Abstract
The invention relates to a traditional Chinese medicine composition for detoxifying and relieving itching, which is prepared from 1-8 parts by weight of sweet wormwood herb, 1-8 parts by weight of purslane, 1-8 parts by weight of biota orientalis, 1-8 parts by weight of climbing groundsel herb and 1-6 parts by weight of peppermint. The traditional Chinese medicine composition can directly act on the skin lesion part, is convenient to use and high in safety, and plays an important role in treating inflammatory exudative skin diseases such as eczema and the like.
Description
Technical Field
The invention relates to the field of medicines, in particular to a traditional Chinese medicine composition for detoxifying and relieving itching.
Background
Eczema is a chronic, recurrent, itchy skin disease, the incidence of which increases year by year worldwide, severely affecting human health and quality of life. The clinical characteristics of eczema include severe itching, erythema, pimple or herpes, edema, blister exudation, repeated disease duration, easy recurrence and chronic trend in the acute stage. 80% of eczema patients are accompanied by itching, severe itching can excite scratching, and skin infection is easy to accompany, thereby generating vicious circle and causing difficulties for local and systemic treatment. Specific causes and mechanisms of eczema remain hot spots of research in the medical community to date. In recent years, with the development of society, the incidence of this disease has been increasing year by year in a fast-paced and high-pressure life state. Therefore, prevention and effective treatment of eczema have become one of the problems to be studied and solved in the medical community.
At present, modern medicine mainly treats eczema by antihistamine medicines, antiallergic medicines, glucocorticoids and the like. Antihistaminic and antiallergic drugs often only achieve the purpose of relieving symptoms, and glucocorticoid can rapidly control symptoms in a short term, but can cause various adverse reactions such as skin atrophy, hormone-dependent dermatitis and the like after long-term use, and can cause skin damage deterioration after drug withdrawal, thus being easy to generate drug dependence.
The eczema is thought to be caused by wind evil, damp evil and heat evil accumulated in the skin striae and muscle surface, and damp heat accumulated in the skin is damaging yin blood, so the ancient books of the traditional Chinese medicine are called 'wet sore', 'immersion sore', 'embroidery ball wind' and the like. The external treatment method of the traditional Chinese medicine has the obvious characteristics and advantages of integral syndrome differentiation, treatment of both symptoms and root causes, small adverse reaction, wide applicable crowd and the like, can directly apply the medicine to the skin damage part, can reduce the use of systemic medicine, has high safety, and has important roles in the treatment of inflammatory exudative skin diseases such as eczema and the like.
At present, the report of the traditional Chinese medicine formula for effectively treating acute and chronic eczema is less, so that the traditional Chinese medicine formula for treating acute and chronic eczema is further developed from the field of traditional Chinese medicines, and the medicine with low side effect is a technical problem to be solved urgently.
Disclosure of Invention
In order to solve the technical problems, the invention comprises the following aspects:
the first aspect of the invention provides a traditional Chinese medicine composition for detoxifying and relieving itching, which is prepared from 1-8 parts by weight of sweet wormwood, 1-8 parts by weight of purslane, 1-8 parts by weight of biota orientalis, 1-8 parts by weight of senecio scandens and 1-6 parts by weight of peppermint.
Preferably, the traditional Chinese medicine composition is prepared from 3-6 parts by weight of sweet wormwood herb, 3-6 parts by weight of purslane, 3-6 parts by weight of biota orientalis, 3-6 parts by weight of senecio scandens and 2-5 parts by weight of mint.
More preferably, the traditional Chinese medicine composition is prepared from 6 parts by weight of sweet wormwood herb, 6 parts by weight of purslane, 6 parts by weight of biota orientalis, 6 parts by weight of climbing groundsel herb and 5 parts by weight of peppermint.
Preferably, the Chinese medicinal composition further comprises pharmaceutically acceptable auxiliary materials.
Preferably, the Chinese medicinal composition is an external preparation.
Preferably, the traditional Chinese medicine composition is in the form of patch, ointment, cream or gel.
More preferably, the traditional Chinese medicine composition is in a gel form.
Preferably, the preparation method of the traditional Chinese medicine composition comprises the following steps:
weighing herba Artemisiae Annuae, herba Portulacae, folium Platycladi, herba Senecionis Scandentis and herba Menthae, adding deionized water 4-6 times of the total weight of the Chinese medicinal materials, heating and reflux extracting for 1-2 hr, filtering the extractive solution, concentrating the filtrate under reduced pressure to give medicinal liquid 1-2 times of the total weight of the Chinese medicinal materials, optionally adding pharmaceutically acceptable adjuvants, and making into Chinese medicinal composition.
Preferably, deionized water with the total weight of 5 times of the total weight of the traditional Chinese medicinal materials is added into the preparation method.
Preferably, the heating reflux extraction time in the preparation method is 1 hour.
Preferably, the filtrate is concentrated under reduced pressure until the weight of the liquid medicine is 1 time of the total weight of the added Chinese medicinal materials.
Preferably, the gel matrix with the prescription amount is weighed, distilled water with the prescription amount is added, the mixture is stirred and then is stood for overnight, the humectant is added after the mixture is completely swelled, the concentrated traditional Chinese medicine liquid is added after the mixture is fully and uniformly stirred, and the preparation is obtained.

Preferably, the gel matrix is selected from one or more of carbomer, gelatin, chitosan or sodium alginate.

Preferably, the weight ratio of the humectant to distilled water is 1 (1-3). More preferably, the weight ratio of humectant to distilled water is 1 (1-2). Further preferably, the weight ratio of humectant to distilled water is 1:1.
Preferably, the weight percentage of the traditional Chinese medicine extract in the gel is 10-20%. More preferably, the weight percentage of the traditional Chinese medicine extract in the gel is 12-18%. Further preferably, the weight percentage of the traditional Chinese medicine extract in the gel is 16%.
Preferably, the gel comprises the above traditional Chinese medicine extract, carbomer 941, propylene glycol and distilled water. More preferably, the gel comprises 15-25g of the above Chinese medicine extract, 0.1-0.5g of carbomer 941, 30-80g of propylene glycol and 30-80g of distilled water. Further preferably, the gel comprises 20g of the above-mentioned Chinese medicine extract, 0.3g of carbomer 941, 50g of propylene glycol and 50g of distilled water.
The second aspect of the present invention provides a preparation method of the above-mentioned Chinese medicinal composition, comprising the following steps:
weighing herba Artemisiae Annuae, herba Portulacae, folium Platycladi, herba Senecionis Scandentis and herba Menthae, adding deionized water 4-6 times of the total weight of the Chinese medicinal materials, heating and reflux extracting for 1-2 hr, filtering the extractive solution, concentrating the filtrate under reduced pressure to give medicinal liquid 1-2 times of the total weight of the Chinese medicinal materials, optionally adding pharmaceutically acceptable adjuvants, and making into Chinese medicinal composition.
Preferably, deionized water with the total weight of 5 times of the total weight of the traditional Chinese medicinal materials is added into the preparation method.
Preferably, the heating reflux extraction time in the preparation method is 1 hour.
Preferably, the filtrate is concentrated under reduced pressure until the weight of the liquid medicine is 1 time of the total weight of the added Chinese medicinal materials.
Preferably, the gel matrix with the prescription amount is weighed, distilled water with the prescription amount is added, the mixture is stirred and then is stood for overnight, the humectant is added after the mixture is completely swelled, the concentrated traditional Chinese medicine liquid is added after the mixture is fully and uniformly stirred, and the preparation is obtained.
The third aspect of the invention provides application of the traditional Chinese medicine composition in preparation of medicines for treating eczema.
Preferably, the eczema is acute eczema or chronic eczema.
The fourth aspect of the invention provides an application of the traditional Chinese medicine composition in preparing medicines for reducing IL-4 content and/or improving INF-gamma content in serum.
Preferably, the invention provides application of the traditional Chinese medicine composition in preparation of medicines for improving IFN-gamma/IL-4 ratio in serum.
The traditional Chinese medicine raw materials selected by the invention have the following common pharmacological effects in the field:
sweet wormwood herb: cold in nature, bitter and pungent in flavor, enters liver and gallbladder meridians. Herba Artemisiae Annuae has effects of clearing heat and removing summer-heat, removing steaming, and preventing malaria, and can be used for treating summer-heat, yin deficiency, night fever, bone steaming, malaria, jaundice due to damp-heat.
Purslane: the traditional Chinese medicine composition has the effects of clearing heat and detoxicating, cooling blood and stopping bleeding, and is clinically used for treating enteritis, bacillary dysentery, furuncle swelling and toxicity, snake and insect bite, hemorrhoids gall, eczema, erysipelas, snake and insect bite, hematochezia, hemorrhoids, metrorrhagia and metrostaxis, acute dermatitis, subacute dermatitis, herpes zoster, postpartum and functional sub-functional bleeding, appendicitis and ancylostomiasis.
Platycladus orientalis leaves, with cold nature, bitter and astringent taste, enter lung meridian, liver meridian and spleen meridian. The cacumen Platycladi has effects of cooling blood, stopping bleeding, promoting hair growth, and blackening hair, and can be used for treating hematemesis, epistaxis, hemoptysis, hematochezia, metrorrhagia, hemorrhage, and blood heat alopecia.
All-grass of groundsel: cold in nature and bitter in flavor, enter liver meridian and lung meridian. The herba Senecionis Scandentis has effects of clearing heat and detoxicating, cooling blood and detumescence, removing liver fire for improving eyesight, killing parasites and relieving pain, and can be used for treating wind-heat type common cold, influenza, upper respiratory infection, acute tonsillitis, sore throat, pneumonia, lobar pneumonia, etc.
Peppermint: pungent in flavor, cool in nature, enters lung and liver meridians, dispels wind-heat, clears head and eyes, relieves sore throat, promotes eruption, soothes liver and moves qi; can be used for treating wind-heat type common cold, initial wind-warm syndrome, headache, conjunctival congestion, pharyngitis, aphtha, rubella, measles, chest distress and hypochondrium distention.
The clinical application method of the traditional Chinese medicine composition comprises the following steps:
150ml of freshly prepared traditional Chinese medicine extract in the embodiment of the invention is taken, 8 layers of gauze are soaked, the traditional Chinese medicine liquid is flatly applied to the affected part for 15 minutes each time of wet application, the residual liquid is wiped off by purified water after the wet application, the treatment is carried out for 2 times a day, and 4 weeks is a treatment course.
The invention has the technical effects that:
1. the traditional Chinese medicine composition disclosed by the invention is reasonable in compatibility, and the inventor optimizes the traditional Chinese medicine composition with obvious eczema treatment effect, especially chronic eczema, by optimizing the components and content ratio of the traditional Chinese medicine.
2. The traditional Chinese medicine composition can directly act on the skin injury part, the potency of the medicine directly reaches the affected part, the use is convenient, the safety is high, and the traditional Chinese medicine composition plays an important role in treating inflammatory exudative skin diseases such as eczema and the like.
Detailed Description
The present invention will be further described with reference to examples, but the embodiments of the present invention are not limited thereto. The experimental methods used in the following examples are conventional methods unless otherwise specified.
Example 1
Weighing 36g of sweet wormwood herb, 36g of purslane, 36g of cacumen biotae, 36g of senecio scandens and 30g of mint, adding 900mL of deionized water, heating, refluxing and extracting for 1 hour, filtering the extracting solution, and concentrating the filtrate under reduced pressure to 200mL to obtain the traditional Chinese medicine extract 1. Weighing 0.3g of carbomer 941, adding 50g of distilled water, stirring, standing overnight, adding 50g of propylene glycol after complete swelling, fully and uniformly stirring, adding 20g of the prepared traditional Chinese medicine extract, and uniformly stirring to obtain the traditional Chinese medicine gel preparation 1.
Example 2
18g of sweet wormwood herb, 54g of purslane, 36g of cacumen biotae, 36g of senecio scandens and 30g of peppermint are weighed, 900mL of deionized water is added, heating reflux extraction is carried out for 1 hour, the extracting solution is filtered, and the filtrate is concentrated to 200mL under reduced pressure, so that the traditional Chinese medicine extract 2 is obtained. Weighing 0.3g of carbomer 941, adding 50g of distilled water, stirring, standing overnight, adding 50g of propylene glycol after complete swelling, fully and uniformly stirring, adding 20g of the prepared traditional Chinese medicine extract, and uniformly stirring to obtain the traditional Chinese medicine gel preparation 2.
Example 3
Weighing 36g of sweet wormwood, 18g of purslane, 54g of cacumen biotae, 36g of senecio scandens and 30g of mint, adding 900mL of deionized water, heating, refluxing and extracting for 1 hour, filtering the extracting solution, and concentrating the filtrate under reduced pressure to 200mL to obtain the traditional Chinese medicine extract 3. Weighing 0.3g of carbomer 941, adding 50g of distilled water, stirring, standing overnight, adding 50g of propylene glycol after complete swelling, fully and uniformly stirring, adding 20g of the prepared traditional Chinese medicine extract, and uniformly stirring to obtain the traditional Chinese medicine gel preparation 3.
Example 4
Weighing 36g of sweet wormwood herb, 36g of purslane, 18g of cacumen biotae, 54g of senecio scandens and 30g of mint, adding 900mL of deionized water, heating, refluxing and extracting for 1 hour, filtering the extracting solution, and concentrating the filtrate under reduced pressure to 200mL to obtain the traditional Chinese medicine extract 4. Weighing 0.3g of carbomer 941, adding 50g of distilled water, stirring, standing overnight, adding 50g of propylene glycol after complete swelling, fully and uniformly stirring, adding 20g of the prepared traditional Chinese medicine extract, and uniformly stirring to obtain the traditional Chinese medicine gel preparation 4.
Comparative example 1 (Replacing of Qianli light into honeysuckle)
Weighing 36g of sweet wormwood herb, 36g of purslane, 36g of cacumen biotae, 36g of honeysuckle flower and 30g of peppermint, adding 900mL of deionized water, heating, refluxing and extracting for 1 hour, filtering the extracting solution, and concentrating the filtrate under reduced pressure to 200mL to obtain the traditional Chinese medicine extract 1 of the comparative example. Weighing 0.3g of carbomer 941, adding 50g of distilled water, stirring, standing overnight, adding 50g of propylene glycol after complete swelling, fully and uniformly stirring, adding 20g of the prepared traditional Chinese medicine extract, and uniformly stirring to obtain the traditional Chinese medicine gel preparation comparative example 1.
Comparative example 2 (substitution of cacumen Platycladi with Sanguisorbae)
Weighing 36g of sweet wormwood herb, 36g of purslane, 36g of garden burnet, 36g of senecio scandens and 30g of peppermint, adding 900mL of deionized water, heating, refluxing and extracting for 1 hour, filtering the extracting solution, and concentrating the filtrate under reduced pressure to 200mL to obtain the traditional Chinese medicine extract 2 of the comparative example. Weighing 0.3g of carbomer 941, adding 50g of distilled water, stirring, standing overnight, adding 50g of propylene glycol after complete swelling, fully and uniformly stirring, adding 20g of the prepared traditional Chinese medicine extract, and uniformly stirring to obtain the traditional Chinese medicine gel preparation comparative example 2.
Experimental example 1 Effect of the Chinese medicinal composition of the invention on the serum IL-4 and IFN-gamma content of chronic eczema model rats
1. Test method
80 rats were randomly divided into a blank group, a model group, the inventive examples 1-4 and the comparative examples 1-2, 10 each. 1 day before molding, except for blank groups, all test groups were dehaired with a shaver in the abdomen of the rat in an area of 2cm×2cm, and then sensitized with 50 μl of 5% dncb acetone solution in the dehaired area for 2 consecutive days, dehaired with an area of 3cm×3cm in the back of the rat for 5 days, and then excited with 100 μl of 0.1% dncb acetone solution in the dehaired area in the back of the rat for 6 days, and then excited with 1 time every 3 days for 4 consecutive times. Blank rats were subjected to depilation treatment on the abdomen and back, and distilled water was applied to the depilation area. On day 2 after the end of the excitation, the traditional Chinese medicine gel prepared in the examples 1-4 and the comparative examples 1-2 of the invention are smeared on the dehairing area on the back of the rat according to the dosage of 2g gel/kg body weight except for a blank group and a model group, and are administrated 1 time a day for 10 days continuously. After 12 hours from the last administration, the femoral artery of each rat in each test group was bled, centrifuged at 3000r/min for 15min, the supernatant was taken, and the content of IL-4 and IFN-gamma in the serum of each rat group was determined by ELISA.
2. Test results
The IL-4 and IFN-gamma content in serum of chronic eczema model rats after administration of the traditional Chinese medicine of the present invention is shown in Table 1 below.

As can be seen from Table 1 above, compared with the blank group, the serum INF-gamma content of the rats in the model group is obviously reduced, the IL-4 content is obviously increased, the IFN-gamma/IL-4 ratio is reduced (the difference has statistical significance), and the unbalance of Th1/Th2 occurs. Compared with the model group and the comparative examples 1-2, the serum INF-gamma content of the rats in the examples 1-4 of the invention is obviously increased, the IL-4 content is obviously reduced, and the IFN-gamma/IL-4 ratio is increased (the difference has statistical significance), so that the balance of Th1/Th2 is promoted. Surprisingly, the traditional Chinese medicine preparation of the embodiment 1 of the invention has the highest range of IFN-gamma/IL-4 ratio in serum, and has an unexpected excellent effect. The test results show that the traditional Chinese medicine composition has the effects of treating eczema and reducing inflammatory response, and the action mechanism of treating chronic eczema is possibly related to regulating Th1 and Th2 cytokines.
Test example 2 clinical treatment cases of the Chinese medicinal composition of the present invention
Clinical curative effect observation shows that the detoxification antipruritic traditional Chinese medicine composition has obvious curative effect on skin diseases such as eczema dermatitis, contact dermatitis and the like, can effectively relieve symptoms such as redness, liquid seepage, itching and the like, and promotes skin injury healing. The following will illustrate some representative examples. If not specified, the wet dressing method of the traditional Chinese medicine in the case of the invention is specifically referred to in the section of clinical application method of the traditional Chinese medicine composition.
Case 1: li Mou female 30 years old, with a diagnosis of general erythema accompanied by itching for 5 years, was treated irregularly and was diagnosed as "eczema". The traditional Chinese medicine composition of the embodiment 1 of the application is applied to the affected part after being taken orally (10 mg/time, 1 time/day) and treated by the external mometasone furoate ointment (1 time/day), the skin injury is light and heavy, the skin injury is not healed repeatedly, and the affected part is wet-applied for 2 times a day for 3 weeks, so that the red spots and itching of the whole body are basically relieved.
Case 2: chen Mou it was diagnosed as "sweat herpes" in the case of men, 45 years old, who had repeated 5 months of diagnosis for herpes, blisters, liquid seepage and itching of both hands and feet. External halomipine ointment (2 times/day), oral cetirizine tablet (10 mg/time, 1 time/day) and the like, and the skin injury is repeated, and the traditional Chinese medicine composition of the embodiment 1 of the application is applied to the affected part after the treatment, and is wet-applied for 2 times per day for 4 weeks, so that the herpes and pruritus of hands and feet basically subside, and slight desquamation is left.
Case 3: a person who is 8 years old, who is a woman who is 8 years old, is diagnosed as "toxic dermatitis and contact dermatitis" because of local redness, liquid seepage and itching occurring 2 days after the right upper limb is contacted with unknown chemicals. The topical mometasone furoate soft extract has poor effect, and the oral cetirizine (10 mg/time, 1 time/day) is taken after the doctor makes a visit, and meanwhile, the traditional Chinese medicine composition of the embodiment 1 of the application is applied to the affected part in a wet way, 4 times a day for 5 days, and the red swelling, the liquid seepage and the itching of the right upper limb are basically resolved.
Case 4: cui Mou, women, 65 years old, were diagnosed as "contact dermatitis" by a 3-day visit of a local erythema and papule blister accompanied with itching after the external application of the wound ointment for right leg pain. External halomicin soft extract (2 times/day), oral desloratadine tablet (10 mg/time, 1 time/day), etc., the improvement of skin lesions is not obvious, the traditional Chinese medicine composition of the application example 1 is applied to the affected part after the treatment, the wet application is carried out for 2 times a day for 3 days, and red rash, pimple and blister itch of the right lower leg are basically resolved, and the color is remained and sinking.
