Abstract
本发明涉及益生菌应用技术领域,特别涉及一种含益生菌的口腔速溶膜及其制备方法。所述益生菌口腔速溶膜外表光滑,质地轻薄,速溶效果好,携带方便,无需饮用水即可食用,特别适用于儿童、老人及有吞咽障碍的人群食用,可广泛应用于食品或保健品或药品生产中。所述速溶膜中凝结芽孢杆菌VHProbi C08活菌含量高且稳定性好,食用该速溶膜有助于清除自由基和降解胆固醇,能有效预防和缓解腹泻,促进肠道健康。此外,所述速溶膜柔韧性好,可折叠,防潮性较好,可通过3D打印技术打印出不同图案,广泛应用于食品包装中,应用前景广阔。

技术领域
本发明涉及益生菌应用技术领域,特别涉及一种可有效防潮的益生菌口腔速溶膜及其制备方法。
背景技术
膜剂(films)是一种将药物溶解或均匀分散于成膜材料中或包裹于成膜材料隔室内,加工成型的单层、多层(复合层)、夹心层的膜剂。从应用部位可分为口腔膜、眼用膜、鼻腔用膜、宫颈及阴道用膜、植入膜剂和经皮给药型膜剂(用作皮肤和黏膜创伤、烧伤或炎症表面的覆盖)等。口腔速溶膜是一种近年来在国外日益广泛应用的口服固体速释新剂型,表现出的许多优点引起广泛的关注,许多制药公司都将快速溶解技术作为一个研究重点。口腔速溶膜剂(oral fast dissolving film,OFDF)是将一定剂量的化学药品或中药提取物的有效成分载入膜材后制成薄膜片,置于舌上后无需饮水即可在 1 min 内快速溶解、释放药物。此剂型在口腔中能够快速分解,形状多为矩形,参照儿童舌上面积其大小一般设计在6 mm 2 以内,病人可以根据用药量按照膜剂上的刻痕裁剪与服用。口腔速溶膜剂相比于其他口服固体制剂来说,有着无需用水送服、不必担心会噎住或吸入,且生产工艺简单,成本较低等优势;相比于口服液体制剂来说,其分剂量准确且便于携带,大大提高了病人用药顺应性与方便性,尤其适用于儿童、老人及有吞咽困难的人群用药。
目前口腔速溶膜剂主要集中在北美市场,如治疗儿童感冒咳嗽的盐酸苯海拉明、美沙芬及治疗感冒鼻塞的盐酸苯福林等。目前我国使用较多的口服制剂包括溶液剂、糖浆剂、混悬剂、泡腾剂、咀嚼片、口崩片等,虽然这些剂型占据着较大的市场份额,但是均存在着分剂量不准确,口服吞咽困难等局限性。近年来我国在开发新的药制剂方面取得了一定的成效,但新剂型所占比重还较小,开发速度也较缓慢。速溶膜剂型国内未见上市产品,均处于在研状态。
益生菌(Probiotics)是一类对宿主有益的活性微生物,是定植于人体肠道、生殖系统内,能产生确切健康功效从而改善宿主微生态平衡、发挥对肠道有益作用的活性有益微生物的总称。人体、动物体内有益的细菌或真菌主要有:酪酸梭菌、乳酸菌、双歧杆菌、嗜酸乳杆菌、放线菌、酵母菌等。益生菌对人体产生的功效可以说非常强大。可以帮助营养物质的消化吸收,许多益生菌株在胃肠道内可产生消化酶,这些酶可帮助人体更好地消化所摄入的食品及吸收食品中的营养成分。益生菌还可竞争性抑制有害微生物吸收营养物质及进入血液循环系统。嗜酸乳杆菌就是这方面的代表菌株,可分泌消化乳糖的乳糖酶从而缓解乳糖不耐症。益生菌还可以产生重要的营养物质,如维生素包括泛酸、尼克酸、B1,B2,B6及维生素K等,同时能产生短链脂肪酸、抗氧化剂、氨基酸等等,对骨骼成长和心脏健康有重要作用。还能抑制有害菌的生长,抑制、清除有害菌产生的毒素。可以预防和治疗某些疾病,如肠道综合症、呼吸道感染、生殖系统感染、过敏、口臭、胃溃疡等益生菌可以分为乳杆菌类,双歧杆菌类,革兰(+)球菌类。益生菌可以防治腹泻,缓解乳糖不耐受症状,预防阴道感染,增强人体免疫力,缓解过敏作用,降低血清胆固醇,预防癌症,抑制肿瘤生长。
目前世界上已开发出数以百计的益生菌保健产品。其中包括含益生菌的酸牛奶、酸乳酪、酸豆奶以及含多种益生菌的口服液、片剂、胶囊、粉末剂、抑菌喷剂等等。益生菌有益健康的理念也已经深入人心,随着科学研究的不断深入,我国对益生菌产品的需求大幅增加,益生菌也迎来了井喷式的发展。尤其是针对儿童的益生菌产品的研究非常火热。儿童肠道比较脆弱,所以家长们更青睐选择益生菌产品来调节儿童肠道的消化、吸收等问题。
针对儿童这一特殊人群,现有的益生菌产品剂型存在儿童不喜其口感、吞咽困难、容易堵塞喉咙等问题,所以研制出一种含有益生菌的口腔速溶膜将是未来儿童益生菌产品发展的新趋势。
发明内容
本发明为解决现有技术问题,提供了一种含益生菌的口腔速溶膜及其制备方法。所述益生菌口腔速溶膜中活菌量高,其稳定性、防潮性和速溶性良好,是一种十分具有发展前景的益生菌新剂型。
本发明一方面涉及一种口腔速溶膜,是将含益生菌的原料加工成膜制得的。
所述原料的组分及其重量份数分别为:糯米粉6~10份、木薯淀粉4~6份、玉米淀粉1.5~3份、蔓越莓粉0.8份、白砂糖1.25份、大豆卵磷脂1.25份、甘油1份、益生菌菌粉1份、纯净水95份。
进一步优选的,所述原料的组分及其重量份数分别为:糯米粉8份、木薯淀粉4份、玉米淀粉2份、蔓越莓粉0.8份、白砂糖1.25份、大豆卵磷脂1.25份、甘油1份、益生菌菌粉1份、纯净水95份。
进一步优选的,所述益生菌菌粉中活菌量不低于109CFU/mL。
进一步优选的,所述益生菌为凝结芽孢杆菌 VHProbi C08(Bacillus coagulansVHProbi C08),已于 2019 年 9 月 23 日保藏于中国武汉大学的中国典型培养物保藏中心,其保藏号为 CCTCC NO:M2019738。
本发明一方面涉及一种益生菌口腔速溶膜的制备方法,包括如下步骤:
(1)按重量份数分别称取糯米粉6~10份、木薯淀粉4~6份、玉米淀粉1.5~3份、蔓越莓粉0.8份、白砂糖1.25份、大豆卵磷脂1.25份、甘油1份、益生菌菌粉1份、纯净水95份;
(2)将木薯淀粉和玉米淀粉混合,加入30%-40%步骤(1)称取的纯净水,充分混匀,70℃水浴1~2h,使淀粉充分糊化;加入大豆卵磷脂、甘油,混匀,得到淀粉糊;
(3)将糯米粉与蔓越莓粉、白砂糖、益生菌菌粉混合,加入剩余的纯净水,充分混匀,制成糯米浆;
(4)待淀粉糊冷却至45℃,与糯米浆混匀,得混合浆液;
(5)将混合浆液制膜、干燥、切割,即得口腔速溶膜。
本发明还提供了所述益生菌口腔速溶膜在食品或保健品或药品生产中的应用。
本发明制备得到的益生菌口腔速溶膜外表光滑,质地轻薄,厚度仅有0.042mm;速溶效果好,入口即化,在30~42℃纯净水中,可以在12~18s内迅速溶解。该速溶膜携带方便,无需饮用水即可食用,特别适用于儿童、老人及有吞咽障碍的人群食用,可广泛应用于食品或保健品或药品生产中。
所述速溶膜中凝结芽孢杆菌VHProbi C08活菌含量高,达7.1×108CFU/g,且稳定性好,60天内活菌量保持稳定。食用该速溶膜有助于清除自由基和降解胆固醇,能有效预防和缓解腹泻,促进肠道健康。
此外,所述速溶膜柔韧性好,可折叠,防潮性较好,还可用于包裹其他益生菌,起到防潮,防粘袋的作用。由于其状态类似于纸,可通过3D打印技术打印出不同图案,广泛应用于食品包装中,应用前景广阔。

具体实施方式
实施例只是对本发明的说明,并不是对本发明保护范围的限制。在不背离本
发明精神和实质的情况下,对本发明方法、步骤或条件所作的修改或替换,均属本发明的范围。
本发明实施例中选用的凝结芽孢杆菌 VHProbi C08(Bacillus coagulansVHProbi C08)筛选自酸白菜样品,其16s rDNA序列为SEQ ID NO:1,已于 2019 年 9 月 23日保藏于中国武汉大学的中国典型培养物保藏中心,其保藏号为 CCTCC NO:M2019738。
凝结芽孢杆菌 VHProbi C08 不产生溶血素,不会溶解血细胞,对常见抗生素敏感,生物安全性良好;能够耐受较高的盐度,最大耐受盐浓度为 7%;能在较广阔的温度范围内生长,并且能产生耐热性很强的孢子,方便该益生菌株后期工业化加工;体外细胞表面疏水性为 8.58%,对肠道细胞具有粘附性。
凝结芽孢杆菌 VHProbi C08 具备较强的抗氧化功能,其中对 DPPH 自由基清除率为 87.80%,HRS 自由基清除率为 47.79%;上清液抗脂质过氧化抑制率为 84.46%,菌体抗脂质过氧化抑制率为 44.88%。
凝结芽孢杆菌 VHProbi C08 具备较强的降解胆固醇能力,其中体外胆固醇降解率达到 39.21%;在高血脂小鼠模型中,用本发明提供的凝结芽孢杆菌VHProbi C08 喂食模型鼠 30 天后预处理组血液胆固醇水平降低了约 34%,后处理组血液胆固醇水平降低了约10%;40 天后,凝结芽孢杆菌 VHProbi C08预处理组血液胆固醇水平降低了约 39%,后处理组血液胆固醇水平降低了约30%,效果非常显著。
凝结芽孢杆菌 VHProbi C08 能有效预防和缓解腹泻,抑制肠蠕动。给大鼠连续服用凝结芽孢杆菌 VHProbi C08 7 天后,能有效延迟腹泻时间,减少粪便量。凝结芽孢杆菌VHProbi C08 代谢产生的有机酸等代谢产物能够抑制有害菌的生长,促进肠道对营养物质的消化和吸收,恢复肠道的微生物屏障功能,刺激机体免疫功能,缩短疾病病程。
下面就结合具体实施例,对本发明做进一步阐述。
实施例1 凝结芽孢杆菌 VHProbi C08 菌粉的制备
1、凝结芽孢杆菌产孢子培养基各组分为:玉米淀粉 10~40g/L、葡萄糖 0. 5~3g/L、豆粕 5~25g/L、碳酸钙 0. 1~1. 5g/L、氯化铵 0. 1~1. 0g/L、磷酸氢二钾0. 1~1. 0g/L、硫酸镁 0. 01~0. 7g/L 和硫酸锰 0. 01~0. 7g/L;将培养基各组分倒入发酵罐中,加水溶解;再调节 pH 为 7. 5~7. 8,然后在 121℃条件下高温灭菌 20min 得到产芽孢培养基。
2、无菌条件下,将凝结芽孢杆菌 VHProbi C08 种子液以 3%的体积比接种于发酵罐中,40℃条件下培养 23~36h,其发酵条件为:转速为 180~200rpm,通风量为 1:1. 2~1. 5,罐压为 0. 05~0. 08MPa,发酵罐的装料量为 60~70%。发酵完成后,将所述发酵罐升温至 50~60℃,维持 20min,得到凝结芽孢杆菌 VHProbiC08 的发酵液。
3、将凝结芽孢杆菌 VHProbi C08 的发酵培养液在 4℃,5000r/min 条件下离心10min,收集菌体后用无菌生理盐水洗涤 2 次,重悬菌体制成菌悬液备用。按照质量比为1:4:1:1:0. 1:1:8 称取大豆蛋白 25g、脱脂奶粉 100g、玉米淀粉25g、蔗糖 25、阿拉伯胶2.5g、低聚果糖 25g 和乳酸钙 200g,加蒸馏水定容至1L,制备成包埋材料溶液。包埋材料溶液和菌悬液按照一定的比例混匀,使凝结芽孢杆菌 VHProbi C08 的终浓度为 2~3×10 9CFU/mL。采用离心式喷雾干燥器进行喷雾干燥处理,干燥塔进风温度为 130℃,出风温度为70℃,雾化压力为 0.5MPa,输送混合液的蠕动泵转速为 11r/min,粒径为 50~90μm,收集菌粉,并测定菌量为 2.35×10 10 CFU/g。
实施例2 一种益生菌口腔速溶膜及其制备方法
一种益生菌口腔速溶膜,是通过如下方法制备得到的:
(1)按重量份数分别称取糯米粉6份、木薯淀粉5份、玉米淀粉1.5份、蔓越莓粉0.8份、白砂糖1.25份、大豆卵磷脂1.25份、甘油1份、凝结芽孢杆菌 VHProbi C08菌粉1份、纯净水95份;

实施例3 一种益生菌口腔速溶膜及其制备方法
一种益生菌口腔速溶膜,是通过如下方法制备得到的:
(1)按重量份数分别称取糯米粉8份、木薯淀粉4份、玉米淀粉2份、蔓越莓粉0.8份、白砂糖1.25份、大豆卵磷脂1.25份、甘油1份、凝结芽孢杆菌 VHProbi C08菌粉1份、纯净水95份;
(2)将木薯淀粉和玉米淀粉混合,加入35%步骤(1)称取的纯净水,充分混匀,70℃水浴1h,使淀粉充分糊化;加入大豆卵磷脂、甘油,混匀,得到淀粉糊;
(3)将糯米粉与蔓越莓粉、白砂糖、凝结芽孢杆菌 VHProbi C08菌粉混合,加入剩余的纯净水,充分混匀,制成糯米浆;
(4)待淀粉糊冷却至45℃,与糯米浆混匀,得混合浆液;
(5)将混合浆液制膜、干燥、切割,即得口腔速溶膜。
实施例4 一种益生菌口腔速溶膜及其制备方法
一种益生菌口腔速溶膜,是通过如下方法制备得到的:
(1)按重量份数分别称取糯米粉10份、木薯淀粉6份、玉米淀粉3份、蔓越莓粉0.8份、白砂糖1.25份、大豆卵磷脂1.25份、甘油1份、凝结芽孢杆菌 VHProbi C08菌粉1份、纯净水95份;
(2)将木薯淀粉和玉米淀粉混合,加入40%步骤(1)称取的纯净水,充分混匀,70℃水浴1h,使淀粉充分糊化;加入大豆卵磷脂、甘油,混匀,得到淀粉糊;
(3)将糯米粉与蔓越莓粉、白砂糖、凝结芽孢杆菌 VHProbi C08菌粉混合,加入剩余的纯净水,充分混匀,制成糯米浆;
(4)待淀粉糊冷却至45℃,与糯米浆混匀,得混合浆液;
(5)将混合浆液制膜、干燥、切割,即得口腔速溶膜。
实施例5 益生菌口腔速溶膜的性能评价及活菌含量测定
5.1 物理性能评价
如图1所示,实施例2、3、4制得的益生菌口腔速溶膜均为透明淡粉色,表面平整光滑,厚度均在0.042~0.058mm之间,。
5.2 水溶性测定
取实施例2、3、4制得的口腔速溶膜各1cm2,分别置于37℃纯净水中,准确记录口腔速溶膜完全溶解的时间。
结果显示:本发明提供的口腔速溶膜均能在12~18s内完全溶解,溶解性良好。
5.3 活菌含量测定
用葡萄糖酵母提取物BC琼脂培养基(1L)进行凝结芽孢杆菌活菌数的测定,其配方如下:酵母粉5.0g,蛋白胨5.0g,葡萄糖5.0g,K2HPO4 0.5g,KH2PO4 0.5g,MgSO4 0.3g,微量元素溶液1.0mL,琼脂粉15.0g,蒸馏水1000mL ,调pH6.3,121℃灭菌15min。
微量元素溶液(mg/mL):NaCl 10g,7H2O·FeSO4 18g,H2O·MnSO4 16g,7H2O·ZnSO4 1.6g,5H2O·CuSO4 1.6g,7H2O·CoSO4 1.6g,过滤除菌。
取实施例2、3、4制得的口腔速溶膜各0.2g,分别加入19.8mL0.85%的生理盐水中,均质机拍打3min,使其完全溶解并混匀,混匀后的样液在无菌环境中进行10倍梯度稀释,直到最后的稀释预计含有大约30个菌落形成单位(CFU)/mL。在10~20分钟内稀释样品,选取最后三个稀释剂用于分析。每个稀释度吸取1mL样品匀液于无菌平皿内,将冷却至48 ℃的BC琼脂培养基倾注入平皿约15mL,转动平皿使混合均匀。40℃±2℃厌氧培养48h。孵育48小时后,计数培养皿上的菌落。计数含有30到300个菌落的平板,只计算以下出现的菌落:在BC琼脂培养基表面菌落直径应在1 mm ~ 5 mm之间,白色至乳白色,凸出,边缘完整,表面光滑;在BC琼脂培养基内部的菌落直径应为0.5 mm~1 mm,呈奶油色针尖状。待长出单菌落后进行计数。计算出口腔速溶膜中活菌量,具体结果见表1。
菌量计算方法:菌量=平均单菌落数*10-5*100(CFU/g)。

从表1的数据可以看出,本发明提供的口腔速溶膜中活菌量非常高,有利于发挥益生菌的益生功能。
5.4 稳定性评价
取实施例3所制备得到的口腔速溶膜,真空包装后,置于25℃的恒温箱中保存,分别在第4、10、17、24、32、39、47、54、60天时,各取0.2g口腔速溶膜测定其活菌含量,具体方法同实施例5.3。
具体结果见表2和图2。

从表2的数据可知,本发明提供的口腔速溶膜在60天内活菌量几乎无变化,稳定性良好。
此外,由常规试验及小鼠模型试验结果可知,食用本发明所述的口腔速溶膜,能有效提高机体的抗氧化性能,降低胆固醇含量,预防和缓解腹泻。儿童食用无吞咽困难、堵塞喉咙等问题,能显著促进儿童肠道对营养物质的消化和吸收,调节肠道菌群平衡与健康,增强儿童的免疫力。
综上所述,本发明提供的益生菌口腔速溶膜外表光滑,质地轻薄,速溶效果好,携带方便,无需饮用水即可食用,特别适用于儿童、老人及有吞咽障碍的人群食用,可广泛应用于食品或保健品或药品生产中。所述速溶膜中凝结芽孢杆菌VHProbi C08活菌含量高且稳定性好,食用该速溶膜有助于清除自由基和降解胆固醇,能有效预防和缓解腹泻,促进肠道健康。此外,所述速溶膜柔韧性好,可折叠,防潮性较好,可通过3D打印技术打印出不同图案,广泛应用于食品包装中,应用前景广阔。

Claims (8)
1.一种口腔速溶膜,其特征在于,所述的口腔速溶膜是将含益生菌的原料加工成膜制得的。
2.如权利要求1所述的口腔速溶膜,其特征在于,所述原料的组分及其重量份数分别为:糯米粉6~10份、木薯淀粉4~6份、玉米淀粉1.5~3份、蔓越莓粉0.8份、白砂糖1.25份、大豆卵磷脂1.25份、甘油1份、益生菌菌粉1份、纯净水95份。
3.如权利要求2所述的口腔速溶膜,其特征在于,所述原料的组分及其重量份数分别为:糯米粉8份、木薯淀粉4份、玉米淀粉2份、蔓越莓粉0.8份、白砂糖1.25份、大豆卵磷脂1.25份、甘油1份、益生菌菌粉1份、纯净水95份。
4.如权利要求1-3任一所述的口腔速溶膜,其特征在于,所述益生菌为凝结芽孢杆菌VHProbi C08(Bacillus coagulans VHProbi C08),已于 2019 年 9 月 23 日保藏于中国武汉大学的中国典型培养物保藏中心,其保藏号为 CCTCC NO:M2019738。
5.如权利要求4所述的口腔速溶膜,其特征在于,所述益生菌菌粉中活菌量不低于109CFU/mL。
6.权利要求1-5任一所述的口腔速溶膜在食品、保健品或药品生产中的应用。
7.一种益生菌口腔速溶膜的制备方法,其特征在于,所述制备方法包括如下步骤:
(1)按重量份数分别称取糯米粉6~10份、木薯淀粉4~6份、玉米淀粉1.5~3份、蔓越莓粉0.8份、白砂糖1.25份、大豆卵磷脂1.25份、甘油1份、益生菌菌粉1份、纯净水95份;
(2)将木薯淀粉和玉米淀粉混合,加入30%-40%步骤(1)称取的纯净水,充分混匀,70℃水浴1~2h,使淀粉充分糊化;加入大豆卵磷脂、甘油,混匀,得到淀粉糊;
(3)将糯米粉与蔓越莓粉、白砂糖、益生菌菌粉混合,加入剩余的纯净水,充分混匀,制成糯米浆;
(4)待淀粉糊冷却至45℃,与糯米浆混匀,得混合浆液;
(5)将混合浆液制膜、干燥、切割,即得口腔速溶膜。
8.如权利要求7所述的制备方法,其特征在于,所述益生菌为凝结芽孢杆菌 VHProbiC08(Bacillus coagulans VHProbi C08),已于 2019 年 9 月 23 日保藏于中国武汉大学的中国典型培养物保藏中心,其保藏号为 CCTCC NO:M2019738。
