Technical field
The method of solid solution perforator (SSP) is made and produced to relate generally to of the present invention, such as with sharp metal or glass needle or Precision Machining and/or the making of molding subsequently solubility microneedle. The invention particularly relates to a kind of manufacture method and use thereof that produces the minisize mould structure of making by curing materials from fine needle arrayed (fine needle array alinment). In addition, the present invention relates to improve the method for microneedle mechanical strength and the method and the use thereof of patch injection/insertion by designing flexible microneedle patch.
Background technology
Percutaneous or Intradermal medicine are sent, and comprise sending of protein and vaccine, are the methods of a kind of very effective acquisition whole body or local pharmacological action. But, when providing enough drug osmotic, have obstacle through skin.Skin is made up of multilamellar.Horny layer is an outermost layer, then is visible epidermal area, is the dermal tissue layer at last. The horny layer thin layer representative of 10~50 μ m is by the topmost obstacle of dermal delivery medicine.According to physics and chemical property, particularly lipophile and the molecular weight of crude drug, horny layer accounts for the skin barrier property of 50%~90% prevention percutaneous dosing.
Because the sampling of intradermal administration or medicine can realize and painless depletion of blood by reducing above-mentioned obstacle, the use microneedle is favourable in dermal delivery and Intradermal are sent." microneedle " used herein speech refers to a plurality of slim-lined constructions, and its long enough enters epidermal area or corium or subcutaneous tissue to penetrate the cutin skin layer.Although need in some cases or wish to be penetrated into skin corium, microneedle generally not like this length can be penetrated into skin corium. As using substituting of hypodermic needle drug administration by injection, the use of microneedle is disclosed in the U.S. Patent No. 3964,482, in this patent, pass horny layer with the array of solid or hollow microneedle and enter into epidermal area.Liquid can be provided and delivered in the following manner: by hollow microneedle or the solid ledge by porous, or around the solid ledge of the not porous that is surrounded by seepy material or hole.Coming the speed of control drug release, the mechanism that medicine transports with membrane material is to absorb.
At the open No.WO98/00193 of PCT, WO97/48440, WO97/48441 discloses the microneedle and miniature sword (microblade) structure of other types among WO97/48442 and the WO96/37256.Microneedle (diameter is less than 1mm) has been used to carry out percutaneous dosing, and microneedle also has been used for the chamber delivering drugs by pin, with outside delivering drugs along needle shaft, and the patch medicinal application after perhaps being used for as skin perforator. For example, utilized micro-processing method or MEMS (MEMS) processing method that silicon system microneedle is developed.Example is referring to U.S. Patent No. 6,334, and 856,6,256,533,6,312,612 and 6,379,324.Unfortunately, the silicon pin can not dissolve in skin, in use can fracture and stays in the skin histology, thereby produce sizable stimulation even infection.Non-silicon system microneedle also is developed, and example is participated in U.S. Patent No. 6,334,856 and 6,091,975.But, in skin, can not dissolve or dissolve slowly (promptly less than several hrs) with the microneedle that metal or plastics are made, therefore be generally used for providing the micro-tube that from medicament reservoir, transports medicine, or be used to produce micropore.
Typically, microneedle is made with the MEMS processing method. In U.S. Patent No. 6,663, the use of polydimethylsiloxane (PDMS) mould of the microneedle that is used to water polymer injection is disclosed in 820 and No.6,334,856, wherein the positivity material of microneedle (positive matter) is to make with MEMS technology. But making mother matrix (master) microneedle array with MEMS can be very expensive and complicated, and the miniature pin of polymer may need medicine to load (loading) or medicine coating (coating), makes pouring procedure not be suitable for large-scale production.
Summary of the invention
The present invention has overcome these problems and the inexpensive and uncomplicated method that is used to produce the SSP drug delivery system that comprises the solubilized microneedle is provided.Particularly, the invention discloses a kind of manufacture method of the positive microneedle mother matrix mould of making by dissimilar fine needle arrays.
The used microneedle of the present invention is by by metal, polymer, or the mould that glass (or other is extensible) material silk is made makes. In order to make positive mother matrix mould by the pin of alignment, the one pin that is used for positive mother matrix be by as the end of polishing silk or wire drawing then the method for sharpening make.Other method that is fit to the sharp pin of manufacturing is known, and will use in this article.Described pin can have different shapes, is that circle or cross section are square as the cross section. The single pin of being made by silk is integrated or be arranged in the mother matrix structure, and be used to pour into a mould the negative norm tool of microneedle of final solubility than manufacturing very fast relatively and cost is low.
Be the process example of in substrate, arranging microneedle below with hole array. The process that pin is integrated in the orifice plate comprises: (1) makes the first and second dull and stereotyped parallel alignment with hole array and (2) make pin pass through the outstanding length of the preliminary election of the requirement on hole to the second flat board of first and second flat boards. The position of needle point can be determined by following method: (1) utilizes the wall that stops that requiring distance with second flat board apart, and (2) utilize the conical bore in second flat board, or (3) utilize the independent addressable array of actuators that moves single pin.
The method of the positive mother matrix mould of another one manufacturing is by retrofit, as milling, polishing or the boring of computer numerical control (CNC) (CNC).See for example " CNC processing handbook (CNC MachiningHandbook, James Madison, Industrial Press, Inc., 1991) and " CNC processing and programming introduction " (An Introduction to CNC Machining and Programming, Gibbs and Grandell of CNC method be discussed, Industrial Press, Inc.1996).For example, from a steel, can on two vertical direction, cut two groove arrays, the pyramid microneedle that can make side angle simultaneously with requirement with predetermined side wall angle (side-wall angle).
The method of the positive mother matrix mould of another one manufacturing is by negative norm tool cast microneedle, and this negative norm tool makes with the MEMS processing method or by the CNC fine processing method such as boring or polishing. The mould that is called " negative norm tool " among the present invention can be made by mother matrix microneedle array structure, and this mould can be used for making the SSP of solubility. The solubility system comprises the solid matrix of solubility (comprising meltability) material, and solid matrix keeps (hold) one or more selected medicines alternatively, and forms one or more perforators from the negative norm tool.Described substrate can be made up of quick dissolving and/or expanding material.Solid solution can be homogeneous solution, non-homogeneous solution, has the suspension of different drug loading states. In order to make soluble SSP, at first utilize above-described method to make positive mother matrix model, then by the mould of this positive mastering silicon or curing materials.Particularly, the making of this auxiliary silicon system negative norm tool can be carried out cost-effective large-scale production, utilizes the proper property of silicon materials simultaneously, as surface tension, flexible, breathability etc.In another embodiment, when using microneedle array, silicon system negative norm tool just can break away from from microneedle array. In this embodiment since silicon materials the price is fair and inexpensive, with the silicon molding jig as packaging material to keep microneedle array intact.
In the another one embodiment of negative norm tool, the microneedle cavities in the negative norm tool has opening in the bottom corner in chamber, thereby is easy to give this chamber gel filled by this hole being applied vacuum, or even gel is pressed into this chamber.
The hydrogel or the similar plastic material that contain medicine by pouring ladle in negative silicon molding jig are made the SSP microneedle array that comprises medicine. In preparation during solid solution, can make medicine concentrate on the tip of microneedle by cast and centrifuging process, such as described in the PCT announcement No.WO 07/030477, its full content is hereby incorporated by.The conical end of " microneedle tip " expression microneedle.Usually, medicine can concentrate on 1/2nd to 1/3rd places, microneedle bottom, preferably in microneedle bottom 1/4th or part still less taper off to a point. Can utilize drug gel and adhesive phase are repeatedly poured into a mould/wiping process, between microneedle, pour into a mould adhesive phase. As basal layer, except the microneedle part, microneedle array becomes sticky with binding agent (particularly water-based adhesive), and the SSP patch no longer needs the peripheral binding agent of other viscosity on basement membrane.Flexible layers can be layered on the adhesive phase.Final microneedle will be to have flexible and microneedle array autohension. In using miniature pin patch, the medicine carrying patch is installed in the box.Box is connected on the syringe. When carrying out the administration of SSP patch with syringe, the adhesive phase between the microneedle can make the microneedle patch be fixed on the skin.
Box can use in injection device, as in U.S. Patent No. 6,945,952, and No.7,182,747 and No.7, described in 211,062, full content is hereby incorporated by.Described medicine-microneedle array patch is attached to the center of box, thereby makes the tip and injection target contact skin of microneedle.Such as by spin locking, sucking fit, detachable glue, connect by magnetic, or by utilizing the interim locking device at the described box of injector tip, described box is installed in the end of syringe.Drug of topical application person can make the penetration depth of microneedle be consistent by the microneedle that impacts in the described box.Typically, described box is flat and thin, preferably is not thicker than about 10mm.The outside of described box can be any different shape and size.Described box can be made up of plastic plastics.Described box can be designed to disposable use, just uses up to throw away.Described box can link to each other with the piston of syringe and piston moves to skin together. In one embodiment, microneedle array is placed with the close position of target skin rather than place on the piston of syringe. This design is easy to use and can carries out large-scale production and do not lose efficient.Another method of using patch is to insert patch with thumb or finger, can or insert power and the persistent period that device control is inserted by the pressure transducer thin film.
The method that another one effectively penetrates into skin is to improve the mechanical strength of microneedle by the process of the preparation of microneedle and after drying.Particularly, by add monosaccharide or disaccharide in matrix polymer (carboxymethyl cellulose), the mechanical strength of microneedle can be improved.Utilize after drying process (or from microneedle substrate, removing extra water constituent) can improve the mechanical strength of microneedle in addition, with after the mold separation.

It is noted that employed singulative " a ", " an " and " the " comprise a plurality of objects in this description and appended claim, unless the content that clearly limits is arranged in addition. Therefore, for example, mention " protein (a protein) " and comprise two or more mixtures of polypeptides, or the like.
Mold Making
What Figure 1A-1C showed is the sectional view of making the positive microneedle array mother matrix of mould 11, mould 11 comprises that the hole with upper surface and lower surface limits dull and stereotyped 12, optional support plate 13, prong 14 is used for determining the dividing plate 15 and the tip alignment plate 16 (needle tip alignment plate) of the length of microneedle.
Can polish taper metal or glass fiber to make prong 14.Described filament can be an any materials, comprises metal, plastics and/or comprises the pottery of glass, and its acutance is by the production method decision of pin.For metal needle, the polishing silk is to desired acutance usually. For glass or plastic material, can obtain prong by the temperature drawn yarn more than glass transition temperature usually. In one embodiment, acupuncture medical needle can be used to make positive mother matrix.Described pin can have any difformity, as circular cross-section, and square sectional, or the like.
Dull and stereotyped 12 and dull and stereotyped 13 hole can form by boring, etching or punching press.Described hole can have any difformity, and they can be by making as the photoetching of use in the MEMS processing and etching afterwards. The hole can be with arrangement arbitrarily, as square, cellular, or arbitrary graphic pattern.
Be an embodiment who makes the mother matrix mould below.
With microneedle 14 and dull and stereotyped 12 and dull and stereotyped 13 integrated processes, comprise that (1) will have the outstanding length that two flat boards 12 of arrangement in identical hole and dull and stereotyped 13 parallel alignments and (2) pass pin hole to the second dull and stereotyped 12 requirement in addition of first dull and stereotyped and second flat board.
Limit beyond dull and stereotyped 12 outstanding length by limits dull and stereotyped 12 and tip alignment plate 16 between dividing plate 15 decisions, tip alignment plate 16 is placed on and limits outstanding length of dull and stereotyped distance and the position parallel with limiting flat board.Described outstanding length difference, a part depends on the length that requires of microneedle, can be any scope in 0.1~5000 mu m range, as 0.5~250 μ m, or 250~1500 μ m, or the random length in these scopes. Can adjust the length that limits the microneedle on dull and stereotyped 12 by the thickness that changes dividing plate 15, this length also depends on the length that requires of the microneedle of being produced, it can be any scope in 1~5000 mu m range, as 1~250 μ m, 250~1500 μ m, or the random length in these scopes.Different with the positive unit mother matrix microneedle of making of MEMS or the meticulous Machining Technology of other CNC, the length of microneedle can be adjusted simply by the thickness of adjusting dividing plate, and the microneedle of different length can design by adjusting single pin in same SSP.The design of the microneedle of this combination different length can advantageously reduce the friction when thrusting skin.Support plate 13 can be to support any structure of pin, as sponge material.Pin can be fixed in dull and stereotyped 13 and/or dull and stereotyped 12 with glue or other fixative or adhesive.
The number that depends on the pin of dull and stereotyped size and existence, the distance between the pin can be different.Typically, pin can be to be placed apart from 5 μ m~5000 μ m, as interval 100~3000 μ m, any distance in 250~1000 μ m, or these scopes at interval each other.Described flat board can comprise the microneedle of arbitrary number, as 1~1, and 000,000, typically 10~100,000, as 50~10,000,100~1000, or the arbitrary number in these scopes.
In an optional embodiment, the hole in the qualification dull and stereotyped 12 can be sharpened the slope identical (Figure 1B) with needle point.Independent control can be the form of addressable array of actuators 18, and wherein each actuator moves each independent pin (Fig. 1 C).The principle of actuator and material can be that piezoelectricity, electroactive polymer, thermal expansion and electrochemistry activate. The real image of foraminous positive mother matrix of tool and needle point 114 is shown in Fig. 1 D in limiting flat board 112.
The negative norm tool is to be formed by positive mother matrix die casting. With curable gel or pourable macromolecular material,, make the negative norm tool that has with positive mother matrix mould similar face profile on the positive mother matrix mould thereby be poured on as uncured silicon or polydimethylsiloxane (PDMS).The method that another one is prepared the negative norm tool is under the situation that does not have assembly 12,15 and 16 positive pin array directly to be immersed in the curable gel or in the thermoplastic. In this case, the cavity of the micropin shape of negative norm tool determines that by the degree of depth that microneedle immerses in the curable gel it is controlled with dividing plate or thin straight-line actuator.
The method that another one is made positive minisize mould is a Precision Machining, as the contoured grinding machine by computer numerical control (CNC) (CNC).For example, on a plate of material, cut, comprise mould to provide, thereby make positive mould with a plurality of bottom surfaces from the outstanding microneedle of substrate along at least two different directions.Referring to for example U.S. Patent No. 7,497,980, its full content is hereby incorporated by.With reference to figure 1E, metal or plastic base plate 221 can repeat polishing as 222 or 223 with predetermined angle 224 along predetermined direction and determine aspect ratio, and can remove plate of material to form the array of polyhedral microneedle 225.Fig. 1 F represents to dissolve the pyramidal microneedle array by the cast of silicon auxiliary mould, and this silicon auxiliary mould is made by the positive mother matrix mould of the contoured grinding machine processing and fabricating of CNC.
The method that another one is made positive mother matrix microneedle mould is by negative norm tool cast microneedle array.Positive mother matrix microneedle pouring piece can be any material, as long as this material is pourable and has suitable cast structural intergrity afterwards. The microneedle array pouring piece can be water-fast, as ethyl cellulose, or water-soluble, as sodium carboxymethyl cellulose (SCMC).The negative norm tool can or burnish and fabricate by CNC precision drilling, milling. For example, microcavity array 232 is boring on polyfluortetraethylene plate 231 in Fig. 1 G, is used to make the positive mother matrix microneedle array of vinyl cellulose. With reference to figure 1H, utilize and the similar precision machining method of the contoured grinding machine of CNC, the edge of plate 242 is cut according to reservation shape and cutting distance, with the justified margin of cutting plate 241 with stack with formation negative norm tool. The method that another one is made the negative norm tool is by the arbitrarily curable materials of cast as PDMS, makes from above-described positive mother matrix mould.
The flow chart of exemplary process that utilizes fabrication techniques SSP described herein is shown in Fig. 2 A and 2B.Duplicating material comprises Merlon, polymethyl methacrylate (PMMA), polrvinyl chloride, polyethylene (PE) and polydimethylsiloxane (PDMS), and any heat or chemistry or photoactivation cross-linked material or thermoplastic.PDMS is preferred mold materials. The precursor of PDMS is the mixture of dimethyl silane and firming agent.A kind of preferable material is a medical grade silicon.Although also be not approved for medical grade silicon so far, (Dow Corning, Midland MI) can be 65 ℃ of full solidification for commercial SYLGARD 184.
The mould of plastics that comprises PDMS that is formed by this positive mother matrix helps making soluble SSP, and this is that the charge is small owing to it, can carry out large-scale production and the simple medium (easy medium) of eliminating the microbubble that may form in hydrogel is provided.Centrifuging process can be used for hydrogel solution is inserted the PDMS mould.Hydrogel can be easy to insert the top of mould when not having external pressure, particularly when silicon molding jig in a vacuum the time.Limit without any specific theory, this may be since the surface property of PDMS uniqueness with and with the compatibility of hydrogel. The explanation that another one is possible is, the inner vacuum that produces of PDMS under low pressure, the particularly inner vacuum in the zone of the cavity wall of microneedle are the pulling force of solution or gel being inserted the microneedle cavities tip.For large-scale production, can utilize the centrifugal action or the vacuum that are applied to negative norm tool bottom, or gel is pressed into the compression stress of microneedle cavities.Explain that as above-mentioned if microbubble is arranged, the ventilation unit of bottom that then is arranged on the miniature pin hole of mould is favourable when large-scale production.Alternatively, when using the negative norm tool to make SSP, can form micropore or porous plate so that mould ventilates, thereby prevent the formation of microbubble in microneedle cavities inside. In case after the hydrogel drying, SSP cuts into the patch element then from mould separating.
Make SSP
To comprise host material and selected medicine or the particulate liquid solution of medicine carrying pours in the negative norm tool of all PDMS as described above, and carry out drying.According to the viscosity of liquid solution and other physics and chemical property, can use additional force such as centrifugal force, vacuum power or compression stress that liquid solution is optionally being filled mould under the high temperature. In order to form solid solution, can use air drying, vacuum drying, lyophilization, convection furnace drying or other any suitable drying means to come dry solvent. For continuous large-scale production, can effectively utilize the flexiplast that comprises PDMS silicon. With reference to figure 2C, for continuous production, the tip, chamber of negative norm tool be open 206 and line up.Because open at this tip, can utilize liquid solution to fill this chamber at an easy rate from the vacuum of bottom or from the external pressure at top.Shown in Fig. 2 D, gel is poured out 207, pour into, suppress 208 or application of vacuum 209 alternatively, and then dry 210.In case behind the bone dry, mould of plastics that the charge is small or silicon molding jig just can be used as packaging material. When using, microneedle and mould can separate, and also can combine.
Because solvent and water constituent can be evaporated in dry run, the size of the mould that seems does not determine the final size of SSP.Therefore, the final size of SSP is littler than the size of mould.Alternatively, in the microneedle a plurality of different layers can by repeat to pour into a mould/solid solution of the identical or different concentration of wiping makes. When after microneedle forms, pouring into a mould adhesive phase, can easily produce viscosity microneedle patch. For example, with reference to figure 2E, the material of cast such as SCMC, and drying 205, the soft baking layer 207 of then pouring into a mould adhesive phase 206 and making by silicon or other soft hydrogel.Utilize repeatedly pouring technology, make viscosity and flexible microneedle patch.Dried SSP cuts into the suitable shape and size that are used for the patch element from mould separating. The description of the shape and size of representational this perforator is seen as U.S. Patent No. 6,945,952, No.7, and 182,747 and No.7,211,062, its full content is hereby incorporated by.
The suitable host material that is used for the SSP perforator comprises soluble polymer, includes but not limited to following material: sodium carboxymethyl cellulose (SCMC), hyaluronate sodium (HA), polyvinyl pyrrolidone (PVP), Polyethylene Glycol (PEG), polyvinyl alcohol (PVA), poly(ethylene oxide) (PEO), polyacrylic acid, polystyrene Sulfonate (polystyrene sulfonate), polypeptide, cellulose, hydroxypropyl cellulose (HPC), hydroxyethyl-cellulose (HEC), hydroxypropyl emthylcellulose (HPMC), dextrin, dextran, monosaccharide and polysaccharide, polyhydric alcohol, gelatin, arabic gum, alginate, the chitosan cyclodextrin, the combination of carbohydrate (carbohydrate) and other water-soluble natural and synthetic polymer and above-mentioned substance.
Carbohydrate derivates, as carbohydrate derivative (for example, trehalose, glucose, maltose, lactose, sucrose, maltulose, isomaltulose, lactulose, fructose, turanose, close in sugar, mannose, melezitose, dextran, maltodextrin, icodextrin, cyclodextrin, maltose alcohol (maltotol), Sorbitol, xylitol, inositol, sugar alcohol (palatinit), mannitol, stachyose and melitriose), can use or mix with above-mentioned substance.According to the physics and the chemical property of each element, the combination by above-mentioned substance designs mechanical performance and rate of dissolution.Carbohydrate can be melted the formation microneedle from mould, or with above-mentioned water-soluble polymer dissolves. In case be dried and and mold separation, can use extra dry run (after drying processing) to remove moisture.By this way, the mechanical strength of microneedle can obtain to improve or adjust, and can control the compressive strength of microneedle simultaneously.
Water-soluble component, as phosphate, nitrate and carboxylate glass (carboxylate glasses), magnesium chloride, potassium chloride and calcium chloride also can mix as host material separately or with matrix polymer. This composition can be used for stable or improve medicine and send or the inoculation ability. For inoculation, the granule of insolubility as depot forming adjuvant (depot adjuvants), may be mixed in in the substrate.Described substrate can also contain vitamin C or vitamin C derivatives.Vitamin C can alleviate contingent dermoreaction.Have been found that adding viscosity that vitamin C can reduce substrate makes substrate and fill so that obtain better mould.
Alternatively, the surface property of mould can improve by different technologies (as silanization, sided corona treatment, Cement Composite Treated by Plasma, face coat, polymer surfaces grafting etc.), so that improve the compatibility of gel and mould, and make after the colloidal sol drying easily and mold separation. The inventor finds that PDMS molding and SCMC hydrogel have good compatibility and can not form microbubble.
The making of SSP patch box and application member
The patch that the method for utilizing the present invention to describe is made preferably uses injection device (application member) to be applied on the skin.Fig. 3 A represents to have the patch application of spring driven application member. There is the box 301 of solid solution patch to be installed on the application member, thereby forms the application member 302 that is mounted with the spring compression that includes spring actuators 303 with compression spring 300.In this embodiment, user can use the microneedle patch alone and not want help. The flat closed form of box (occlusive flat form) has the advantage that volume reduces in storing and transporting. In the box of flat form, the piston of application member impacts the patch that places on the skin, and this may make impact force or Impact energy maximum, thereby helps SSP to be penetrated into massively in the target skin.Described box can not contact with environment from two aspects of physics and chemistry protection SSP, when using.Thin film that splits easily or tear or film can be protected the SSP in the box.In using the embodiment of flat box, microneedle and contact skin or place the place very approaching with skin, the piston portion of application member clashes into microneedle array in the skin then. The principle that this microneedle is inserted is equal to or is better than when microneedle and places on the skin and the situation in space greatly arranged between microneedle and the target skin.
The other example of Fig. 3 B and 3C difference the Show Button type application member 310 (Fig. 3 B) and mouse-type application member 313 (Fig. 3 C).Miniature box for needle or pin 312 can be connected on the application member 310, when pushing application member 310, start trigger 311.In mouse-type application member 313, trigger 314 is on the top of mouse. The vertical view of described box and side view are described in Fig. 3 D and Fig. 3 E respectively. But microneedle 318 is fixed on the fracturing diaphragm 319 of disposable plastic box 320 inside, and microneedle 318 is by 322 protections of the closure film on the surface 321.Fig. 3 F illustrates the insertion pattern that working pressure detects film 323.
By the SSP administration
Fig. 4 illustrates another patch examples of applications with the allotment gel that comprises cream and/or emulsion.According to application, described allotment gel can comprise the identical or different active component of active component among one or more and the SSP.This allotment gel can comprise the medicament to skin beneficiating, as humidification adjuvant or irritation or antimicrobial agent. In this example 40, before using patch, will allocate gel 42 and put on the target skin 43.Having described patch in 44 is applied on the pretreated skin.In 45 and 46, patch puts on the skin, and after the SSP dissolving, the allotment gel puts on position 43.In this case, can send active component in the gel by the hole that patch inserts and dissolving produces.
SSP
It maybe can be taper shape, pyramid, wedge shape or sheet shape that the SSP perforator can have axle straight or taper, is pre-determined by positive mother matrix. In preferred embodiment, the external diameter of SSP perforator is maximum in its bottom or second end, about 1~2000 μ m, and near the external diameter of the perforator first end is preferably 1~100 μ m.The length of SSP perforator is typically in 10~5000 mu m ranges, more preferably in 100~2000 mu m ranges.Skin is not slick surface, but has the different degree of depth on the uneven microcosmic. In addition, the elasticity of cuticular thickness and skin changes along with the different of people and people, changes along with the difference of the position on a certain specified person. For penetrating of effective administration and relative painless depletion of blood, ideal penetration depth has a scope, rather than single value. The penetration depth of SSP perforator can influence pain can influence delivery efficiency simultaneously. In certain embodiments, the penetration depth of perforator is in the scope of 10~1000 μ m.When percutaneous was used, " penetration depth " of SSP perforator was preferably less than 500 μ m, so that can not penetrate epidermal area by the perforator of horny layer insertion skin. This is a kind of best approach that can be avoided contacting neural and blood vessel.Because the roughening on the elasticity of skin and surface, the basal layer relevant with the SSP system can not insert skin fully, and therefore, in this was used, the physical length of SSP perforator can be longer.
According to medical needs, may need to be penetrated into the perforator of skin corium in some applications. In these cases, utilizing the SSP system when handling the situation of instant administration is the selection of reality.By regulating variable (the SSP length of perforator, size, speed and impact force that the mechanical performance of substrate or substrate layer and SSP perforator insert) and consider the elasticity of target skin, the hardness and the rough surface of skin, can optimize the penetrating component of SSP perforator. The major function of SSP perforator is to thrust horny layer carrying out instant administration from substrate, and alternatively, applies the passage of smearing gel or cream or emulsion after helping to be used in or stays open from the passage of memorizer. As long as the SSP perforator has rational dissolution velocity, and enough firmly can thrust horny layer, any bio-compatible material can be used as the SSP perforator. In some applications, insoluble microneedle is very useful. At this moment, can use water-fast hydrogel in the manufacture method of Miao Shuing in the above, as ethyl cellulose.
In some cases, need the pointed portion of SSP to have spissated medicine. This SSP can design by the method and/or the spissated method of granule of the repeatedly cast/wiping of description before.Fig. 5 A and 5B show the real image that utilizes the SSP that silicon system negative norm tool makes that is made of sodium carboxymethylcellulose pyce.In another embodiment, utilize said method can be easy to make flexible and adhesive substrate with microneedle array. For example, SCMC fills microneedle mould, sequentially pours into a mould adhesive phase and soft hydrogel formulation.Resulting patch has hard microneedle and viscosity/soft bottom microneedle array, and this patch does not need other bonding lining form or covering.
SSP patch system
SSP patch system comprises bin and one or more perforator alternatively, the gel that this bin includes liquid or constitutes with ancillary drug, and perforator extends from least a portion of this reservoir surface.Thereby described SSP perforator and patch the system horny layer of transdermal together strengthen percutaneous dosing and instant administration are provided. When medicine when basal layer scatters, utilize lining form can realize continuing medication from basal layer. In the patch system, SSP perforator and bin can be configured to individual unit or isolating unit.
According to application, system puts on skin with the SSP patch, enters epidermis or skin corium so that one or more SSP perforator penetrates horny layer. In another method, can use SSP and gel, cream and/or emulsion. For example, described gel can comprise medicine and/or desired accessory drugs, and described gel can be applied in or be sprayed at the position of requirement.Insert the SSP patch afterwards.Perhaps, can after using, patch apply gel.
The medicament for the treatment of and/or preventing property can be transmitted in the SSP system, comprises medicine and vaccine and other bioactive molecule, passes through or enter skin and other tissue. The SSP device allows carrying out administration and arriving body fluid through skin or other tissue disorder organizing under injury, pain and/or the stimulation with minimum. When administration, the SSP perforator mainly is made of active medicine (or drug particles itself), and gel combination (comprising cream and emulsion) can design by drug distribution curve as required.According to application, osmotically active or counteirritantia chemical compound or antimicrobial medicament can have useful effect. In diagnostic application, the SSP perforator can comprise or be made up of the sensitive material with specific analyte that exists or metabolite reaction that loads. In order to change or to control injection speed, can have under the tectal situation, the utilization external physical strengthens system, as utilizes ionotherapy, electrophoresis, sound wave to penetrate the combination of method (sonophoresis), piezoelectric response, heating element heater, magnetic cell or similar response or above response.
The medicine of sending by the SSP system
The medicine of sending can be protein, peptide, nucleotide, DNA (deoxyribonucleic acid) (DNA), ribonucleic acid (RNA), siRNA (siRNA, small interfering RNA), gene, polysaccharide and synthetic organic and inorganic compound.Representational medicament includes but not limited to: the medicine of anti-infective, hormone, growth regulator, the effect of adjusting heart or blood flow and the medicine of pain management.Described medicine can be used for vaccination or topical therapeutic or carry out zone or whole body therapeutic.
Many medicines all can various treatment rate be sent, control this treatment rate by changing some design factors, this design factor comprises: the frequency of utilization of the size of the size of the rate of dissolution of the size of SSP, the drug loading among the SSP, substrate, the quantity of SSP perforator, SSP patch, gel (comprising cream and emulsion) and composition and device etc.Although can adopt the penetration length by prolonging the SSP patch that medicine is directly sent in the blood flow, yet the application of most SSP medicine dermal delivery is an epidermal area.
SSP patch disclosed herein system is also very useful by transporting of the tissue except that skin for control.Other non-skin histology that is used for administration comprises nasal cavity or vagina, oral cavity, eye, tooth regions or enters organization internal or insert other accessibility mucous layer to promote medicine and be transported to these organization internals or through these tissues under laparoscopically assistance. For example, the eye that the SSP patch can be inserted the patient is delivered to medicine in the eyes with the actuator that slowly moves helping, thus control or correct conjunctiva, sclera and/or corneal problems. The medicine of allotment rests in the tissue, even still can keep administration after taking down patch. The SSP patch also can insert in the oral cavity that comprises buccal mucosa, carries out quick whole body administration or short delivery time, for example, breaks through pain management (breakthrough pain management) and carries out dental care. Can send the medicine of the muscle relaxant in using as orthodontics through oral mucosa, so that in oral cavity or gingiva, carry out topical therapeutic. The another one example can be used the SSP system in vivo, and for example, the covering gastrointestinal tract promotes the picked-up to oral drugs, or the covering blood vessel promotes medicine to infiltrate blood vessel wall.Under the situation that interior tissue is used, use biological adhesive SSP material can help SSP to stop the longer time in vivo. For example in case of emergency, can use the food patch that comprises essential amino acid, fat and vitamin.
Intradermal administration is used
The another one important use is vaccination and is used for the treatment of and prevents allergy.Owing to contain the network of antigen presenting cell such as islet cells and corium dendritic cell in the skin, so skin is the desired site that effective vaccine is sent. The SSP system that is used for skin immunization can reduce the dosage of vaccine, makes medicine be delivered to the skin dendritic cell fast, thereby and can produce storage effect vaccination better. Can be easy to design the SSP system that is used for polyvalent vaccine, it is expected at when transporting and storing has better stability than liquid vaccine.
Another one important use of the present invention is to be used for cosmeceutical (cosmeceutical).Have particulate SSP system can be effectively and be used for safely removing or reduce that wrinkle forms, skin aging hyperhidrosis and alopecia. For example, can utilize system as herein described delivery of botulinum toxins (Botox), alkyd, vitamin and vitamin derivative, epidermal growth factor (EGF), adenosine, arbutin etc.Described system can also be used for the treatment of pathological changes or unusual skin characteristic, as is common in pimple, acne, clavus, wart, callus, thumb cyst, actinic keratosis and hyperkeratosis scleroderma on face, arm, lower limb or the foot. The SSP system also can be used for tattoo (tattoo) formation/removal patch in the cosmetic applications. The SSP system of active or false (sham) also can be used for acupuncture.
