Abstract
The invention discloses a sample liquid diluting and uniformly mixing method suitable for a chemiluminescence immunoassay analyzer, which comprises the steps of firstly vertically moving a sampling needle downwards to the upper part of a reaction cup needing to be diluted and uniformly mixed, wherein the longitudinal central line of the sampling needle is offset from the longitudinal central line of the reaction cup; moving the sampling needle downwards until the liquid suction port is positioned below the liquid level of the sample liquid; extracting a sample liquid; simultaneously, the sampling needle synchronously descends along with the liquid level, and liquid pumping is stopped when the sampling needle descends to a set position; injecting liquid into the reaction cup by the sampling needle, and synchronously lifting the sampling needle to the liquid level position of the sample liquid along with the lifting of the liquid level to stop; and repeatedly extracting the sample liquid until the sample liquid in the reaction cup meets the specified uniformity requirement. The invention realizes the efficient dilution and uniform mixing of a small-volume sample solution. Meanwhile, the control logic is simple, the working efficiency is high, and the method is suitable for the in vitro diagnosis field of the combination of a biochemical analyzer, a biochemical analyzer and a chemiluminescence analyzer and the like.

Technical Field
The invention relates to a chemiluminescence immunoassay analyzer, in particular to a sample liquid diluting and uniformly mixing method suitable for the chemiluminescence immunoassay analyzer.
Background
The chemiluminescence immunoassay method has the advantages of high sensitivity, good selectivity, high analysis speed and wide linear range, so that the chemiluminescence immunoassay analyzer taking the chemiluminescence immunoassay method as a carrier is widely applied to clinical examination.
When the full-automatic chemiluminescence immunoassay analyzer works, a sampling needle needs to precisely sample liquid. Injecting the high-concentration sample liquid into a reaction cup, diluting and uniformly mixing in the reaction cup so as to facilitate the test analysis of the instrument. Because the volume of the sample liquid is small, the sample liquid needs to be uniformly diluted to dozens or hundreds of times in a short time, and the requirement on the dilution and uniform mixing efficiency is high. If the uniform mixing effect is poor, sample liquid layering or local density unevenness is easily caused, so that the test result of the instrument has deviation or the repeatability of sample detection is poor.
At present, methods for diluting and uniformly mixing sample liquid mainly include methods of suction-injection uniform mixing, vibration uniform mixing, rotation uniform mixing and the like. The vibration mixing is to dilute and mix the sample by using mechanical vibration of a sample adding needle or a reaction cup, and the mixing mode has high noise and poor mixing effect and is not suitable for diluting and mixing the sample with small volume. The rotating and mixing process utilizes a mechanical structure to drive the reaction cup to rotate, so that the effect of sample dilution and mixing is achieved. The existing suction-injection mixing method utilizes a sampling needle to repeatedly suck and inject sample liquid and diluent in a reaction cup to achieve the effect of diluting and mixing the sample, but the existing suction-injection mixing method has the defects of low mixing efficiency and easy vertical layering of the diluted sample liquid in the reaction cup.

step 4, injecting liquid into the reaction cup by the sampling needle, and synchronously lifting the sampling needle to the position of the step 2 along with the lifting of the liquid level to stop;
and 5, repeating the step 3 and the step 4 until the sample liquid in the reaction cup meets the specified uniformity requirement.
In the step 1, the horizontal distance L of the longitudinal central line of the sampling needle offset from the longitudinal central line of the reaction cup meets the condition that L is more than or equal to 0.25D and less than 0.5D; wherein D is the inner diameter of the sampling needle.
In the step 3, the speed of the sampling needle for extracting the sample liquid is 0.6-1.8 m/s; in the step 4, the liquid injection speed of the sampling needle into the reaction cup is 0.6-1.8 m/s.
The invention has the advantage of realizing efficient dilution and uniform mixing of small-volume sample liquid. Meanwhile, the control logic is simple, the working efficiency is high, and the method is suitable for the in vitro diagnosis field of the combination of a biochemical analyzer, a biochemical analyzer and a chemiluminescence analyzer and the like.

step 3, extracting sample liquid; the sampling needle 1 synchronously descends along with the liquid level while extracting the sample liquid, and the liquid extraction is stopped when the sampling needle 1 descends to a set position; the speed of the sampling needle for pumping 1 sample liquid is 0.6 m/s-1.8 m/s;
step 4, as shown in fig. 2, injecting liquid into the reaction cup 2 by the sampling needle 1 at a speed of 0.6-1.8 m/s to promote the sample liquid in the reaction cup 2 to form a vortex 6; the sampling needle 1 synchronously rises to the position of the step 2 along with the rise of the liquid level and stops;
and 5, repeating the step 3 and the step 4 until the sample liquid in the reaction cup 2 meets the specified uniformity requirement.
The invention abandons the conventional standing, whipping and standing self-mixing (long mixing time and poor effect) adopted for small-volume sample liquid, controls the depth of the sampling needle below the liquid level and the force for drawing and injecting the sample liquid by offsetting the longitudinal central line of the sampling needle to the longitudinal central line of the reaction cup, and repeatedly draws and injects the sample liquid through the pump body to dilute and mix the sample. Because the sample needle is offset from the reaction cup, the sampling needle can form vortex in the reaction cup by reasonably controlling the pumping and injecting speed and the liquid amount of the pump in the process of pumping and injecting the sample liquid, and the risk of layering and uneven local density of the diluted sample liquid is avoided.
