug Effect 1
Test subjects: spodoptera frugiperda (Spodoptera frugiperda) in corn field
The test method comprises the following steps: test set 2 medicament treatment, using clear water as a control, repeating the treatment for 3 times, setting a protection row with the area of each cell being 25m2, and randomly arranging and combining the cells. The medicine is applied in a stem and leaf spraying mode and is uniformly sprayed. And (3) respectively surveying the population density of each cell by adopting a five-point sampling method 1d before pesticide application and 7d after pesticide application, surveying 1m2 per point, recording the population number and damage condition of each point, and respectively calculating the population decline rate and the control effect.
