The administration of Omega-3 enhances learning, memory and concentration. Since Omega-3 unsaturated fatty acids are not self-synthesized in the body, Omega-3 unsaturated fatty acids can be ingested only from food, whereas Omega-3 unsaturated fatty acids contained in daily foods in small amounts do not satisfy the needs of the human body, and thus it is necessary to improve the health condition of the human body by supplementing such essential unsaturated fatty acids through external ingestion.
At present, commercially available products rich in Omega-3 unsaturated fatty acids mainly appear in the form of capsules, such as DHA soft capsules, fish oil soft capsules, krill oil capsules and the like, however, the storage period of the existing products is not long, the oil loading amount in the capsules is not large, the oil rich in Omega-3 unsaturated fatty acids is poor in solubility, is not easy to absorb by a human body, and has low bioavailability, so that the product performance is influenced.
Therefore, there is a need to develop an unsaturated fatty acid product with long shelf life and high bioavailability.
Disclosure of Invention
Aiming at the defects of the prior art, the invention aims to provide the fructus cannabis unsaturated fatty acid-containing drop which has the advantages of reasonable ratio of all nutrient components, high in vivo bioavailability, stable quality and long storage period, and has various effects of assisting in enhancing memory, preventing senile dementia, reducing blood fat and the like; the invention also aims to provide a preparation method of the drop, which has the advantages of simple operation and suitability for large-scale production.
In one aspect, the invention provides a drop containing fructus cannabis unsaturated fatty acid, which comprises the following components in percentage by mass:
DHA algal oil: 5 to 25 percent;
fructus cannabis unsaturated fatty acids: 5 to 65 percent;
soybean lecithin: 3 to 15 percent;
flavoring agent: 0.1 to 0.8 percent;
hemp seed oil: and (4) the balance.
In a preferred embodiment, the drops containing edestin unsaturated fatty acids according to the invention comprise the following components in mass percent:
DHA algal oil: 10 to 20 percent;
fructus cannabis unsaturated fatty acids: 10 to 60 percent;
soybean lecithin: 5 to 10 percent;
flavoring agent: 0.3 to 0.5 percent;
hemp seed oil: and (4) the balance.
The DHA algae oil is a general name of all oil substances in algae, is a slightly fishy faint yellow liquid at normal temperature, and has the main effective components of EPA, DHA and the like. DHA is an indispensable material basis for brain cell development and exercise. The level of DHA in vivo directly affects the proliferation of brain cells, nerve conduction, and the growth and development of synapses. Human memory and mental function are maintained and enhanced by DHA. The DHA supplement can promote brain cell full development, delay intelligence decline and amnesia, and prevent Alzheimer disease.
The fructus cannabis unsaturated fatty acid is high-grade health edible oil, the content of the unsaturated fatty acid is more than or equal to 92 percent, the omega-6 of linoleic acid is more than or equal to 56 percent, the omega-3 of linolenic acid is more than or equal to 14 percent, and the fructus cannabis unsaturated fatty acid is rich in natural VA, VD and other nutrient substances. The fructus cannabis unsaturated fatty acid has the effects of strengthening brain and nourishing brain, the fructus cannabis unsaturated fatty acid does not contain cholesterol, the senile dementia can be effectively prevented, and abundant linoleic acid and linolenic acid can remove impurities metabolized in blood vessels, purify blood and provide fresh blood for brain. Alpha-linolenic acid is a polyunsaturated fatty acid with three double bonds, is an omega-3 essential fatty acid, is used for improving intelligence, resisting thrombus, protecting liver and the like, and is one of essential nutrients for people.
The soybean phospholipid is obtained by introducing unsaturated fatty acids such as medium carbon chain grease, conjugated linoleic acid, vegetable catalpol or n-3 series polyunsaturated fatty acids and the like into phospholipid molecules through ester exchange reaction, not only maintains good emulsification property of the phospholipid, but also greatly improves the nutritional value of the phospholipid, and has better absorption promoting effect when being matched with DHA algal oil and fructus cannabis unsaturated fatty acids. The soybean lecithin is similar to a key or a carrier of a blood brain barrier, carries nutrient components such as DHA in DHA algae oil and linolenic acid omega-3 in fructus cannabis unsaturated fatty acid, enters the brain through the blood brain barrier, supplements nutrition for the brain of a human body, improves memory and prevents senile dementia.
The nutrient substances in the drop containing the fructus cannabis unsaturated fatty acid are reasonable in proportion and meet the international requirements. The above nutritional ingredients are reasonably matched, and the product has effects of improving memory, preventing senile dementia, reducing blood lipid, etc.
In a preferred embodiment, in said drops containing cannabis unsaturated fatty acids, said flavouring agent is a mixture consisting of ethyl maltol and a food flavour, wherein the mass ratio of ethyl maltol to food flavour is 2: 3.
The ethyl maltol and the food essence in the fructus cannabis unsaturated fatty acid-containing drop are diffused into the mouth, so that the bad smell of the raw material components in the drop is completely covered, and the drop is easier to eat.
In a preferred embodiment, said drops containing edestin fatty acids further comprise an antioxidant in a quantity comprised between 0.1% and 1.5% by mass, preferably between 0.5% and 1% by mass.
In a preferred embodiment, in said drops containing edestin fatty acids, the antioxidant may be vitamin C, vitamin E or a combination thereof.
In a preferred embodiment, the drops containing unsaturated fatty acids of fructus cannabis according to the invention have a particle size of 350-500nm, preferably 400-450 nm.
The drop is a liquid preparation, is a heterogeneous liquid dispersion system formed by mutually dispersing multiple phases, and has the particle size of 350-500nm, preferably 400-450nm, so that the drop is more stable and has longer shelf life.
In yet another aspect, the present invention provides a method for preparing drops containing hemp seed unsaturated fatty acid, comprising the steps of:
1) uniformly mixing DHA algae oil, fructus Cannabis unsaturated fatty acid, soybean phospholipid, correctant and fructus Cannabis oil to obtain mixed solution;
2) introducing nitrogen into the prepared mixed solution for protection, and discharging at the temperature of 20 ℃;
3) sterilizing, vacuum defoaming, and packaging to obtain drop containing fructus Cannabis unsaturated fatty acid.
In a preferred embodiment, in the method for the preparation of drops containing edestin unsaturated fatty acids according to the invention, an antioxidant is also added in step 2), wherein the antioxidant may be vitamin C, vitamin E or a combination thereof.
The invention adopts a step-by-step mixing mode, and the raw materials are sequentially and gradually added into a mixing system to be fully dispersed, the obtained drop is stable in dispersion, the stability is improved under the protection of nitrogen gas, and the storage period is prolonged.
In conclusion, compared with the prior art, the fructus cannabis unsaturated fatty acid-containing drops and the preparation method thereof have the following beneficial effects:
(1) after the DHA algae oil, the fructus cannabis unsaturated fatty acid, the soybean lecithin and the fructus cannabis oil are combined in a reasonable proportion, the drop containing the fructus cannabis unsaturated fatty acid is reasonable in proportion of nutritional ingredients and high in bioavailability of the nutritional ingredients, can especially remarkably improve the bioavailability of the DHA in vivo, and has various effects of assisting in enhancing memory, preventing senile dementia, reducing blood fat and the like; the drop adopts a nitrogen gas protection mode, and the particle size of the drop is 350-500nm, preferably 400-450nm, so that the drop is more stable, the shelf life is longer and can be prolonged to 2 years, and therefore, the drop containing the hemp seed unsaturated fatty acid has the advantage of long storage period.
(2) The preparation method of the drop has the advantages of simple operation and suitability for large-scale production.
Detailed Description
For a better understanding of the present invention, the following examples are set forth to illustrate, but are not to be construed as the limit of the present invention.
Hereinafter, the reagents used in the examples of the present invention are all conventional in the art, if not specifically described. The corrigent is a mixture of ethyl maltol and food essence according to the mass ratio of 2: 3. The selected DHA algae oil is a raw material with DHA content of 75%, and the fructus Cannabis unsaturated fatty acid is a raw material with alpha-linolenic acid content of 55%.
Example 1: preparation of drops containing fructus Cannabis unsaturated fatty acid
The components and the corresponding mass percentages thereof are shown in table 1 and are prepared by the following steps:
1) uniformly mixing DHA algae oil, fructus Cannabis unsaturated fatty acid, soybean phospholipid, correctant and fructus Cannabis oil to obtain mixed solution;
2) introducing nitrogen into the prepared mixed solution for protection, and discharging at the temperature of 20 ℃;
3) sterilizing, vacuum defoaming, and packaging to obtain drop containing fructus Cannabis unsaturated fatty acid.
Example 2 to example 6: preparation of drops containing fructus Cannabis unsaturated fatty acid
Drops containing hemp seed unsaturated fatty acid were prepared according to the same procedure as in example 1, with the mass percentages of the components in examples 2 to 6 in table 1.

Example 7: preparation of drops containing fructus Cannabis unsaturated fatty acid
The components and their contents were the same as in example 1, except that they were prepared by the following steps:
1) uniformly mixing DHA algae oil, fructus Cannabis unsaturated fatty acid, soybean phospholipid, correctant and fructus Cannabis oil to obtain mixed solution;
2) discharging the prepared mixed solution at 20 ℃;
3) sterilizing, and packaging to obtain drop containing fructus Cannabis unsaturated fatty acid.
Example 8: preparation of drop containing fructus Cannabis unsaturated fatty acid
The DHA algae oil, fructus cannabis unsaturated fatty acid, soybean phospholipid and flavoring agent have the same mass percentage content as that in example 1, and are different from example 1 in that the soybean phospholipid and flavoring agent also contain 0.1 mass percentage of antioxidant vitamin C, and the balance is fructus cannabis oil. The preparation method comprises the following steps:
1) uniformly mixing DHA algae oil, fructus Cannabis unsaturated fatty acid, soybean phospholipid, correctant and fructus Cannabis oil to obtain mixed solution;
2) adding an antioxidant into the prepared mixed solution, uniformly mixing, introducing nitrogen for protection, and discharging at the temperature of 20 ℃;
3) sterilizing, vacuum defoaming, and packaging to obtain drop containing fructus Cannabis unsaturated fatty acid.
Example 9: preparation of drops containing fructus Cannabis unsaturated fatty acid
The DHA algae oil, fructus cannabis unsaturated fatty acid, soybean phospholipid and flavoring agent have the same mass percentage content as that in example 1, and are different from that in example 1 in that the DHA algae oil, fructus cannabis unsaturated fatty acid, soybean phospholipid and flavoring agent also contain 1 mass percentage of antioxidant vitamin E, and the balance is fructus cannabis oil. The preparation method is the same as example 8.
Example 10: preparation of drop containing fructus Cannabis unsaturated fatty acid
The DHA algae oil, fructus cannabis unsaturated fatty acid, soybean phospholipid and flavoring agent are the same as those in example 1 in mass percentage, and the DHA algae oil, fructus cannabis unsaturated fatty acid, soybean phospholipid and flavoring agent are different from those in example 1 in that the DHA algae oil, fructus cannabis unsaturated fatty acid, soybean phospholipid and flavoring agent also contain 0.5% of antioxidant in mass percentage, the antioxidant is a mixture of vitamin C and vitamin E according to the mass ratio of 1:1, and the balance is fructus cannabis oil. The preparation method is the same as example 8.
In comparative examples 1 to 3 below, the content of the substitute component was the same as that of the component substituted in example 1, and the other components and their contents were the same as in example 1.
Comparative example 1: preparation of oral drops
The preparation was carried out according to the same procedure as in example 1, except that monoglyceride was used instead of soybean phospholipid in example 1, as in example 1.
Comparative example 2: preparation of oral drops
The preparation was carried out according to the same procedure as in example 1, except that olive oil was used instead of the hemp seed oil in example 1 from example 1.
Comparative example 3: preparation of oral drops
The preparation was carried out according to the same procedure as in example 1, except that olive oil was used instead of the hemp seed unsaturated fatty acid in example 1 from example 1.
Comparative example 4: preparation of oral drops
The preparation was carried out in the same manner as in example 1, except that the unsaturated fatty acid content of hemp seed was 70%, the DHA algal oil, soybean phospholipids and taste-masking agent were added in the same manner as in example 1, and the balance was supplemented with hemp seed oil, as in example 1.
Comparative example 5: preparation of oral drops
The preparation was carried out according to the same procedure as in example 1, except that the DHA algal oil was added in an amount of 30%, the hemp seed unsaturated fatty acid, soybean phospholipid, and taste modifier were added in the same amounts as in example 1, and the balance was supplemented with hemp seed oil.
Examples of the experiments
And (3) bioavailability test:
male Wistar rats (provided by the Experimental animal center of the department of medicine of Beijing university) with a weight of 260-. Rats were intragastrically perfused with the drops of examples 1-10 and comparative examples 1-6, respectively, and after administration for 0.25h, 0.5h, 1h, 2h, 4h, 6h, 8h, 12h and 24h, orbital venous plexus blood was collected at 0.5mL in heparinized tubes, incubated with an oil bath of hemp at 37 ℃ for 30min, then centrifuged at 5000r/min for 10min, plasma was separated and the supernatant was collected and stored at-20 ℃ for determination of drug concentration. The data from the DHA blood concentration-time curve measurements were calculated by DAS pharmacokinetics and are shown in table 2 below.

As can be seen from the test data in table 2, the interaction between the components in the drops, the replacement of the components and the variation of the distribution ratio of the components all have an effect on the absorption of the components. The application protects that the DHA bioavailability of the drops with the specific proportioning components is improved. Specifically, the bioavailability of DHA in drops by rats in examples 1-10 was significantly higher than that of drops by rats in comparative examples 1-5, and especially the bioavailability of DHA was the highest in the best examples 9-10.
The above description is only a preferred embodiment of the present invention, and the protection scope of the present invention is not limited to the above embodiments, and all technical solutions belonging to the idea of the present invention belong to the protection scope of the present invention. Claims (8)
1. A drop containing fructus cannabis unsaturated fatty acid comprises the following components in percentage by mass:
DHA algal oil: 5 to 25 percent;
fructus cannabis unsaturated fatty acids: 5 to 65 percent;
soybean lecithin: 3 to 15 percent;
flavoring agent: 0.1 to 0.8 percent;
hemp seed oil: and (4) the balance.
2. A drop containing fructus cannabis unsaturated fatty acid as claimed in claim 1, comprising the following components in mass percent:
DHA algal oil: 10 to 20 percent;
fructus cannabis unsaturated fatty acids: 10 to 60 percent;
soybean lecithin: 5 to 10 percent;
flavoring agent: 0.3 to 0.5 percent;
hemp seed oil: and (4) the balance.
3. Drops containing edestin fatty acids according to claim 1 or 2, wherein the flavouring is a mixture of ethyl maltol and food essence, wherein the mass ratio of ethyl maltol to food essence is 2: 3.
4. Drops containing edestin fatty acids according to claim 1 or 2, wherein they further comprise an antioxidant in a percentage by mass ranging from 0.1% to 1.5%, preferably from 0.5% to 1%.
5. Drops containing edestan unsaturated fatty acids according to claim 4, wherein said antioxidant is vitamin C, vitamin E or a combination thereof.
6. Drops containing fructus cannabis unsaturated fatty acid according to claim 1 or 2, wherein the particle size of the drops containing fructus cannabis unsaturated fatty acid is 350-500nm, preferably 400-450 nm.
7. A method for preparing drop containing fructus Cannabis unsaturated fatty acid comprises the following steps:
1) uniformly mixing DHA algae oil, fructus Cannabis unsaturated fatty acid, soybean phospholipid, correctant and fructus Cannabis oil to obtain mixed solution;
2) introducing nitrogen into the prepared mixed solution for protection, and discharging at the temperature of 20 ℃;
3) sterilizing, vacuum defoaming, and packaging to obtain drop containing fructus Cannabis unsaturated fatty acid.
8. The method according to claim 7, wherein an antioxidant is further added in step 2), wherein the antioxidant is vitamin C, vitamin E or a combination thereof.
