背景技术

天然果胶(Citrus Pectin简称CP)是从水果果皮、果肉中提取的一种多糖复合物,其分子是以一种长链碳水化合物结构存在,分子量为5-30万Da,酯化度为20-75%。根据纯度形状等可分为食品级、医药级果胶,高脂、低脂果胶,高凝、中凝、低凝果胶。天然果胶是纯天然无毒副作用的水溶性膳食纤维,作为食品添加剂使用时,有很好的胶凝、稳定、乳化、增稠、悬浮功能。因此,将果胶用于半固化胃肠内营养物具有良好的应用前景。目前已经有多个关于果胶调节胃肠内营养物粘度的专利,市场上也出现了的相应的产品。但临床和动物实验表明,这类调节剂主要通过改变胃肠内营养物的物理性状来预防食物反流。然而受众人群往往伴有胃肠道消化能力弱,肠道菌群失调等问题,仅仅改变营养物粘度无法解决这些问题,还有可能带来消化不良、便秘等副作用。因此开发一种具有适度调节胃肠内营养物粘度、促进胃肠道蠕动、改善消化系统内环境功能的产品十分有必要。

其中,优选地,所述猕猴桃提取物的制备方法,包括下述步骤:
将猕猴桃洗净去皮后置于研钵中研磨破碎,添加2倍量的蒸馏水,85℃加热4h,过滤,滤液于3000r/min离心10min,取上清,4℃静置1d后于3000r/min离心10min,留上清得猕猴桃水提物。

下述实施例中猕猴桃提取物的制备方法,包括下述步骤:
将猕猴桃洗净去皮后置于研钵中研磨破碎,添加2倍量的蒸馏水,85℃加热4h,过滤,滤液于3000r/min离心10min,取上清,4℃静置1d后于3000r/min离心10min,留上清得猕猴桃水提物。
实施例1
本实施例提供一种胃肠内营养消化吸收调节剂,是由低脂柑橘果胶、卡拉胶、低聚异麦芽糖、猕猴桃提取物和水组成的水溶液,所述低脂柑橘果胶的质量浓度为50g/L,所述卡拉胶的质量浓度为10g/L,所述低聚异麦芽糖的质量浓度为20g/L,所述猕猴桃提取物的质量浓度为5g/L。
其中,所述低脂柑橘果胶的粘均分子量为10000,酯化度为5%。
本实施例胃肠内营养消化吸收调节剂的制备方法,包括下述步骤:
(1)取低脂柑橘果胶50g和卡拉胶10g,加入到85℃的600ml水中,水浴保持90min,然后800rpm搅拌2h,即得到均匀混合溶液;
(2)取猕猴桃提取物5g和低聚异麦芽糖20g加入混合溶液中,溶液补水至1L,搅拌均匀,即得。
本实施例的胃肠内营养消化吸收调节剂用NDJ-8S型粘度计(上海方瑞仪器有限公司),在25℃的液体温度测定样品的粘度,测得粘度值5mPa·s。
实施例2
本实施例提供一种胃肠内营养消化吸收调节剂,是由低脂柑橘果胶、卡拉胶、低聚果糖、猕猴桃提取物和水组成的水溶液,所述低脂柑橘果胶的质量浓度为40g/L,所述卡拉胶的质量浓度为15g/L,所述低聚果糖的质量浓度为10g/L,所述猕猴桃提取物的质量浓度为8g/L。
其中,所述低脂柑橘果胶的粘均分子量为50000,酯化度为20%。
本实施例胃肠内营养消化吸收调节剂的制备方法,包括下述步骤:
(1)取低脂柑橘果胶40g和卡拉胶15g,加入到85℃的600ml水中,水浴保持90min,然后800rpm搅拌2h,即得到均匀混合溶液;
(2)取猕猴桃提取物8g和低聚果糖10g加入混合溶液中,溶液补水至1L,搅拌均匀,即得。
本实施例的胃肠内营养消化吸收调节剂用NDJ-8S型粘度计(上海方瑞仪器有限公司),在25℃的液体温度测定样品的粘度,测得粘度值6mPa·s。
实施例3
本实施例提供一种胃肠内营养消化吸收调节剂,是由低脂柑橘果胶、卡拉胶、低聚半乳糖、猕猴桃提取物和水组成的水溶液,所述低脂柑橘果胶的质量浓度为120g/L,所述卡拉胶的质量浓度为5g/L,所述低聚半乳糖的质量浓度为30g/L,所述猕猴桃提取物的质量浓度为3g/L。
其中,所述低脂柑橘果胶的粘均分子量为80000,酯化度为25%。
本实施例胃肠内营养消化吸收调节剂的制备方法,包括下述步骤:
(1)取低脂柑橘果胶120g和卡拉胶5g,加入到85℃的600ml水中,水浴保持90min,然后800rpm搅拌2h,即得到均匀混合溶液;
(2)取猕猴桃提取物3g和低聚半乳糖30g加入混合溶液中,溶液补水至1L,搅拌均匀,即得。
本实施例的胃肠内营养消化吸收调节剂用NDJ-8S型粘度计(上海方瑞仪器有限公司),在25℃的液体温度测定样品的粘度,测得粘度值7mPa·s。
实施例4
本实施例提供一种胃肠内营养消化吸收调节剂,是由低脂柑橘果胶、卡拉胶、甲壳低聚糖、猕猴桃提取物和水组成的水溶液,所述低脂柑橘果胶的质量浓度为45g/L,所述卡拉胶的质量浓度为12g/L,所述甲壳低聚糖的质量浓度为15g/L,所述猕猴桃提取物的质量浓度为6g/L。
其中,所述低脂柑橘果胶的粘均分子量为100000,酯化度为30%。
本实施例胃肠内营养消化吸收调节剂的制备方法,包括下述步骤:
(1)取低脂柑橘果胶45g和卡拉胶12g,加入到85℃的600ml水中,水浴保持90min,然后800rpm搅拌2h,即得到均匀混合溶液;
(2)取猕猴桃提取物6g和甲壳低聚糖15g加入混合溶液中,溶液补水至1L,搅拌均匀,即得。
本实施例的胃肠内营养消化吸收调节剂用NDJ-8S型粘度计(上海方瑞仪器有限公司),在25℃的液体温度测定样品的粘度,测得粘度值4mPa·s。
实施例5
本实施例提供一种胃肠内营养消化吸收调节剂,是由低脂柑橘果胶、卡拉胶、低聚木糖、大豆低聚糖、猕猴桃提取物和水组成的水溶液,所述低脂柑橘果胶的质量浓度为55g/L,所述卡拉胶的质量浓度为8g/L,所述大豆低聚糖的质量浓度为15g/L,所述低聚木糖的质量浓度为10g/L,所述猕猴桃提取物的质量浓度为5g/L。
其中,所述低脂柑橘果胶的粘均分子量为150000,酯化度为40%。
本实施例胃肠内营养消化吸收调节剂的制备方法,包括下述步骤:
(1)取低脂柑橘果胶55g和卡拉胶8g,加入到85℃的600ml水中,水浴保持90min,然后800rpm搅拌2h,即得到均匀混合溶液;
(2)取猕猴桃提取物5g、低聚糖25g加入混合溶液中,溶液补水至1L,搅拌均匀,即得。
本实施例的胃肠内营养消化吸收调节剂用NDJ-8S型粘度计(上海方瑞仪器有限公司),在25℃的液体温度测定样品的粘度,测得粘度值6mPa.s。
性能试验
一、组合物样品粘度稳定性
测试本发明的胃肠内营养消化吸收调节剂在放置1个月和3个月、6个月、12个月粘度稳定性(表1)。
用NDJ-8S型粘度计(上海方瑞仪器有限公司),在25℃的液体温度测定样品的粘度,测得粘度值。

由上述数据可以看出,本发明的胃肠内营养消化吸收调节剂放置12个月的过程中,无明显沉淀产生,粘度无明显变化。
二、体外模拟实验
通过调节溶液pH在体外模拟胃消化食物时的内环境(pH3.5),测试本发明的粘度调节剂与肠内营养乳剂(华瑞肠内营养乳剂-瑞能)在模拟胃环境下的增稠效果。
用NDJ-8S型粘度计(上海方瑞仪器有限公司),在37℃的液体温度测定样品的粘度,测得粘度值(表2)。

由上述数据可以看出,本发明的胃肠内营养消化吸收调节剂与肠内营养乳剂在体外模拟胃环境下混合后能迅速提升粘度值,且十分稳定。
三、离体肠断动力学实验
选用BL-420F生物机能实验仪与JH-2型肌张力传感器连接,观察和记录(37±0.5℃)麦氏浴槽中离体肠段的运动。以台氏液(37±0.5℃)为基础对照、肾上腺素(0.01%)为阴性对照、乙酰胆碱(0.01%)为阳性对照,分别观测实施例和对照例对小鼠离体肠段运动的影响(见表3)。

由上述数据可以看出,本发明的胃肠内营养消化吸收调节剂能有效提高肠段收缩频率,促进肠道蠕动。
四、动物实验
选健康昆明种小鼠70只,雌雄各半,随机分成空白对照组、实验对照组和5个实验组。各组小鼠禁食不禁水16h后,按表4方案饲喂。30min后立即脱颈椎处死小鼠,打开腹腔分离肠系膜,剪取上端自幽门、下端至回盲部的肠管,将小肠平铺于白色瓷盘中,用生理盐水洗去表面血迹,轻轻将小肠拉成直线,测量肠管长度为“小肠总长度”,从幽门至酚红前沿为“肠道蠕动长度”(表5)。

肠道推进实验结果表明,本发明的胃肠内营养消化吸收调节剂能有效延长胃肠内营养剂在消化系统中的停留时间;在促进胃肠道蠕动方面本发明明显优于单纯的果胶溶液。

Claims (4)
1.一种胃肠内营养消化吸收调节剂,其特征在于是由低脂柑橘果胶、卡拉胶、低聚糖、猕猴桃提取物和水组成的水溶液,所述低脂柑橘果胶的质量浓度为40-120g/L,所述卡拉胶的质量浓度为5-15g/L,所述低聚糖的质量浓度为10-30g/L,所述猕猴桃提取物的质量浓度为3-8g/L。
2.根据权利要求1所述的一种胃肠内营养调节剂,其特征在于:所述低脂柑橘果胶的粘均分子量为10000-150000,酯化度为5%-40%。
3.根据权利要求1所述的一种胃肠内营养调节剂,其特征在于:所述低聚糖为低聚异麦芽糖、低聚果糖、低聚木糖、低聚半乳糖、大豆低聚糖、甲壳低聚糖中的一种或几种。
4.根据权利要求1所述的一种胃肠内营养调节剂,其特征在于:所述猕猴桃提取物的制备方法,包括下述步骤:
将猕猴桃洗净去皮后置于研钵中研磨破碎,添加2倍量的蒸馏水,85℃加热4h,过滤,滤液于3000r/min离心10min,取上清液,4℃静置1d后于3000r/min离心10min,留上清液得猕猴桃水提物。
