Abstract
The invention discloses a composition for resisting skin aging. It comprises cholesterol, 1,2 pentanediol and palmitoyl tripeptide-5. When the usage amount of the palmitoyl tripeptide-5 is the same, the anti-aging effect of the palmitoyl tripeptide-5 can be improved. Less palmitoyl tripeptide-5 may be added to the product under certain effect expectations. The cost of the 1,2 pentanediol and the cholesterol is far less than that of palmitoyl tripeptide-5, so the invention can save the cost of products on the premise of ensuring the efficacy.
Description
Technical Field
The invention belongs to the field of cosmetics, and particularly relates to a composition for resisting skin aging.
Background
Skin aging refers to the aging and sexual damage of skin function, and the protective ability, regulation ability and the like of the skin on the body are reduced, so that the skin cannot adapt to the change of internal and external environments, and the overall appearance conditions such as color, luster, shape, texture and the like are changed. Aging of skin is classified into endogenous aging and exogenous aging. Endogenous aging refers to the natural aging of the skin with age. The skin is whitened, fine wrinkles, elasticity is reduced, skin is relaxed and the like are generated. The most important cause of extrinsic aging is photoaging due to sun exposure. Is manifested by wrinkles, skin laxity, rough, yellowish or sallowness skin discoloration, telangiectasia, pigmentation, etc.
Slowing down skin aging is a need for all women and is a hot spot for research by numerous skin care companies and high tech raw materials companies. The anti-aging technology mainly used at present comprises better maintenance (antioxidation, moisturizing, anti-free radical, improving skin barrier function and the like) for skin, repairing subcutaneous injury, improving cell activity, stimulating subcutaneous collagen production, relaxing subcutaneous muscle memory tension and the like. Meanwhile, technologies for preventing photoaging such as sun protection, blue light prevention and the like are widely applied to the field of skin care products for aging resistance.
Palmitoyl tripeptide-5 is an effective raw material for anti-wrinkle cosmetics, and achieves the aims of smoothing wrinkles and tightening skin and anti-wrinkle by stimulating skin collagen synthesis through tissue growth factor (TGF-beta). Palmitoyl tripeptide-5 promotes skin cell growth, inhibits oxygen and hydroxyl radicals, promotes synthesis of matrix proteins (matrixproteins), especially collagen, and may also increase the production of elastin, hyaluronic acid, glycosaminoglycans, and fibronectin. The collagen peptide promotes collagen synthesis by increasing stromal cell activity, making the skin appear more elastic and youthful. The action mode is similar to tretinoin, but HAs no side effect of tretinoin, and increases synthesis of collagen and Hyaluronic Acid (HA) of cell stroma, so that fine lines are not seen, and the skin is firm.
At present, palmitoyl tripeptide-5 is widely applied to the field of skin care products, but the raw materials are expensive, and the cost of the product is high.
The invention comprises the following steps:
the invention aims to provide a composition using three skin anti-aging components, which can obviously promote skin penetration of palmitoyl tripeptide-5, thereby reducing the usage amount of palmitoyl tripeptide-5 or obtaining better anti-aging effect under the same usage amount.
The invention adopts the following technical scheme:
a composition for skin anti-aging comprising three components of cholesterol, 1,2 pentanediol, and palmitoyl tripeptide-5.
The palmitoyl tripeptide-5 is 0.0001-0.01% by mass, the 1,2 pentanediol is 1.0-10.0% by mass, the cholesterol is 0.01-0.1% by mass, and the solvent is water.
The mass percentage of the palmitoyl tripeptide-5 is 0.001-0.005%.
The mass percentage of the 1,2 pentanediol is 3.0-6.0%.
The mass percentage of the cholesterol is 0.03-0.05%.
The palmitoyl tripeptide-5 comprises 0.01% of palmitoyl tripeptide-5, 10.0% of 1,2 pentanediol, 0.1% of cholesterol and water as a solvent.
The palmitoyl tripeptide-5 comprises 0.0025% of palmitoyl tripeptide-5, 4% of 1,2 pentanediol and 0.04% of cholesterol by mass, and the solvent is water.
The formulation of the composition is a variety of formulations including, but not limited to, solutions, emulsions, creams, gels, solids, lyophilized powders, and liposomes.
Palmitoyl Tripeptide-5 (palmitoyl-lysyl-lysine) is a synthetic signal peptide, also known as palmitoyltribide-3, pal-KVK and SYN-COLL, which mimics the thrombospondin and results in the binding of the sequence Arg-PheLys to the inactive transforming growth factor-beta form, inducing the release of active transforming growth factor-beta (TGF-beta). Briefly, palmitoyl tripeptide-5 mimics the mechanism of the human body itself and accelerates collagen synthesis in the skin.
Animal studies have shown that palmitoyl tripeptide-5 may increase collagen synthesis. In vitro studies have shown that palmitoyl tripeptide-5 can prevent collagen degradation by interfering with MMP1 and MMP3 collagen degradation. In addition, in one control trial, 60 healthy volunteers received 2.5% palmitoyl tripeptide-5 versus 10% palmitoyl pentapeptide-3 cream versus placebo cream twice daily for 84 days. Palmitoyl tripeptide 5 significantly reduced the mean and maximum remission parameters by 12%.
Palmitoyl tripeptide-5 may be used as an important ingredient in anti-aging skin care products not only because it may address damage to the extracellular matrix and inhibit skin inflammation, but also because it may compact and moisturize the skin.
One of the most effective ways to select anti-aging products is to know how the ingredients contained therein affect your skin. The composition is a proprietary artificial composition intended to communicate with healthy skin cells for healthy operation. In skin care and anti-aging, cellular communication is a relatively new concept. Skin care has been around the addition of nutritional oils and ingredients to the skin for centuries. Recent studies have shown that components such as Palmitoyl Tripeptide-5 can make skin healthier and younger by improving collagen production and by supporting skin cell functions to tighten the skin.
It is also believed that palmitoyl tripeptide-5 may interact with the hyaluronic acid and elastin production system. The combined effect of these features is to increase water content, improve the appearance of fine lines and wrinkles, and thicken the skin. In addition, studies have shown that it helps to support the healthy function of cells, improving toxin removal.
Cholesterol is a derivative of cyclopentanol phenanthrene. Is white or light yellow crystal, is a main steroid compound in mammals, and plays an important role in basic cell life activities. Cholesterol has been found from cholelithiasis as early as 18 th century, and this lipid-like substance was named cholesterol by the chemist of Daucan 1816. Cholesterol is widely found in animals, especially brain and nerve tissue, and is most abundant in kidneys, spleen, skin, liver and bile. It has similar solubility to fat, is insoluble in water, and is easily dissolved in diethyl ether, chloroform, etc. Cholesterol is an essential substance indispensable to animal tissue cells, and it is involved not only in the formation of cell membranes, but also as a raw material for the synthesis of bile acids, vitamin D and steroid hormones. Cholesterol is also metabolized to bile acid, steroid hormone, 7-dehydrocholesterol, and 7-dehydrocholesterol is converted to vitamin D3 by uv irradiation, so cholesterol is not a substance harmful to the human body.
Cholesterol is an important constituent of cell membrane, which surrounds every cell of human body, and is an essential constituent of cholesterol, accounting for more than 20% of plasma membrane lipids. It has been found that animals are fed cholesterol-deficient foods, and as a result, the red blood cells of these animals are increased in fragility, which tends to cause cell rupture. Research shows that cholesterol can prevent disorder of the bilayer when the temperature is high; the low temperature can interfere the ordering of the liquid crystal, prevent the formation of the liquid crystal and keep the fluidity of the liquid crystal. Thus, it is conceivable that without cholesterol, cells cannot maintain normal physiological functions and life will also terminate.
1.2 pentanediol is an organic compound, colorless transparent liquid, and has a relative density of 0.971, and is dissolved in organic solvents such as alcohol, ether, and ethyl acetate. 1, 2-pentanediol is a key raw material for synthesizing propiconazole as a bactericide. The 1, 2-pentanediol has broad-spectrum antibacterial activity, can also play the roles of moisturizing and conditioning skin, is often added into skin care products, is suitable for nursing delicate skin such as sensitive skin, infants and the like and aging dry skin, and can also be used for hair care. The 1, 2-pentanediol product used for personal care products has high purity, is colorless and odorless, and does not contain irritating impurities.
The 1, 2-pentanediol has good inhibition effect on gram positive bacteria, gram negative bacteria and mould, can be added into propiconazole serving as a bactericide, can be smeared on skin after being prepared into cream, has a certain antibacterial effect, and is beneficial to inhibiting pathogenic bacteria from breeding;
the 1, 2-pentanediol is a humectant with good effect, can achieve the moisturizing effect when being added into cosmetics, can play a role in nourishing skin, can rapidly supplement water required by the skin, improve the dry state of the skin, relieve the conditions of skin peeling and chip falling, and can also improve the water resistance in sun-proof products;
the 1, 2-pentanediol can be added into skin care products, such as water, cream, essence and the like, and also can be added into eye cream, and the skin care products can nourish skin, enable the skin to become more tender and elastic, have the effect of conditioning the skin, improve the dry skin and the large pores, but can achieve the effect only by adhering to the coating.
In summary, among the three components of the composition of the present invention, cholesterol, 1,2 pentanediol and palmitoyl tripeptide-5 play an anti-aging role; cholesterol and 1,2 pentanediol can improve the penetration of palmitoyl tripeptide-5, and can make more palmitoyl tripeptide-5 molecules enter the dermis; the cholesterol and the 1, 2-pentanediol can improve the water environment of the skin surface and the moisture content in the horny layer, thereby prolonging the permeation duration of the palmitoyl tripeptide-5. The three components have synergistic effect, so that the utilization efficiency of the palmitoyl tripeptide-5 can be effectively improved, and the anti-aging effect of the palmitoyl tripeptide-5 is enhanced.
Compared with the prior art, the invention has the beneficial effects that:
1. when the usage amount of the palmitoyl tripeptide-5 is the same, the anti-aging effect of the palmitoyl tripeptide-5 can be improved. Less palmitoyl tripeptide-5 may be added to the product under certain effect expectations.

Example 1
Palmitoyl tripeptide-5,1,2 pentanediol, cholesterol, and deionized water were added in the following proportions, with 0.1wt% of methyl paraben as a preservative. Stirring until all the materials are dissolved for standby.

The test result analysis method comprises the following steps:
0.1 g of anti-wrinkle toner for test is uniformly smeared in a square area with the inner side edge of an arm of one subject being 2 cm long. After waiting for 4 hours, the skin tissue was sequentially taped in the center of a 2 cm square area. The skin tissue sample obtained in the tenth time was subjected to liquid chromatography to obtain the concentration data of palmitoyl tripeptide-5 in this skin tissue as the amount of palmitoyl tripeptide-5 reaching the dermis layer. The synergistic penetration of 1,2 pentanediol and cholesterol is clearly seen in the ratio obtained by dividing the amount of palmitoyl tripeptide-5 obtained in comparative example 1 by the amount of palmitoyl tripeptide-5 obtained in other examples and comparative examples.
Test results: as can be seen from FIG. 1, the amount of palmitoyl tripeptide-5 used in example 1 was only twenty-fifth of that in comparative example 1 after 4 hours of use of the examples and comparative example, but a similar osmotic effect to that in example 1 was obtained. In example 3, the same amount of palmitoyl tripeptide-5 as in comparative example 1 was used to obtain a 15-fold permeation effect. With reference to the permeation results of comparative examples 2 and 3 using only 1,2 pentanediol or cholesterol, a certain permeation-promoting effect can be obtained by using only 1,2 pentanediol or cholesterol, but not by using them simultaneously. Thus, the simultaneous use of 1,2 pentanediol and cholesterol can play a significant role in synergetic permeation promotion.

Claims (8)
1. A composition for skin anti-aging comprising cholesterol, 1,2 pentanediol, and palmitoyl tripeptide-5.
2. The composition for skin aging resistance according to claim 1, wherein the palmitoyl tripeptide-5 is 0.0001-0.01% by mass, the 1,2 pentanediol is 1.0-10.0% by mass, the cholesterol is 0.01-0.1% by mass, and the solvent is water.
3. The composition for skin aging according to claim 2, wherein the amount of palmitoyl tripeptide-5 is 0.001 to 0.005% by mass.
4. The composition for skin aging resistance according to claim 2, wherein the mass percentage of 1,2 pentanediol is 3.0-6.0%.
5. The composition for skin aging according to claim 2, wherein the cholesterol content is 0.03 to 0.05% by mass.
6. The composition for skin aging resistance according to claim 1, wherein the palmitoyl tripeptide-5 is 0.01% by mass, the 1,2 pentanediol is 10.0% by mass, the cholesterol is 0.1% by mass, and the solvent is water.
7. The composition for skin aging resistance according to claim 1, wherein the palmitoyl tripeptide-5 is 0.0025% by mass, the 1,2 pentanediol is 4% by mass, the cholesterol is 0.04% by mass, and the solvent is water.
8. The composition for skin anti-aging according to claim 2, wherein the formulation of the composition is a variety of formulations including, but not limited to, solutions, emulsions, creams, gels, solids, lyophilized powders, and liposomes.
